Electrophoretic analysis of some Lathyrus L. species based on seed storage proteins

İrfan Emre

Present study reported the variation in electrophoretic seed subfraction (albumins, globulin A, globulin B, glutelins) patterns in some Lathyrus species collected from their natural habitats different localities in Turkey. Electrophoretic data were documented by using a gel documentation system (Bio-Rad, USA) and analysed by using Quantity 1-D analysis software and also the dendograms were formed with 4.0% tolerance in UPGAMA (Unweighed Pair-Group Arithmetic Mean). The differences among species were observed and all seven species were clearly identifiable from the protein patterns. The dendograms obtained from cluster analysis of seed storage proteins data showed examined species have displayed a number of distinct relationships. According to our results, it seems justified to recommend a wider use of electrophoretic analysis of seed storage proteins in taxonomic investigations of leguminous plants.

Abstract In the present study, fatty acid composition in mature seeds of the Lathyrus L. taxa were determined by using gas chromatography. Present results indicate that the seed oils of studied Lathyrus taxa have palmitic acid (C16:0) and stearic acid (C18:0) as major saturated fatty acids. Moreover, the present data show that oleic acid (C18:1), linoleic acid (C18:2 n 6) and a—linolenic acid (C18:3 n 3) are major unsaturated fatty acids in studied Lathyrus taxa. The highest oleic acid (C18:1 n 9) content is found in L. boissieri (34,1%) and L. inconspicuus (24,5%) while L. clymenum and L. ochrus, which are members of section Clymenum, have highest polyunsaturated fatty acids, 66,7%, 73,6%, respectively.

Abstract In the present study, levels of vitamin and fatty acid composition in mature seeds of some Lathyrus L. taxa [L. roseus Stev., L. tuberosus L., L. belinensis Maxted & Goyder, L. annuus L., L. gorgoni Parl. var. gorgoni, L. cicera L., L. sativus L., L. stenophyllus Boiss. & Heldr., L. hirsutus L., L. chloranthus Boiss. et L. aphaca L. var. affinis (Guss.) Arcang.] were determined by using HPLC (high-performance liquid chromatography) and VPC (vapour gas chromatography). Palmitic acid (C16:0), stearic acid (C18:0), oleic acid (C18:1 n-9), linoleic acid (C18:2 n-6) and α-linolenic acid (C18:3 n-3) were found as major fatty acid components in all studied eleven taxa. In addition, δ-tocopherol, α-tocopherol, α-tocopherol acetate and vitamin D3 were found highest level and retinol, retinol acetate, vitamin D2 and vitamin C content (apart from L. belinensis and L. hirsutus) were found lowest ratio in the examined Lathyrus taxa.

This study used karyological techniques to determine the chromosome numbers and morphology of eight species of Onobrychis L. (O. caput-galli (L.) Lam, O. aequidentata (Sibth. & Sm.) d’ Urv, O. fallax Freyn & Sint. var. fallax, O. lasiostachya Boiss, O. viciifolia Scop., O. oxyodonta Boiss. subsp. armena (Bois. & Huet) Aktoklu, O. hypargyrea Boiss. and O. cappadocica Boiss.). The results of this study determined the chromosome numbers of O. cappadocica as 2n = 16; O. viciifolia as 2n = 28 and the other species as 2n = 14 The karyotypes of species consisted of median-centromeric (m) or submedian-centromeric (sm) chromosomes. However, O. oxyodonta Boiss. subsp. armena (Bois. & Huet) Aktoklu was found to have only the median-centromeric (m) chromosomes. According to the results of the present study, of the eight Onobrychis taxa, only O. hypargyrea has a pair of satellite chromosomes (sat-chromosome). Furthermore, this study detected karyotype asymmetry.

Abstract In the present study, fatty acid composition in mature seeds of the Vicia L. taxa were determined by using gas chromatography. Present results indicate that the seed oils of studied Lathyrus taxa have palmitic acid (C16:0) and stearic acid (C18:0) as major saturated fatty acids. Moreover, the present data showe that oleic acid (C18:1), linoleic acid (C18:2 n 6) and α-linolenic acid (C18:3 n 3) were major unsaturated fatty acids in studied taxa. The highest oleic acid (C18:1 n 9) content was found in V. crocea (Desf.) B. Fedstch. (26.5%) but it was found at the lowest level in V. mollis (7.8%). In addition, present study determine that investigated Vicia species have linoleic acid (46.4–79.1%) and linolenic acid (3.8–21.3%) contents.
