Isolation and diversity analysis of resistance gene analogues (RGAs) from cultivated and wild strawberries

Degenerate oligonucleotide primers, designed based on conserved regions of Nucleotide Binding Site (NBS) domains from previously cloned plant resistance genes, were used to isolate Resistance Gene Analogues (RGAs) from wild and cultivated strawberries. Seven distinct families of RGAs of the NBS-LRR type were identified from two related wild species, Fragaria vesca and F. chiloensis, and six different Fragaria × ananassa cultivars. With one exception (GAV-3), the deduced amino acid sequences of strawberry RGAs showed strong similarity to TIR (Toll Interleukin I Receptor)-type R genes from Arabidopsis, tobacco and flax, suggesting the existence of common ancestors. GAV-3 seemed to be more closely related to the non-TIR type. Further studies showed that the recombination level and the ratio of non-synonymous to synonymous substitutions within families were low. These data suggest that NBS-encoding sequences of RGAs in strawberry are subject to a gradual accumulation of mutations leading to purifying selection, rather than to a diversifying process. The present paper is the first report on RGAs in strawberry.

Isolation and purification of a 316 Da preformed compound from strawberry (Fragaria ananassa) leaves active against plant pathogens

An antibiotic called fragarin showing activities against bacterial and fungal plant pathogens was isolated and purified by FPLC chromatography from the soluble fraction of strawberry leaves. The molecular weight value determined by mass spectrometry is 316 Da. Fragarin remains fully active after protease treatment or alkaline hydrolysis at 100°C for 20 min. Biological and chemical analyses suggest that fragarin may be a new type of an antimicrobial preformed compound – phytoanticipin – and would constitute a primary non‐specific barrier of strawberry defense.

Global sensitivity analysis in dynamic metabolic networks

Abstract In this work, we have performed global sensitivity analysis on a large-scale dynamic metabolic network through variance-based techniques. Time profiles for sensitivity indices have been calculated for each parameter, based on Sobol’ approach (2001) . The global sensitivity analysis has been carried out on a dynamic model for the Embden–Meyerhof–Parnas pathway, the phosphotransferase system and the pentose-phosphate pathway of Escherichia coli K-12 strain W3110 ( Chassagnole et al., 2002 ). The model comprises eighteen dynamic mass balance equations for extracellular glucose and intracellular metabolites, thirty kinetic rate expressions and seven additional algebraic equations that represent concentration profiles for co-metabolites. Each parameter has been considered to have a normal probability distribution centered on its nominal value and sample sizes of two thousand and five hundred scenarios have been considered. The preceding analysis has allowed identification of eleven parameters as the most influential ones on the complex metabolic network under study.

A strawberry disease caused by Acremonium strictum

We report Acremonium strictum as the causal agent of a new disease in strawberry plants (Fragaria x ananassa Duch.) in the Northwest of Argentina. Both the structure of conidiophores and the sequence spanning the internal transcribed spacers 1 and 2 (ITS1 and ITS2) of the nuclear ribosomal DNA (rDNA) allowed confirming the affiliation of the isolate, corresponding to A. strictum. An analysis of symptoms and lesions caused by the strain of A. strictum in susceptible cultivars showed that the typical symptoms are as follows: in an early stage, small necrotic light-brown spots in leaves and petioles increase in number and size as the disease progresses; in a more advanced stage, dark necrotic areas expand over petioles and leaves causing strangulation of petioles and the plant wilt. Crown rot was not observed even at a very advanced stage of the disease.
