Analysis of pathogenicity by restriction-endonuclease digestion of amplified genomic DNA of Entamoeba histolytica isolated in Pernambuco, Brazil

The pathogenicity of 47 strains ofEntamoeba histolytica isolated in Pernambuco, Brazil, was examined using the polymerase chain reaction (PCR) followed by restriction-endonuclease digestion. Electrophoretic patterns of PCR products digested withHinfI revealed that all strains were nonpathogenic. The results were entirely in accord with phenotypic properties such as isoenzyme patterns and the failure to bind a pathogenic-isolatespecific monoclonal antibody. When the sensitivity of PCR was examined, amplified products could be detected from template DNA equivalent to five trophozoites. These observations indicate that PCR amplification of genomic DNA and subsequent restriction-enzyme digestion is a useful strategy for obtaining a sensitive and accurate diagnosis. The present study also demonstrates that nonpathogenic strains ofE. histolytica predominate in northeastern Brazil.

Colonização pelo Staphylococcus aureus em profissionais de enfermagem de um hospital escola de Pernambuco

Estudio realizado para identificar prevalencia de colonizacion por Staphylococcus aureus en profesionales de enfermeria de hospital universitario de Pernambuco, asi como evaluar el perfil de resistencia de la bacteria aislada. Se realizo un estudio transversal en el que se recolectaron muestras biologicas de manos y cavidad nasal. La identificacion del S. aureus se realizo mediante cultivo en agar-sangre, agar-manitol salado y mediante pruebas de catalasa y coagulasa. El perfil de sensibilidad se determino por tecnica de Kirby Bauer y para la determinacion de resistencia a meticilina se realizo screening en placa con oxalacina, con adicion de 4% de NaCl. De 150 profesionales evaluados, 39 estaban colonizados, lo que demostro prevalencia de 25,8%. Entre las variables estudiadas, faja etaria y cantidad de EPI se presentaron asociadas con la colonizacion por la bacteria. De todas las cepas aisladas, apenas cinco presentaron resistencia a meticilina.This study was performed with the objective to identify the prevalence of colonization by Staphylococcus aureus in nursing professionals from a teaching hospital in Pernambuco, and evaluate the resistance profile of these isolates. S. aureus was identified using agar (blood agar and mannitol salt) via catalase and coagulase tests. The sensitivity profile was determined by Kirby Bauer technique and determination of methicillin resistance was performed with oxacillin screening with sodium chloride (NaCl) addition. Of the 151 professionals evaluated, 39 were colonized which showed a prevalence of 25.8%. Among the variables studied, age and use of PPE were associated with colonization by the organism. Of all the isolates, only five were resistant to methicillin.

Ocorrência de Entamoeba histolytica/Entamoeba dispar em pacientes ambulatoriais de Recife, PE

The objective this study was to determine the occurrence of the species Entamoeba histolytica/Entamoeba díspar in clinical samples of ambulatory patients in Pernambuco. A specific assay for Entamoeba histolytica was used in this study, which identified no positive fecal samples among the 213 patients. These results confirm that E. dispar is the dominant species in Pernambuco State.

Prevalence and pathogenicity of Entamoeba histolytica in three different regions of Pernambuco, Northeast Brazil

Parasitological examinations were carried out on 663 individuals of three different cities of Pernambuco State, Northeastern Brazil: Recife, Palmares and Bodocó. The population from a drought area of Pernambuco State, Bodocó, was investigated for amoebiasis and compared with Recife, metropolitan city (about 1.3 million of inhabitants) and another inland community, Palmares, located inside of the sugar-cane plantation region of the State. No evidence of invasive strains of E. histolytica were found in these inhabitants, provided that the isolated zymodemes I, III, IV, VIII, IX, X, XVII and XVIII are recognized as nonpathogenic strains of E. histolytica. Furthermore, the prevalence of intestinal helminths and other protozoan infections showed that these individuals are infected by other agents responsible for diarrhoeal diseases.

Portadores assintomáticos de infecções por Streptococcus pyogenes em duas escolas públicas na cidade do Recife, Pernambuco

OBJECTIVES: assess the prevalence of Streptococcus pyogenes in throat secretions of students from two public elementary schools in Recife. METHODS: an epidemiological and clinical-microbiological study was performed. From two schools, 753 individuals were examined. Throat secretion cultures were performed in blood sheep agar 5% and SBGA strains identified by bacitracin, Pyr and latex agglutination tests. RESULTS: the school children were aged from five to 19 years old, 54,3% of the sample were male and 45,7% female. Six asymptomatic SBGA carriers were identified. Following treatment with penicillin, ASLO sera titration was performed. All carriers had antibody titers below 200UT. CONCLUSIONS: SBGA prevalence rate of 0,8% was estimated in asymptomatic carriers, this percentage was low when compared with other results from similar studies. The authors suggest that epidemiological studies be accomplished to estimate SBGA prevalence in children with pharyngitis and correlation with acute rheumatic fever.
