The influence of modified 14-3-3 protein synthesis in potato plants on the nutritional value of the tubers

Abstract The recently created six transgenic potato genotypes with overexpressed or underexpressed P14-3-3a (29G) and P14-3-3c (20R) isoforms of 14-3-3 protein were field-trialled (1998–2001). The contents of protein, starch, reducing sugars, sucrose and lipids were determined in the transgenic and control tubers harvested from the field. The obtained results showed a significant increase in crude protein content in potatoes with repression of P14-3-3c isoform and in potatoes with blocked P14-3-3a synthesis in comparison to the control line. A stable increase in lipid content of potatoes with overexpression of 14-3-3 protein from Cucurbita pepo in the field trials was observed. The variability of the investigated genotypes, in respect to the nutritional components, was statistically analysed using discriminant function and cluster analyses. The dominant influence of the variability of the genotypes exerted significant differentiation of protein, lipid and starch contents. These components showed the greatest discriminant power in the variability of genotypes. These results confirm the suggestion that 14-3-3 protein co-ordinates primary metabolite synthesis in plants.

Boron content in daily meals for preschool children and school youth

Boron content in daily me als for preschool children and school youth was studied in this research. The boron content was measured in the meals taken from Wroclaw preschool and boarding school in December, March, June, and September. The whole meal was homogenized. Four grams of sample were mineralized and analyzed by inductively coupled plasma-optical emission spectroscopy (ICP-OES). Statistical assessment of the results was made with the Statistica v.5.1 program. The recovery was determined by the use of the standard additions method to evaluate the accuracy of the procedure. The mean boron content in 1 kg dry weight of a preschool meal was 2.31 mg B/kg dry wt, and in boarding-school meals, it was 2.15 mg B/kg dry wt. Based on these results, the estimated daily boron intake was calculated. The daily boron intake from food by preschool children was found to be 1.01 mg B/d for children and 1.2 mg B/d for youth. The actual intake of boron from food was determined after having related the quantity of the element to 1 kg of body weight. In the case of children, the mean intake of boron was 0.05 mg B/kg body wt/d, whereas in teenagers, it was 0.016 mg B/kg body wt/d. The results of this research show no significant differences between daily boron intake for preschool children and boarding-school youth.

Fe, Ca and Mg contents in selected fast food products in Poland

The Fe and Mg contents in selected fast food products available in restaurants and fast food outlets in Poland were determined by AAS, and the Ca content by AES. The mean Fe contents in the studied fast food products were from 0.7 to 2.3 mg/100 g, or from 0.6 to 2.3 per single serving. The highest means for this element were found in a serving of hamburger (2.3 mg), fishburger (2.0 mg) and chicken sandwich (2.0 mg). The mean Ca contents in the studied products were from 11.6 to 192.2 mg/100 g, or 10 to 192.2 mg per serving. The highest means for this element were found in a serving of pizza (192.2 mg) and cheeseburger (134.8 mg). The mean Mg contents in the studied products were from 6.8 to 34.1 mg/100 g1 or 5.9 to 37.3 mg per serving. The highest means for this element were found in a serving of french fries (37.3 mg), chicken sandwich (34.7 mg) and fishburger (30.4 mg). Based on the Fe, Ca and Mg contents found in these products, the percentage of the Recommended Dietary Allowance (RDA) of these elements was calculated for one serving of each product. These calculations were done for various groups of people in Poland. The highest percentage of the recommended Fe intake could be covered by one serving of hamburger (15-23% RDA), fishburger (14-20% RDA), or chicken sandwich (13-20% RDA). The highest percentage of the recommended Ca intake could be covered by one serving of pizza (17-24% RDA) or cheeseburger (12-17% RDA); and for Mg one serving of french fries (11-19% RDA), chicken sandwich (10-17% RDA), or fishburger (9-15% RDA). From the conducted studies it may be concluded that some fast food products can serve as a source of Fe, Ca and Mg in the diet of people of various ages.

Dietary carbohydrate content influences boron's effect on lipid and protein indices in rats.

The aim of the study was to establish whether an excess or deficiency of dietary carbohydrates would influence the effect of boron (B) on lipid, protein, and glucose metabolism in laboratory rats. Sixty male Buffalo rats were used in the study, divided into six groups fed a control diet or a low- or high-carbohydrate diet with or without a B supplement (3 mg B/kg fodder). The hemoglobin concentration and hematocrit were assessed in whole-blood samples, and the total protein, albumin, creatine, glucose, total lipid, and high-density lipoprotein (HDL) choloesterol contents in the serum were established. The total cholesterol and triacylglycerol contents in liver lipid extracts were also measured.A low carbohydrate content in a B-supplemented diet led to an increase in the total protein and albumin contents in the serum of the rats compared to the levels for the rats on the control and high-carbohydrate B-supplemented diets. Under conditions of an excess or deficiency of carbohydrates in the diet, B did not significantly influence the cholesterol and total lipid contents in the serum. Boron’s influence on the other metabolic indexes (glucose, HDL cholesterol, and triacyglycerol contents in the serum; cholesterol and triacylglycerol contents in the liver) was unaffected by the carbohydrate content in the diet.

Occurrence of dietary risk factors in inflammatory bowel disease:Influence on the nutritional status of patients in clinical remission

BACKGROUND Among the complex factors that may favor the occurrence of inflammatory bowel disease (IBD), genetic, immunological and environmental initiators, including nutritional factors, are listed. So far, there have been no previous studies on the type and frequency of dietary risk factors for IBD in Poland and their effect on the nutritional status of patients. OBJECTIVES The aim of the study was to assess the influence of the frequency and type of dietary risk factors for IBD on the nutritional status of patients with ulcerative colitis (UC) and Crohns disease (CD). MATERIAL AND METHODS In the study, the dietary habits and nutritional status of patients were assessed using the cross-check dietary history method and the Mini Nutritional Assessment (MNA) questionnaire. The study group consisted of 162 IBD patients: 61 individuals with CD and 101 with UC. The data was compared to the results of a control group (129 healthy volunteers). RESULTS The results obtained showed that IBD patients during a period of remission disclosed such dietary risk factors as inadequate consumption of fiber and excessive consumption of red meat and meat products, animal fats, and sugars in comparison to the control group. Only low fiber intake was associated with a worse nutritional status of patients with UC. No consistent influence of the number of IBD dietary risk factors on the nutritional status of patients was found. CONCLUSIONS The nutritional status of IBD patients in remission was related to the type of dietary risk factors, but did not depend on the number of them.

The usefulness of routinely used malnutrition screening tools in predicting anemia in lung cancer patients

BACKGROUND Anemia and malnutrition are frequently observed during lung cancer development, and the associations between them have been researched. However, no study concerning the utility of routinely used nutritional screening tools in predicting anemia in lung cancer has been performed. OBJECTIVES The aim of this study was to assess the usefulness of routinely used malnutrition screening tools in predicting anemia in lung cancer patients. MATERIAL AND METHODS Eighty-five male patients were recruited to this study. Blood counts, serum iron concentration, total iron binding capacity (TIBC) and serum transferrin saturation (STS), measurements of selected anthropometric parameters, Mini Nutritional Assessment (MNA) and Glasgow Prognostic Score (GPS) were performed for the subjects. To evaluate the differences in the distribution of hematological and iron status parameters according to nutritional status, a t-test (Mann-Whitney U test for non-parametric data) and an analysis of variance (ANOVA) were performed. Tukeys post hoc test was performed for intergroup comparison of parametric data. The sensitivity, specificity, positive and negative predictive values of MNA and GPS were compared to blood counts and biochemical parameters of iron status. RESULTS Using the MNA test, we observed that ca. 60% of subjects had deteriorated nutritional status. About half of the patients had inflammation cumulated with malnutrition. A similar part of the subjects had anemia. The MNA test showed a significant difference in the distribution of Hb and Htc, while GPS showed the distribution of Fe and TIBC among lung cancer patients. We did not observe any influence of fat-free mass index (FFMI) on hematological and iron status parameters. The MNA test had very high specificity and positive predictive values (PPV) for all the hematological parameters evaluated as well as GPS for serum Fe concentration and TIBC. CONCLUSIONS Our data demonstrates that an evaluation of nutritional status with the MNA test can provide additional predictive information regarding anemia, while GPS may do the same with type of anemia in lung cancer patients.

Effect of high‐fat cholesterol enriched diets on hypolipemic action of Oenothera paradoxa oil in rats. Part 1. Blood serum and liver lipids

The effect on Oenothera paradoxa oil on blood serum and liver lipids metabolism in rats fed a semisynthetic high-fat cholesterol enriched diets was investigated. The source of fats was sunflower oil or lard in 15% quantities and the source of protein was soybean protein isolate in 27% quantity. The diets were enriched with 0.5% cholesterol. This dietary experiment was carried on for 8 weeks. For the first 4 weeks rats were fed standard diet and for the next 4 weeks Oe. paradoxa oil (300 mg/day/rat) was additionally given by stomach-tube. At the end of experiment the contents of total cholesterol, HDL-cholesterol, triglycerides and free fatty acids in blood serum as well as cholesterol and triglycerides level in liver were determined. It was found that the addition of cholesterol to the diet decreased the hypocholesterolemic and hypotriglyceridemic effects of Oe. paradoxa oil both in blood serum and liver. It have not had any significant effect on the free fatty acid concentration in blood serum decrease by Oe. paradoxa oil intake.
