Prevalence of Influenza A viruses in wild migratory birds in Alaska: Patterns of variation in detection at a crossroads of intercontinental flyways

BackgroundThe global spread of the highly pathogenic avian influenza H5N1 virus has stimulated interest in a better understanding of the mechanisms of H5N1 dispersal, including the potential role of migratory birds as carriers. Although wild birds have been found dead during H5N1 outbreaks, evidence suggests that others have survived natural infections, and recent studies have shown several species of ducks capable of surviving experimental inoculations of H5N1 and shedding virus. To investigate the possibility of migratory birds as a means of H5N1 dispersal into North America, we monitored for the virus in a surveillance program based on the risk that wild birds may carry the virus from Asia.ResultsOf 16,797 birds sampled in Alaska between May 2006 and March 2007, low pathogenic avian influenza viruses were detected in 1.7% by rRT-PCR but no highly pathogenic viruses were found. Our data suggest that prevalence varied among sampling locations, species (highest in waterfowl, lowest in passerines), ages (juveniles higher than adults), sexes (males higher than females), date (highest in autumn), and analytical technique (rRT-PCR prevalence = 1.7%; virus isolation prevalence = 1.5%).ConclusionThe prevalence of low pathogenic avian influenza viruses isolated from wild birds depends on biological, temporal, and geographical factors, as well as testing methods. Future studies should control for, or sample across, these sources of variation to allow direct comparison of prevalence rates.

Genetic evidence of intercontinental movement of avian influenza in a migratory bird: the northern pintail (Anas acuta)

The role of migratory birds in the movement of the highly pathogenic (HP) avian influenza H5N1 remains a subject of debate. Testing hypotheses regarding intercontinental movement of low pathogenic avian influenza (LPAI) viruses will help evaluate the potential that wild birds could carry Asian‐origin strains of HP avian influenza to North America during migration. Previous North American assessments of LPAI genetic variation have found few Asian reassortment events. Here, we present results from whole‐genome analyses of LPAI isolates collected in Alaska from the northern pintail (Anas acuta), a species that migrates between North America and Asia. Phylogenetic analyses confirmed the genetic divergence between Asian and North American strains of LPAI, but also suggested inter‐continental virus exchange and at a higher frequency than previously documented. In 38 isolates from Alaska, nearly half (44.7%) had at least one gene segment more closely related to Asian than to North American strains of LPAI. Additionally, sequences of several Asian LPAI isolates from GenBank clustered more closely with North American northern pintail isolates than with other Asian origin viruses. Our data support the role of wild birds in the intercontinental transfer of influenza viruses, and reveal a higher degree of transfer in Alaska than elsewhere in North America.

EFFECTS OF CHRONIC DIETARY LEAD IN AMERICAN KESTRELS (FALCO SPARVERIUS)

American kestrels were fed a diet containing 0, 10, or 50 ppm lead (Pb) powder for at least 5 mo. Blood δ-aminolevulinic acid dehydratase (ALAD) activity in birds receiving 50 ppm Pb was as low as 20% of controls but no significant effects were noted in packed cell volume (PCV) or hemoglobin concentration (Hb). Mean liver Pb residues in birds fed 50 ppm Pb were 1.3 and 2.4 ppm (dry wt) for males and females, respectively. Liver Pb residues in birds fed 10 ppm Pb were not significantly greater than controls. There was no significant correlation between blood ALAD activity and blood Pb concentration, no consistent histopathological lesions were noted, and body and organ weights were not affected.

Avian influenza at both ends of a migratory flyway: characterizing viral genomic diversity to optimize surveillance plans for North America

Although continental populations of avian influenza viruses are genetically distinct, transcontinental reassortment in low pathogenic avian influenza (LPAI) viruses has been detected in migratory birds. Thus, genomic analyses of LPAI viruses could serve as an approach to prioritize species and regions targeted by North American surveillance activities for foreign origin highly pathogenic avian influenza (HPAI). To assess the applicability of this approach, we conducted a phylogenetic and population genetic analysis of 68 viral genomes isolated from the northern pintail (Anas acuta) at opposite ends of the Pacific migratory flyway in North America. We found limited evidence for Asian LPAI lineages on wintering areas used by northern pintails in California in contrast to a higher frequency on breeding locales of Alaska. Our results indicate that the number of Asian LPAI lineages observed in Alaskan northern pintails, and the nucleotide composition of LPAI lineages, is not maintained through fall migration. Accordingly, our data indicate that surveillance of Pacific Flyway northern pintails to detect foreign avian influenza viruses would be most effective in Alaska. North American surveillance plans could be optimized through an analysis of LPAI genomics from species that demonstrate evolutionary linkages with European or Asian lineages and in regions that have overlapping migratory flyways with areas of HPAI outbreaks.

Intercontinental reassortment and genomic variation of low pathogenic avian influenza viruses isolated from northern pintails ( Anas acuta ) in Alaska: examining the evidence through space and time

Migration and population genetic data for northern pintails (Anas acuta) and phylogenetic analysis of low pathogenic avian influenza (LPAI) viruses from this host in Alaska suggest that northern pintails are involved in ongoing intercontinental transmission of avian influenza. Here, we further refine this conclusion through phylogenetic analyses which demonstrate that detection of foreign lineage gene segments is spatially dependent and consistent through time. Our results show detection of foreign lineage gene segments to be most likely at sample locations on the Alaska Peninsula and least likely along the Southern Alaska Coast. Asian lineages detected at four gene segments persisted across years, suggesting maintenance in avian hosts that migrate to Alaska each year from Asia or in hosts that remain in Alaska throughout the year. Alternatively, live viruses may persist in the environment and re-infect birds in subsequent seasons.

Use of Serum Biochemistry to Evaluate Nutritional Status and Health of Incubating Common Eiders (Somateria mollissima) in Finland

During 1997–1999, we collected serum samples from 156 common eider (Somateria mollissima) females incubating eggs in the Finnish archipelago of the Baltic Sea. We used serum chemistry profiles to evaluate metabolic changes in eiders during incubation and to compare the health and nutritional status of birds nesting at a breeding area where the eider population has declined by over 50% during the past decade, with birds nesting at two areas with stable populations. Several changes in serum chemistries were observed during incubation, including (1) decreases in serum glucose, total protein, albumin, β‐globulin, and γ‐globulin concentrations and (2) increases in serum uric acid, creatine kinase, and β‐hydroxybutyrate concentrations. However, these changes were not consistent throughout the 3‐yr period, suggesting differences among years in the rate of carbohydrate, lipid, and protein utilization during incubation. The mean serum concentrations of free fatty acids, glycerol, and albumin were lowest and the serum α‐ and γ‐globulin levels were highest in the area where the eider population has declined, suggesting a role for nutrition and diseases in the population dynamics of Baltic eiders.

Lead fishing weights and other fishing tackle in selected waterbirds

Abstract From 1995 through 1999, 2,240 individuals of 28 species of waterbirds were examined in the United States for ingested lead fishing weights. A combination of radiography and visual examination of stomachs was used to search for lead weights and blood and liver samples from live birds and carcasses, respectively, were collected for lead analysis. Ingested lead weights were found most frequently in the Common Loon (Gavia immer) (11 of 313 = 3.5%) and Brown Pelican (Pelecanus occidentalis) (10 of 365 = 2.7%), but also in one of 81 (1.2%) Double-crested Cormorants (Phalacrocorax auritus) and one of 11 (9.1%) Black-crowned Night Herons (Nycticorax nycticorax). Birds with ingested lead fishing weights (including split shot, jig heads, and egg, bell, and pyramid sinkers) were found in California, Florida, Maine, New Hampshire, North Carolina, and Wisconsin. The size and mass of ingested lead weights ranged from split shot of 7 mm in the longest dimension, weighing less than 2 g, to a 22 × 39 mm pyramid sinker that weighed 78.2 g. Six ingested lead weights were more than 25.4 mm in the longest dimension. Lead concentrations in the blood and liver of birds with lead fishing weights in their stomachs ranged up to 13.9 ppm and 26.0 ppm (wet weight basis), respectively. During the study, we also noted the presence of ingested or entangled fishing tackle, with no associated lead weights, in eight species.

Surveillance for High Pathogenicity Avian Influenza Virus in Wild Birds in the Pacific Flyway of the United States, 2006–2007

Abstract In 2006 the U.S. Department of Agriculture, U.S. Department of Interior, and cooperating state fish and wildlife agencies began surveillance for high-pathogenicity avian influenza (HPAI) H5N1 virus in wild birds in the Pacific Flyway of the United States. This surveillance effort was highly integrated in California, Oregon, Washington, Idaho, Nevada, Arizona, Utah, and western Montana, with collection of samples coordinated with state agencies. Sampling focused on live wild birds, hunter-killed waterfowl during state hunting seasons, and wild bird mortality events. Of 20,888 samples collected, 18,139 were from order Anseriformes (waterfowl) and 2010 were from order Charadriiformes (shorebirds), representing the two groups of birds regarded to be the primary reservoirs of avian influenza viruses. Although 83 birds were positive by H5 real-time reverse transcription polymerase chain reaction (rRT-PCR), no HPAI H5N1 virus was found. Thirty-two virus isolates were obtained from the H5-positive samples, including low-pathogenicity H5 viruses identified as H5N2, H5N3, and H5N9.

Isolation and Characterization of a Reovirus from Common Eiders (Somateria mollissima) from Finland

SUMMARY. Samples of brain, intestine, liver, lung, spleen, and bursa of Fabricius were collected from five common eider (Somateria mollissima) duckling carcasses during a die-off in the western Gulf of Finland (59°50′N, 23°15′E) in June 1996. No viral activity was observed in specific-pathogen-free chicken embryos inoculated with tissue suspensions, but samples of bursa of Fabricius from three birds were positive when inoculated into Muscovy duck (Cairina moschata) embryo fibroblasts. The isolates were characterized as nonenveloped RNA viruses and possessed several characteristics of the genus Orthoreovirus. Virus particles were icosahedral with a mean diameter of 72 nm and were stable at pH 3.0; their genome was separated into 10 segments by polyacrylamide gel electrophoresis. Mallard (Anas platyrhynchos) ducklings experimentally infected with the eider reovirus showed elevated serum activities of aspartate aminotransferase, creatine kinase, and lactate dehydrogenase enzymes and focal hemorrhages in the liver, spleen, and bursa of Fabricius. During 1997–99, the prevalence of neutralizing antibodies to the isolated virus ranged from 0 to 86% in 302 serum samples collected from incubating eider hens at three nesting areas along coastal Finland. The highest seroprevalence was found in Hanko in 1999, just weeks before reports of an uninvestigated mortality event resulting in the death of an estimated 98% of ducklings at that location. These findings raise the question of potential involvement of the virus in poor duckling survival and eider population declines observed in several breeding areas along coastal Finland since the mid-1980s.

ENZYME ACTIVITIES IN PLASMA, LIVER AND KIDNEY OF BLACK DUCKS AND MALLARDS

Activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST), creatine phosphokinase (CPK), alkaline phosphatase (ALP), and lactate dehydrogenase (LDH) were measured in plasma, liver, and kidney, and gammaglutamyl transferase (GGT) was measured in liver and kidney of black ducks (Anas rubripes). Activities of ALT, AST, GGT, and ornithine carbamyl transferase (OCT) were assayed in plasma, liver, and kidney of game-farm mallards (Anas platyrhynchos). Appreciable OCT and AST activity occurred in both liver and kidney. Activities of ALT, CPK, ALP and GGT were higher in kidney, while LDH was higher in liver. GGT was detected in plasma from one of four mallards.
