This study sought to improve the reproductive performance of anoestrous high-producing dairy cows by including equine chorionic gonadotrophin (eCG) after progesterone-releasing intravaginal device (PRID) removal. In Experiment I, 806 cows at 51-57 days post-partum were randomly assigned to a PRID (treated with PRID), PRID-500 (treated with PRID plus 500 IU of eCG) or PRID-750 (treated with PRID plus 750 IU of eCG) group. In Experiment II, 422 cows showing a long anoestrus period (animals with no oestrus signs nor luteal tissue 35 days before treatment) were randomly assigned to the PRID, PRID-500 or PRID-750 groups. The dependent variables considered in binary logistic regression analyses for both experiments were the rates of oestrus, ovulation and conception after treatment, the cumulative conception rate on Day 120 post-partum and pregnancy loss. In Experiment I, interaction between treatment and season showed a significant effect on the oestrous response. Thus, during the warm season, PRID group cows were 8.9 times more likely to express oestrus than the remaining cows. Moreover, inseminated cows with two or more corpora lutea 8-14 days after treatment were more likely to become pregnant (by a factor of 2.4) than cows with a single corpus luteum. Finally, cows without luteal structures treated with PRID were 0.4 less likely to be pregnant on Day 120 post-partum, compared with the remaining cows. In Experiment II, cows in the PRID group treated during the warm or cool season were less likely to exhibit oestrus (by a factor of 0.06 or 0.2, respectively) or ovulate (by a factor of 0.004 or 0.14, respectively) than the remaining cows. In conclusion, in anoestrous cows in both experiments, the addition of eCG to the use of an intravaginal progesterone device to induce oestrus was beneficial. The recommended dose of eCG is 500 IU.

The aim of the present study was to characterize the early postpartum period in clinically healthy dairy cows by ultrasonography (US), endometrial cytology (EC), and white blood cell counts, and determine possible relationships between postpartum findings and subsequent reproductive performance. Fifty-three dairy cows were examined on Days 15 to 21 (Visit 1), 22 to 28 (Visit 2), and 29 to 35 (Visit 3) postpartum. The clinical examination included: examination of vaginal fluid, EC, transrectal palpation and ultrasonography of the genital tract (cervical diameter, endometrial thickness, presence of a corpus luteum [CL] or intrauterine fluid [IUF] and its echogenicity). Luteal activity (presence of a CL in a single visit), return to cyclicity (presence of a CL in 2 consecutive visits), and conception rate at 70 and 120 days postpartum were considered as the dependent variables in four consecutive binary logistic regression analyses. Factors affecting leukocyte counts were established by general linear model (GLM) repeated measures analysis of variance. Based on the odds ratio (OR), the likelihood of luteal activity was higher in multiparous than primiparous cows (OR = 3.75) and tended to diminish in cows showing increased endometrial thickness in Visit 1 (V1) (OR = 0.06). The likelihood of returning to cyclicity decreased for each centimeter increase in cervical diameter in V1 (OR = 0.14) and that of conception on Day 70 was lower in cows showing the presence of echogenic or anechogenic IUF in V1 (OR = 0.09 or OR = 0.13, respectively) compared with cows lacking IUF. Effects of parity and IUF were observed on neutrophil counts. Positive EC results were unrelated to the cumulative conception rate at 70 and 120 days in milk, whereas cows returning a positive EC result in V1 showed a greater likelihood of increased endometrial thickness. In conclusion, measuring cervical diameter, endometrial thickness, and detecting the echogenicity of IUF by ultrasonography from Days 15 to 21 postpartum in clinically normal cows is an appropriate tool to predict subsequent reproductive performance. Vaginal examination and transrectal palpation alone did not emerge as valuable predictors.

This study was designed to assess the safety of phase I vaccination against Coxiella burnetii in advanced pregnancy and the effect of vaccination on subsequent reproductive performance of high producing dairy cows. C. burnetii serostatus was determined in 719 dairy cows by individual serological testing. According to their serostatus, cows were randomly assigned to a control (n=359) or vaccine (n=360) group (inactivated phase I on Days 171-177 and 192-198 of gestation, Coxevac-Ceva Sante Animale). Using a χ(2)-test, vaccination had no effect on abortion before parturition, retention of placenta and stillbirth, either in seropositive as in seronegative cows. Coxs proportional hazards model revealed that cows in the vaccine group were 1.22 times more likely to conceive during the first 150 days in milk than cows in the control group. Moreover, the likelihood of pregnancy was lower in multiparous cows, cows with a retained placenta and cows undergoing first AI during the warm season compared to the remaining animals (by factors of 0.75, 0.69 and 0.69, respectively). In animals testing seronegative for C. burnetii, the likelihood of pregnancy was 1.25 times higher in vaccinated cows compared to non-vaccinated seronegative animals. No effect of vaccination on subsequent fertility was detected in seropositive animals. In conclusion, the results of this study indicate that phase I vaccination against C. burnetii during advanced pregnancy in dairy cows is safe and improves subsequent fertility of C. burnetii seronegative animals.

The objective was to examine risk factors for the interval to resumption of ovarian cyclicity (ROC), multiple ovulations (MCL), ovarian follicular cysts (OC), and presence of intrauterine fluid (IUF) at 22 to 28 [visit (V) 1] and 36 to 42 (V2) days in milk (DIM) in dairy cows. The study was conducted retrospectively by evaluating records from 1,155 Holstein cows from 3 herds. Ovaries and uteri were examined at V1 and V2 by transrectal ultrasonography to determine ovarian structures and IUF. Based on the odds ratio, multiparous cows were more likely to have ROC at V1 by a factor of 1.79 compared with primiparous cows. The likelihood of ROC at V1 was lower in cows with higher milk production, in cows with retained fetal membranes (RFM) or cows with IUF at V1 by factors of 0.98 (for each kg of milk increased), 0.52, and 0.61, respectively. Based on the odds ratio, cows diagnosed with IUF at V2 were 2.85 times more likely to have attained ROC at V2. Multiparous cows and cows that delivered twins were 2.73 and 2.16 times, respectively, more likely to have MCL at V1, whereas cows with RFM were 0.38 times less likely to have MCL at V1. The likelihood of MCL at V2 was higher in cows with MCL and OC at V1 by factors of 2.67 and 1.91, respectively. Multiparous cows were 8.51 times more likely to have OC at V1 than primiparous cows. Higher producing cows were more likely to have OC at V2 by a factor of 1.04 compared with lower producing cows. Parity, stillbirth, RFM, and ROC at V1 were all identified as risk factors for IUF at V1. Cows with RFM and delivering twins were more likely to be diagnosed with IUF at V2 by a factor of 3.43 and 4.07, respectively. In summary, parity, twinning, RFM, metritis, IUF, and milk production were all associated with altered ovarian structures, and the presence of IUF was related to parity, twinning, RFM, and ROC in postpartum dairy cows.

This study compared the responses shown by lactating dairy cows to four different P4-based protocols for AI at estrus. Cows with no estrous signs 96 h after progesterone intravaginal device (PRID) removal were subjected to fixed-time AI (FTAI), and their data were also included in the study. In Experiment I, follicular/luteal and endometrial dynamics were assessed every 12 h from the beginning of treatment until AI. The estrous response was examined in Experiment II, and fertility was assessed in both experiments. The protocols consisted of a PRID fitted for five days, along with the administration of different combinations of gonadotropin releasing hormone (GnRH), equine chorionic gonadotropin and a single or double dose (24 h apart) of prostaglandin F2α. In Experiment I (40 cows), animals receiving GnRH at the start of treatment showed a significantly higher ovulation rate during the PRID insertion period while estrus was delayed. In Experiment II (351 cows), according to the odds ratios, cows showing luteal activity at the time of treatment were less likely to show estrus than cows with no signs of luteal activity. Treatment affected the estrous response and the interval from PRID removal to estrus but did not affect conception rates 28–34 days post AI. Primiparous cows displayed a better estrous response than multiparous cows. Our findings reveal acceptable results of 5-day P4-based protocols for AI at estrus in high-producing dairy cows. Time from treatment to estrus emerged as a good guide for FTAI after a 5-day P4-based synchronization protocol.

The impact of long-term vaccination against Coxiella burnetii on the fertility of cows was studied. Double vaccinations three weeks apart at the start of the third trimester of gestation in each of two consecutive pregnancies were applied. The final study population consisted of 410 cows after the first vaccination round. Based on the odds ratios, the likelihood of early fetal loss (pregnancy loss following a positive pregnancy diagnosis before Day 90 of gestation) was higher in control cows (OR = 1.42) than in vaccinated cows. The final study population consisted of 336 cows after the second round of vaccination. According to the odds ratios, vaccinated C. burnetii seronegative cows were less likely to be subfertile (> 3 AI) (OR = 0.4) compared to non-vaccinated seronegative animals, and the likelihood of early fetal loss was lower in vaccinated C. burnetii seropositive animals (OR = 0.3) compared to non-vaccinated seronegative cows. Seropositivity to C. burnetii was positively related to twin pregnancy after the two rounds of vaccination (OR = 2.1 and 3.5, respectively). These results indicate that two consecutive vaccination rounds against C. burnetii in advanced gestation reduce subfertility and early fetal loss in dairy cows.

Abstract This study evaluated the effect of a once-used progesterone (P4) intravaginal device (PRID) associated with four different shortened P4-based estrus synchronization (ES) protocols on estrous response (ER) and pregnancy per AI (P/AI) in cyclic and acyclic lactating dairy cows. Cows (n=465) were randomly assigned to one of the following protocols: 1) 2PGG, cows were given a PRID-Delta and 100 μg GnRH i.m. at PRID insertion (day 0). The PRID was left for 5 d, and 25 mg of dinoprost (PGF2α) i.m. given twice at PRID removal and 24 h later; 2) 2PGGe, same treatments as 2PGG plus 500 IU of eCG i.m. at PRID removal; 3) 2PGe, same treatments as 2PGGe, except GnRH was not given at PRID insertion; 4) PGe, same treatments as 2PGe, except PGF was only given at PRID removal. A total of 258 cows received a new PRID-Delta containing 1.55 g of P4, whereas 207 cows received a once-used PRID. Estrus was determined from P4 device removal until 96 h after using an automated heat detection system. Cows in estrus were given a second GnRH at AI and those without signs of estrus by 96 h after PRID removal were given GnRH and timed-AI (TA I). All inseminations were performed by one technician with commercially available frozen-thawed semen. Ultrasonography was performed at initiation of protocol and 28-34 days post AI to determine cyclicity and pregnancy status, respectively. Cows receiving once-used P4 devices had greater ER than cows receiving a new device (59.9 vs. 50.0; P=0.029), but P/AI did not differ between P4 devices, respectively (P>0.1; 40.6 vs 40.7%). Cyclic cows were less likely to display estrus than acyclic cows by a factor of 0.66 (P=0.036). Cows subjected to the 2PGe (2.41; P<0.01) protocol were more likely to display estrus than cows subjected to the 2PGG, whereas cows subjected to the PGe protocol did not differ (0.94; P=0.8) from those in the 2PGG group. Despite differences in ER, neither cyclicity nor estrus synchronization protocol affected P/AI (overall 40.6%). In summary, cyclic cows, those given a new P4 device and those subjected to either 2PGG or PGe protocol had reduced ER. However, all the factors examined had no significant effect on P/AI. All the estrus synchronization protocols resulted in acceptable fertility.
