The handicap principle of sexual selection theory states that colourful phenotypic traits signal aspects of individual quality because only individuals in prime condition can afford to produce and bear conspicuous traits. Melanin-based pigments participate in the elaboration of many secondary sexual characters and, given their role in sexual selection, melanin-based coloration may therefore honestly reflect individual quality. Although the expression of melanism is usually under genetic control, in some species it is condition dependent. However, the underlying physiological mechanism is yet unknown. Based on the negative feedback link between corticosterone and melanogenesis (melanocortins, tyrosinase) in response to stressful environmental factors, we hypothesize that corticosterone mediates the condition-dependent component of melanism. This hypothesis predicts that stressful factors induce a rise in circulating corticosterone which inhibits the secretion of melanocortins and tyrosinase and in turn melanin production. We tested this prediction by manipulating the level of corticosterone at the time of melanin production in nestling barn owls, Tyto alba, a species showing heritable variation in the degree of phaeomelanism from reddish-brown to white. The finding that corticosterone-implanted nestlings produced feathers with less phaeomelanic coloration than placebo-implanted nestlings is consistent with the hypothesis that the environment-mediated reduction in the degree of melanism is, at least in part, caused by a rise in corticosterone. In species in which the expression of melanin-based coloration is condition dependent, we now need a test showing that individuals with less corticosterone and more melanin-based signals are individuals in better condition.

Biomineralization research on mollusc shells has mostly focused on nacre formation. Chitin, silk fibroin protein, and acidic macromolecules are important components for shell formation. Although the principle concept behind shell calcification was developed many years ago, the individual components have not been well scrutinized. Besides that, Mollusca are the second largest invertebrate phylum, but comprehensive biochemical research involving a comparison of different taxa is still rare. This study reconsiders the above three components with adding some biochemical data of aculiferans. The presence of chitin in polyplacophorans sclerites was confirmed by IR and pyrolysis GC/MS. DMMB staining data inferred that sulphated groups present in aplacophoran cuticle but not in polyplacophorans cuticle. These insight suggested importance of comparison between acuriferans and conchiferans.

The review article is devoted mainly to the description of applications of gold nanoparticles (GNPs) in separation sciences, especially in electromigration and chromatographic techniques. The applications of GNPs in particular separation methods, CE, microchip CE, MEKC, CEC, HPLC and GC, are classified according to the molecular size of the analytes from low-molecular-mass compounds via medium sized substances to biopolymers (proteins and nucleic acids). A very recent and promising utilization of GNPs for sample preparation, preconcentration and preseparation of selected analytes from complex matrices is presented as well. Moreover, in two introductory sections, typical preparation procedures of the GNPs and their modifications are presented and physico-chemical and analytical methods employed for characterization of the native and modified GNPs are briefly introduced.

Abstract Eggshell pigment constituents were determined by a high-performance liquid chromatography method. Most of the work was done on whole clutches of red-backed shrike ( Lanius collurio ). In addition to the known pigments (protoporphyrin IX, biliverdin), a new pigment, zinc-containing protoporphyrin IX, was found as well. Its content was highly variable—from 0% to 100%. The content of total protoporphyrin IX (with and without zinc) ranged from 1.72 to 114.84 nmol (average for whole clutches was 39.93 nmol), while the content of zinc-deficient protoporphyrin IX was ND (not detectable) -110.46 nmol (average for whole clutches was 34.02 nmol) and the content of zinc-containing protoporphyrin IX was ND - 32.28 nmol (average for whole clutches was 5.91 nmol). Zinc-containing protoporphyrin IX was absent in 33% of eggs, while zinc-deficient protoporphyrin IX was absent in a single case only (2%). If clutches from relatively “polluted” and “unpolluted” regions were compared, no differences have been found. Based on these findings is the conclusion that the high variability in eggshell pigment content is likely to reflect physiological influences (e.g. order of egg laying and the whole condition of the nesting female) rather than environmental interferences. This conclusion is in agreement with our previous findings. Additional data regarding the pigment content of seven other bird species are also included.

Microemulsions prepared by using either sodium dodecyl sulfate (SDS) or trimethyltetradecylammonium bromide (TTAB) were tested with regard to the migration time, selectivity and number of theoretical plates using a set of both water-soluble and lipid-soluble vitamins. While in the separations where SDS was used as the surface-active agent for microemulsion preparation, better resolutions were obtained with both hydrophilic and lipophilic vitamins, with TTAB the best separations were obtained with the micelle-forming reagent only or with diethyl ether as the microemulsion-forming component. In the SDS-based separations nicotinic acid was moving with the same velocity (relative to the endoosmotic flow) regardless of the emulsion or micellar phase used. It is proposed that owing to its behaviour it can serve as an ideal internal standard in further studies on microemulsion separations.

OBJECTIVE The induction of obesity, dyslipidemia, and insulin resistance by high-fat diet in rodents can be prevented by n-3 long-chain polyunsaturated fatty acids (LC-PUFAs). We tested a hypothesis whether AMP-activated protein kinase (AMPK) has a role in the beneficial effects of n-3 LC-PUFAs. RESEARCH DESIGN AND METHODS Mice with a whole-body deletion of the α2 catalytic subunit of AMPK (AMPKα2−/−) and their wild-type littermates were fed on either a low-fat chow, or a corn oil-based high-fat diet (cHF), or a cHF diet with 15% lipids replaced by n-3 LC-PUFA concentrate (cHF+F). RESULTS Feeding a cHF diet induced obesity, dyslipidemia, hepatic steatosis, and whole-body insulin resistance in mice of both genotypes. Although cHF+F feeding increased hepatic AMPKα2 activity, the body weight gain, dyslipidemia, and the accumulation of hepatic triglycerides were prevented by the cHF+F diet to a similar degree in both AMPKα2−/− and wild-type mice in ad libitum-fed state. However, preservation of hepatic insulin sensitivity by n-3 LC-PUFAs required functional AMPKα2 and correlated with the induction of adiponectin and reduction in liver diacylglycerol content. Under hyperinsulinemic-euglycemic conditions, AMPKα2 was essential for preserving low levels of both hepatic and plasma triglycerides, as well as plasma free fatty acids, in response to the n-3 LC-PUFA treatment. CONCLUSIONS Our results show that n-3 LC-PUFAs prevent hepatic insulin resistance in an AMPKα2-dependent manner and support the role of adiponectin and hepatic diacylglycerols in the regulation of insulin sensitivity. AMPKα2 is also essential for hypolipidemic and antisteatotic effects of n-3 LC-PUFA under insulin-stimulated conditions.

A mixture of ten steroids was separated by microemulsion and micellar (SDS and glycodeoxycholate) electrokinetic chromatography systems. Separations were done on a 50 cm (to the detector) x 50 microns I.D. fused-silica capillary. Complete separation of all the test compounds in the micellar mode was obtained with glycodeoxycholate (50 mM) in 25 mM borate buffer, pH 6.5, as the micelle-forming agent. The best results, however, were obtained using microemulsion electrokinetic chromatography in which higher aliphatic alcohols were used as the microemulsion-forming modifiers. The system consisted of n-hexanol (0.81%), SDS (3.31%) and n-butanol (6.61%) in 20 mM phosphate buffer, pH 10.0 (89.28%, w/w). In the microemulsion mode, linear calibration for steroid standards was obtained in the concentration range 3 x 10(-4)-3 x 10(-5) mol 1(-1) with a detection limit of 1 pmol. The method was validated and applied to an 11 beta-hydroxysteroid dehydrogenase assay in tissues.

The AMP‐activated protein kinase (AMPK) cascade is a sensor of cellular energy charge that promotes catabolic and inhibits anabolic pathways. However, the role of AMPK in adipocytes is poorly understood. We show that transgenic expression of mitochondrial uncoupling protein 1 in white fat, which induces obesity resistance in mice, is associated with depression of cellular energy charge, activation of AMPK, downregulation of adipogenic genes, and increase in lipid oxidation. Activation of AMPK may explain the complex metabolic changes in adipose tissue of these animals and our results support a role for adipocyte AMPK in the regulation of storage of body fat.

It has been proposed that blue colouration in eggs has evolved as a signal of female quality that males can use to modulate their parental investment. This hypothesis is based in the antioxidant properties of biliverdin whose costly deposition in the eggshell is expected to signal female antioxidant capacity and egg quality. Since maternally derived androgens are costly to produce and may adaptively affect offspring phenotype, high-quality females may benefit by signalling their androgen investment through egg colouration. Our aim was to investigate whether egg colour variation in the spotless starling reflected the amount of pigments on the eggshell and whether egg pigmentation was related to female and egg quality. Chromatography analyses revealed that spotless starling eggshells contained two different pigments: biliverdin and protoporphyrin IX with no correlation between them. Biliverdin contents correlated positively with egg colouration indicating that darker eggs with a higher peak in the blue–green segment of the spectrum contained higher amounts of biliverdin. Eggs containing more biliverdin were laid by high-quality females and contained higher yolk testosterone levels. However, despite the strong correlation between biliverdin and colorimetric variables, egg colouration did not reflect accurately female and egg quality. Our results provide evidence that eggshell pigmentation in the spotless starling is related to female and egg quality as shown by the yolk testosterone levels. However, the lack of relation between egg colour and female condition and egg quality do not provide evidence to support the signalling function of egg colouration.

The protein composition of the insoluble avian eggshell matrix was studied. The determination of these proteins insoluble in water (EDTA-insoluble) was carried out using enzymatic cleavage followed by a high-performance liquid chromatography–mass spectrometry method. The influence of various enzymes on the protein splitting also was studied. The distribution of proteins depends on the type of layer (localization within the eggshell): ovocalyxin-32 was found mainly in the outer layer (the cuticle); ovocleidin-116 and 17 and ovocalyxin-36 were found throughout the whole eggshell, whereas ovalbumin was only found in the inner layer, the mammillary. The pigment (protoporphyrin IX) was mainly found in the cuticle and is incorporated into the protein network.
