Pharmacologic Manipulation of Fertility

The professional application of agents to the manipulation of fertility of cows requires basic and applied knowledge of the physiologic mechanisms that are affected and of the pharmacologic agents that are used. In all areas of the pharmacologic manipulation of fertility, the achievement is less than the ideal, and further research is required to improve the efficiency of treatments. The induction of estrus in acyclic animals can involve a reduction in the depth of anestrus, pretreatment with progestagen to ensure estrous behavior and the formation of a normal corpus luteum, and then treatment with exogenous gonadotropin. Responsiveness to treatment can be variable and reflects the depth of anestrus of the animals. Improved treatment regimens require a knowledge of the basic mechanisms involved with the depth of anestrus, a means of assessing the depth of anestrus, and an understanding of the hormonal requirements of ovarian follicles for development and maturation in animals at different depths of anestrus. The optimal precision in the synchronization of estrus (and ovulation) in cyclic animals requires the synchronization of both follicular waves and the end of progestational phase. The end of progestational phase can be synchronized effectively using prostaglandin F2a (or analogs), or by treatment with progestagens with or without luteolytic agents. Procedures to synchronize follicular waves need to be established. The induction of superovulation can be achieved readily using gonadotropins prior to estrus synchronization using prostaglandin F2a. The responses to standard treatments in terms of ovulation rates and yield of transferable embryos are highly variable. The development of procedures to reduce this variability requires an understanding of the intra-ovarian mechanisms involved in recruitment of follicles for a wave of follicular growth, in the selection of dominant follicles for further development, and in the mechanisms controlling follicular atresia. Cystic ovarian disease can be treated effectively using HCG or GnRH (follicular cysts) or prostaglandin F2a (luteal cysts). The basic mechanisms resulting in failure of estrogen positive feedback on LH secretion (that results in cystic follicles) remain to be determined. Small but significant increases in pregnancy rates can be achieved treating cows with prostaglandin during the post-partum period, with prostaglandin to induce estrus for insemination, with GnRH or HCG at estrus, and with GnRH or progestagen treatment during diestrus. Beneficial effects of treatment have been shown in some trials but not in others.(ABSTRACT TRUNCATED AT 400 WORDS)

Use of postpartum vaginoscopic (visual vaginal) examination of dairy cows for the diagnosis of endometritis and the association of endrometritis with reduced reproductive performance

OBJECTIVE To assess the association between the findings from vaginoscopic examination of postpartum dairy cows with peripartum reproductive disease and their subsequent reproductive performance, and to determine the factors that might be associated with a positive vaginoscopic examination (VV). METHODS Cows (n=1325) from 17 seasonal calving dairy herds were enrolled if they had been calved at least 7 days and had at least one of the following conditions that placed them at risk of endometritis: retained fetal membranes (RFM), dystocia, a dead calf, hypocalcaemia, twin birth, calving induction or an observed vulval discharge (VD). Examination was undertaken 28-37 days before mating start date (MSD) within each herd. All cows were body condition scored (BCS) and were VV scored on a scale of 0 (clear) to 3 (purulent). The uterus and ovaries were assessed by rectal palpation. RESULTS Cows with a positive VV score (1-3) were less likely to conceive to first service and had a lower pregnancy proportion than cows that had a negative VV score (score 0). Mean MSD to conception interval was longer in VV-positive cows. A positive VV score was associated with a low BCS, primiparity, intrapelvic uterus, poor uterine tone, large uterus, RFM, VD, dystocia, dead calf and twins. CONCLUSION In the present study a positive VV score was associated with reduced reproductive performance and was more common in primiparous and low BCS cows. Of the at-risk cows, those with RFM, VD, dystocia, dead calf or twins were more likely to have a positive VV score.

Comparison of two methods of detecting purulent vaginal discharge in postpartum dairy cows and effect of intrauterine cephapirin on reproductive performance

OBJECTIVE Part 1: compare the use of a Metricheck device (a stainless steel probe with a semi-spherical rubber cup attached at one end) to sample the contents of the anterior vagina with a vaginal speculum examination for the diagnosis of pus in the vagina of postpartum dairy cows and to investigate the association of that pus with reproductive performance. Part 2: assess the effect of a single intrauterine infusion of 500 mg cephapirin in cows diagnosed with vaginal purulent or mucopurulent discharge 7 to 28 days after calving on reproductive performance. PROCEDURE Six herds were visited fortnightly to examine cows that had calved between 7 and 28 days (n = 423) with both the Metricheck device and a vaginal speculum to score, by each method, the vaginal discharge from 0 (clear or absent) to 3 (purulent) for each animal included in the study. Half of the cows that had a positive discharge score (1 to 3 by either examination method) were then treated with an intrauterine infusion of 500 mg of cephapirin. The relationship between Metricheck score, vaginoscopy score, treatment and reproductive performance was assessed. RESULTS There was a substantial measure of agreement between each method when scores were analysed by status. Cows that were positive with either method had inferior reproductive performance compared with cows with a score of zero. Treatment of cows diagnosed with a purulent or mucopurulent discharge with intrauterine cephapirin improved reproductive performance in both the vaginoscopy and Metricheck groups.

Oriental theileriosis in dairy cows causes a significant milk production loss

BackgroundOriental theileriosis is a tick-borne, protozoan disease of cattle caused by members of the Theileria orientalis-complex. Recent outbreaks of this disease in eastern Australia have caused major concerns to the dairy and beef farming communities, but there are no published studies of the economic impact of this disease. On a farm in Victoria, Australia, we assessed whether oriental theileriosis has an impact on milk production and reproductive performance in dairy cows.MethodsBlood samples collected from all 662 cows on the farm were tested using an established molecular test. For individual cows, milk production and reproductive performance data were collected. A clinical assessment of individual cows was performed. Based on clinical findings and molecular test results, the following groups of cows were classified: group 1, with cardinal clinical signs of oriental theileriosis and molecular test-positive for T. orientalis; group 2, with mild or suspected signs of theileriosis and test-positive; group 3, with no clinical signs and test-positive; and group 4, with no clinical signs and test-negative. Milk production and reproductive performance data for groups 1, 2 and 3 were each compared with those for group 4 using linear and logistic regression analyses, respectively.ResultsAt 100 days of lactation, group 1 cows produced significantly less milk (288 l; P = 0.001), milk fat (16.8 kg; P < 0.001) and milk protein (12.6 kg; P < 0.001) compared with group 4. At this lactation point, group 2 also produced significantly less milk fat (13.6 kg; P = 0.002) and milk protein (8.6 kg; P = 0.005) than group 4. At 305 days of lactation, group 1 cows produced significantly less milk (624 l; P = 0.004), milk fat (42.9 kg; P < 0.001) and milk protein (26.0 kg; P < 0.001) compared with group 4 cows. Group 2 cows also produced significantly less milk fat (21.2 kg; P = 0.033) at this lactation point. No statistically significant difference in reproductive performance was found upon pairwise comparisons of groups 1–3 with group 4 cows.ConclusionsThe present findings demonstrate that clinical oriental theileriosis can cause significant milk production losses in dairy cattle.

Identification of Streptococcus uberis Multilocus Sequence Types Highly Associated with Mastitis

ABSTRACT Multilocus sequence typing analysis of Streptococcus uberis has identified a cluster of isolates associated with clinical and subclinical mastitis and a cluster associated with cows with low somatic cell counts in their milk. Specific groups of genotypes (global clonal complex [GCC] sequence type 5s [ST5s] and GCC ST143s) were highly associated (P = 0.006) with clinical and subclinical mastitis and may represent a lineage of virulent isolates, whereas isolates belonging to GCC ST86 were associated with low-cell-count cows. This study has, for the first time, demonstrated the occurrence of identical sequence types (ST60 and ST184) between different continents (Australasia and Europe) and different countries (Australia and New Zealand). The standardized index of association and the empirical estimation of the rate of recombination showed substantial recombination within the S. uberis population in Australia, consistent with previous multilocus sequence type analyses.

Effect of intrauterine treatment with cephapirin on the reproductive performance of seasonally calving dairy cows at risk of endometritis following periparturient disease

OBJECTIVE To determine the effect of intrauterine cephapirin (0.5 g) treatment on the reproductive performance of cows diagnosed as at risk of developing endometritis. METHODS Cows (n = 1325) from 17 seasonal calving dairy herds were enrolled if they had been calved for more than 6 days and had a condition that placed them at risk of endometritis. Half were treated with intrauterine cephapirin at an examination performed 28-37 days prior to mating start date (MSD). Cows were scored for body condition, their vaginal discharge was scored using vaginoscopy (VV), and the uterus and ovaries were assessed by rectal palpation. RESULTS Overall, there was no significant treatment effect on reproductive performance. In 945 modified at risk (MAR) cows the effect of treatment varied with the calving to treatment interval (CTI) and VV status. For the subset of 229 MAR cows with CTI < or = 28 days, treatment improved the proportions of first-service conception (OR 3.1; P < 0.01) and 6-week in-calf (OR 2.1; P < 0.05), and reduced the mean MSD to conception interval by 20 days (P < 0.05). The difference was greater in cows with a positive VV score (1-3). Treated VV-negative cows with CTI > 48 days had a reduced proportion of cows in-calf by 6 weeks (OR 0.58; P = 0.056) compared with untreated cows. CONCLUSION Intrauterine cephapirin infusion of MAR cows improved reproductive performance, depending on CTI. The greater treatment effect in VV-positive cows suggests that VV scoring is a useful tool for the diagnosis of endometritis.

The use of an internal teat sealant in combination with cloxacillin dry cow therapy for the prevention of clinical and subclinical mastitis in seasonal calving dairy cows.

Cows (n=2,053) from 6 seasonally calving dairy herds were enrolled in a trial to compare the efficacy of 2 dry cow treatments. Cows received either a combination dry cow therapy of 600 mg of cloxacillin (CL) followed by an internal teat sealant (ITS) containing 2.6 g of bismuth subnitrate in all 4 quarters immediately following their final milking for the season, or only an intramammary infusion of 600 mg of CL. All cases of clinical mastitis were recorded and cultured during the first 150 d of lactation in each herd, and cow somatic cell count (SCC) was measured between 7 and 50 d postcalving. A large difference was found between treatment groups in the rate at which cows were diagnosed with clinical mastitis over the first 21 d of lactation, after which time the rate at which cows were diagnosed with clinical mastitis was similar between treatment groups. Analysis of the relative proportions of cows with clinical mastitis was performed at both the gland and cow levels. The relative risk (RR) of clinical mastitis diagnosed within 21, 30, and 100 d of calving in a gland treated with the ITS-CL combination was, respectively, 0.30 [95% confidence interval (CI)=0.21-0.44], 0.39 (0.28-0.53), and 0.58 (0.46-0.75) that of the CL group. An interaction between treatment and previous SCC was found when clinical mastitis was analyzed at the cow level. In a subset of cows that had low SCC in their previous lactation, the RR of mastitis in cows with the ITS-CL combination within 21, 30, and 100 d of calving was, respectively, 0.54 (95% CI=0.33-0.87), 0.57 (0.37-0.88), and 0.69 (0.50-0.99) that of cows that received only CL at drying off. In the subset of cows that had at least 1 high SCC in the previous lactation, the RR of mastitis in the ITS-CL combination group within 21, 30, and 100 d of calving was, respectively, 0.26 (95% CI=0.16-0.44), 0.37 (0.24-0.57), and 0.72 (0.55-0.96) that of the CL-only group. The ITS-CL combination of dry cow treatments was associated with a reduction in subclinical mastitis [SCC ≥250,000 cells/mL; RR=0.80 (95% CI=0.65-0.98)] when compared with treatment with CL alone. The use of an ITS in combination with CL dry cow treatment was associated with significantly lower clinical and subclinical mastitis in the following lactation, with a greater difference found in cows that had a history of subclinical mastitis in the previous lactation.

Molecular epidemiology of recurrent clinical mastitis due to Streptococcus uberis: Evidence of both an environmental source and recurring infection with the same strain

This study was undertaken because clinicians and farmers have observed that a considerable number of cows diagnosed with Streptococcus uberis mastitis have recurrences of mastitis in the same or a different quarter. The study was an attempt to answer whether these recurring cases were due to treatment failure (in which case a search would have begun for a better treatment for Strep. uberis mastitis) or due to reinfection with a different strain of Strep. uberis. Using pulsed-field gel electrophoresis (PFGE), we determined that the majority of recurrences (20 of 27) were caused by a new strain of Strep. uberis, indicating that treatment of the initial infection had been successful. A small number of recurrences (5 of 27) were caused by the initial strain, indicating persistence. The remaining 2 recurrences occurred in a new quarter but with the initial strain of Strep. uberis, indicating either spread between quarters or reactivation of a previous subclinical infection. Analysis of the PFGE profiles failed to reveal any strain-specific propensity to persist, because strains causing recurrences occurred in most of the major clusters.

Use of multiplexed tandem PCR to estimate the prevalence and intensity of Theileria orientalis infections in cattle

This study employed a semi-quantitative, multiplexed tandem PCR (MT-PCR) to assess the prevalence and infection intensity of four genotypes (buffeli, chitose, ikeda and type 5) of Theileria orientalis in cattle in Australia. Genomic DNA samples from blood samples (n=448) collected from 27 to 32 dairy cows from each of 15 dairy herds with a history of recent theileriosis outbreaks (Group 1), and from blood samples available from 24 cows with or without oriental theileriosis (Group 2) were tested using MT-PCR. Results revealed that all four genotypes were present in Group 1 cattle; genotype buffeli had the highest prevalence (80.5%), followed by genotypes ikeda (71.4%), chitose (38.6%) and type 5 (20.3%). Genotype ikeda had the highest average infection intensity in the cattle (relating to 55,277 DNA copies), followed by buffeli, chitose and type 5 (6354-51,648 copies). For Group 2, results indicated that genotype ikeda had a significantly higher average intensity of infection than buffeli in symptomatic cattle (P<0.001), and symptomatic cattle had a higher intensity of ikeda than asymptomatic cattle (P=0.004). Future studies should assess the utility of the present MT-PCR assay as a diagnostic and epidemiological tool in other parts of Australasia and the world.

The effects on claw health of supplement feeding grazing dairy cows on feed pads.

The effects of feeding and management systems on the health and welfare of grazing dairy cows were investigated by comparing the claw health of cows fed grain during milking and pasture silage in the paddock (Control), with cows fed a grain-based partial mixed ration (PMR) on a concrete feed pad. Cows were assessed on three occasions during lactation: (1) early lactation (20-81 days in milk [DIM]) before allocation to feeding treatments; (2) mid-lactation (97-158 DIM) immediately following an intensive feeding experiment, and (3) late lactation (173-243 DIM) several months after return to initial management groups. At the final examination, claw puncture resistance was measured. The results showed that for the most prevalent lesions (white line disease, paintbrush haemorrhage and traumatic bruising), there was no effect of feeding system or amount of supplement on the presence of the moderate to severe forms in early lactation, but cows were more likely to have a particular lesion at the second assessment if it was present in early lactation. Puncture resistance of the claw was not related to presence of a lesion for any of the most prevalent lesion types. It was concluded for this herd that for most indicators of claw health, there was no overall effect of different feeding systems (supplement fed during milking or on a feed pad) or amount of supplement.
