Separation of polyphenols in Canary Islands wine by capillary zone electrophoresis without preconcentration

Jiří Pazourek; G. González; Alma L Revilla; Josef Havel

A method for separation and determination of polyphenols in wine by capillary zone electrophoresis (CZE) without any preconcentration step is described. The sensitivity and limits of detection for gentisic and p-coumaric acid are better than those previously published. The effect of a possible C18 solid-phase extraction prior to the CZE analysis was examined. The developed optimized method (without any extraction step) was applied to the analysis of wines from Tenerife, Canary Islands.

Electrophoretic behavior of adamantane derivatives possessing antiviral activity and their determination by capillary zone electrophoresis with indirect detection

Nad’a Reichová; Jiří Pazourek; Pavla Polášková; Josef Havel

Separation and determination of adamantane derivatives with antiviral activity, namely amantadine (1‐adamantan amine), memantine (1‐amino‐3,5‐dimethyl adamantane) and rimantadine (α‐methyl‐1‐adamantane methylamine), were examined by capillary zone electrophoresis. After optimization, an indirect detection method using 5 mM 4‐methylbenzylamine in ethanol/water solution (1:4) as simultaneously absorbing and buffering background electrolyte with detection at 210 nm was found suitable for determination of the individual compounds (limit of detection was 0.35 mgL–1 for memantine hydrochloride, S/N = 3). Baseline separation of all the three compounds was reached by addition of α‐ or β‐cyclodextrins to the electrolyte in concentrations of 20 and 2 mM, respectively.

Simultaneous determination of trans-resveratrol and sorbic acid in wine by capillary zone electrophoresis

Zuzana Dobiášová; Jiří Pazourek; Josef Havel

A method for separation and determination of sorbic acid, a food and beverage preservative, and trans‐resveratrol, a biomedically active substance, in wine by capillary zone electrophoresis is described. A solid‐phase extraction step on C18‐column prior to the electrophoretic separation providing lower detection limits was used for trans‐resveratrol determination. For determination of sorbic acid direct analysis of wine (without a preconcentration step) was used. The method is rapid and sensitive and was applied to the analysis of wines from Alsace, France.

Validation of a Capillary Zone Electrophoresis Method for Determination of Rimantadine Hydrochloride in Rimantadin100 Tablets and the Method Application to Dissolution Test Monitoring

Jiří Pazourek; Alma L Revilla; Dagmar Gajdošová; Josef Havel

A capillary zone electrophoretic method with indirect UV‐detection for determination of rimantadine, an antiviral drug against influenza A, in tablets was validated. Instrumental precision, the method precision, accuracy, calibration curve linearity, selectivity, robustness, and time stability of the sample and the standard were tested. The method was also applied to monitor dissolution tests of the tablets. The possibility of addition of an internal standard for improvement of the method precision was discussed.

Minimization of adsorption in capillary zone electrophoresis of proteins and neuroprotective peptides

Ondřej Šulák; Miroslava Spanilá; Dagmar Gajdošová; Jiří Pazourek; Josef Havel

Four types of coating procedures suitable for protein and peptides separation: covalent coating using polyethyleneimine (PEI), 3-glycidoxy-propyltrimethoxysilane (GPTMS), 3-aminopropyltriethoxysilane (APS) and a new approach of dynamic coating with linear polyacrylamide (LPA) have been studied. The capillary surface was characterized by determination of the electroosmotic flow (EOF); for optimization a model mixture of proteins (cytochrome C, lysozyme and ribonuclease A) was used. Separation of a neuroprotective peptide [Gly-14]humanin was demonstrated in the optimized system.
