PER3 Polymorphism Predicts Sleep Structure and Waking Performance

Circadian rhythmicity and sleep homeostasis interact to regulate sleep-wake cycles [1-4], but the genetic basis of individual differences in sleep-wake regulation remains largely unknown [5]. PERIOD genes are thought to contribute to individual differences in sleep timing by affecting circadian rhythmicity [6], but not sleep homeostasis [7, 8]. We quantified the contribution of a variable-number tandem-repeat polymorphism in the coding region of the circadian clock gene PERIOD3 (PER3) [9, 10] to sleep-wake regulation in a prospective study, in which 24 healthy participants were selected only on the basis of their PER3 genotype. Homozygosity for the longer allele (PER3(5/5)) had a considerable effect on sleep structure, including several markers of sleep homeostasis: slow-wave sleep (SWS) and electroencephalogram (EEG) slow-wave activity in non-rapid eye movement (non-REM) sleep and theta and alpha activity during wakefulness and REM sleep were all increased in PER3(5/5) compared to PER3(4/4) individuals. In addition, the decrement of cognitive performance in response to sleep loss was significantly greater in the PER3(5/5) individuals. By contrast, the circadian rhythms of melatonin, cortisol, and peripheral PER3 mRNA expression were not affected. The data show that this polymorphism in PER3 predicts individual differences in the sleep-loss-induced decrement in performance and that this differential susceptibility may be mediated by its effects on sleep homeostasis.

Sleep quantity, sleep difficulties and their perceived consequences in a representative sample of some 2000 British adults

Sleep problems and sleep restriction are popular topics of discussion, but few representative data are available. We document Britains sleep based on a nationally representative sample of 1997, 16–93 year olds, who participated in face‐to‐face interviews. Fifty‐eight per cent of respondents reported sleep problems on one or more nights the previous week and 18% reported that the sleep they obtained was insufficient on the majority of nights. Sleep durations were longest in the youngest participants (16–24 years), who slept on average 1 h longer than the 7.04 (SD 1.55) sample average. Sleep duration showed no appreciable change beyond middle age. Men and women reported sleeping similar amounts but women reported more sleep problems. Men reported sleeping less when there were more children in their household. Workers (i.e. employees) reported sleeping less on workdays than on non‐workdays, but those based at home and those not employed did not. Inability to switch off from work was related to sleep duration on non‐workdays. Across all participants average sleep duration exhibited a non‐monotonic association with quality of life (i.e. contribution of sleep to energy, satisfaction and success in work, home and leisure activities). Quality of life was positively associated with sleep duration, for durations up to 9 h, but negatively associated with quality of life beyond this. Comparison of our data with the US national sleep poll revealed that Britain sleeps as little or less, whereas a comparison with data reported 40 years ago revealed no statistically reliable reductions. Although we may not sleep less than four decades ago, when we report sleeping less we also tend to associate that lack of sleep with poor performance and quality of life.

Risk perception and decision taking during the transition between novice and experienced driver status

Abstract Young drivers are statistically overrepresented in road accidents. Their elevated risk is a complex function of chronological age and driving experience, both of which are associated with acceptance and misperception of risk on the road. A better understanding of the cognitive changes which accompany their transition from novice to experienced driver status should allow us to elucidate the factors underlying their acceptance and misperception of traffic risk. With, such an understanding, we would be in a better position to take decisions about improving their safety via training and education schemes rather than via legislative constraints on their exposure to risk. This paper considers evidence that young drivers underestimate certain traffic hazards and overestimate their own driving abilities. The potential contribution of these misperceptions to their faulty decision taking during skills acquisition is discussed in the light of recent findings from relevant research. Effects of partial and acute total sleep deprivation on performance across cognitive domains, individuals and circadian phase.

Background Cognitive performance deteriorates during extended wakefulness and circadian phase misalignment, and some individuals are more affected than others. Whether performance is affected similarly across cognitive domains, or whether cognitive processes involving Executive Functions are more sensitive to sleep and circadian misalignment than Alertness and Sustained Attention, is a matter of debate. Methodology/Principal Findings We conducted a 2 × 12-day laboratory protocol to characterize the interaction of repeated partial and acute total sleep deprivation and circadian phase on performance across seven cognitive domains in 36 individuals (18 males; mean ± SD of age = 27.6±4.0 years). The sample was stratified for the rs57875989 polymorphism in PER3, which confers cognitive susceptibility to total sleep deprivation. We observed a deterioration of performance during both repeated partial and acute total sleep deprivation. Furthermore, prior partial sleep deprivation led to poorer cognitive performance in a subsequent total sleep deprivation period, but its effect was modulated by circadian phase such that it was virtually absent in the evening wake maintenance zone, and most prominent during early morning hours. A significant effect of PER3 genotype was observed for Subjective Alertness during partial sleep deprivation and on n-back tasks with a high executive load when assessed in the morning hours during total sleep deprivation after partial sleep loss. Overall, however, Subjective Alertness and Sustained Attention were more affected by both partial and total sleep deprivation than other cognitive domains and tasks including n-back tasks of Working Memory, even when implemented with a high executive load. Conclusions/Significance Sleep loss has a primary effect on Sleepiness and Sustained Attention with much smaller effects on challenging Working Memory tasks. These findings have implications for understanding how sleep debt and circadian rhythmicity interact to determine waking performance across cognitive domains and individuals.

Sleep, diurnal preference, health, and psychological well-being: a prospective single-allelic-variation study.

Individual differences in sleep and diurnal preference associate with physical and mental health characteristics, but few genetic determinants of these differences have been identified. A variable number tandem repeat (VNTR) polymorphism in the PERIOD3 (PER3) gene (rs57875989) has been reported to associate with diurnal preference, i.e., preferred timing of waking and sleep. Here, the authors investigate in a prospective single-candidate genetic variant study whether allelic variation for this polymorphism associates also with reported actual sleep timing and sleep duration, as well as psychological and health measures. Six hundred and seventy-five subjects, aged 20 to 35 yrs, completed questionnaires to assess sleep and psychological and health characteristics and were genotyped for the PER3 VNTR. Homozygosity for the longer allele (PER35/5) of the VNTR was associated with increased morning preference, earlier wake time and bedtime, and reduced daytime sleepiness. Separate analyses of work and rest days demonstrated that the increase in time in bed during rest days was greatest in PER35/5 homozygotes. PER3 genotype modified the effects of sleep timing and duration on fluid intelligence and body mass index. Genotype was not associated with physical or psychological characteristics as assessed by the SF-36 Health Questionnaire, the General Health Questionnaire, the Big Five Inventory, the Behavioral Inhibition System–Behavioral Activation System scales, and the Positive and Negative Affect Scale, even though these measures varied significantly with diurnal preference as assessed by the Morningness-Eveningness Questionnaire. Whereas diurnal preference also predicts mental health and psychological characteristics, as well as sleep timing, the PER3 VNTR specifically affects measures of sleep timing and may also modify the effects of sleep on health outcome measures. Youthfulness, inexperience, and sleep loss: the problems young drivers face and those they pose for us

Young inexperienced drivers are more likely to be involved in road traffic crashes than drivers who are older and more experienced. This paper argues that neither age nor inexperience are, in and of themselves, sufficient explanations of the association between age, experience, and casualty rates. The aim here is to consider what it is about inexperienced young drivers in particular that may increase crash risk. Evidence is reviewed showing differential sleep loss among different teenage groups, which may relate to recently presented evidence that young teenagers are more crash involved than drivers in their early twenties. Potential acute and chronic effects of sleep loss among teenagers and young adults are described.

Differential effects of a dual orexin receptor antagonist (SB-649868) and zolpidem on sleep initiation and consolidation, SWS, REM sleep, and EEG power spectra in a model of situational insomnia.

Orexins have a role in sleep regulation, and orexin receptor antagonists are under development for the treatment of insomnia. We conducted a randomised, double-blind, placebo-controlled, four-period crossover study to investigate the effect of single doses of the dual orexin receptor antagonist SB-649868 (10 or 30 mg) and a positive control zolpidem (10 mg), an allosteric modulator of GABAA receptors. Objective and subjective sleep parameters and next-day performance were assessed in 51 healthy male volunteers in a traffic noise model of situational insomnia. Compared with placebo, SB-649868 10 and 30 mg increased total sleep time (TST) by 17 and 31 min (p<0.001), whereas after zolpidem TST was increased by 11.0 min (p=0.012). Wake after sleep onset was reduced significantly by 14.7 min for the SB–6489698 30 mg dose (p<0.001). Latency to persistent sleep was significantly reduced after both doses of SB–6489698 (p=0.003), but not after zolpidem. Slow wave sleep (SWS) and electroencephalogram (EEG) power spectra in non-REM sleep were not affected by either dose of SB-640868, whereas SWS (p< 0.001) and low delta activity (<=1.0 Hz) were increased, and 2.25–11.0 Hz activity decreased after zolpidem. REM sleep duration was increased after SB-649868 30 mg (p=0.002) and reduced after zolpidem (p=0.049). Latency to REM sleep was reduced by 20.1 (p=0.034) and 34.0 min (p<0.001) after 10 and 30 mg of SB-649868. Sleep-onset REM episodes were observed. SB-649868 was well tolerated. This dual orexin receptor antagonist exerts hypnotic activity, with effects on sleep structure and the EEG that are different from those of zolpidem.

Research on drivers and driving covers a wide range of topics and approaches within psychology. The driving task, as complex everyday task, provides a focus for a wide range of theories and methodologies and allows these to be tested and elaborated in a highly relevant applied setting. This article gives a very brief overview of the relevant topics such as driver perception and cognition and the social psychology of driving. Traffic psychology also significant contributions to make to the development of accident countermeasures. This does not only include traditional approaches such as road user education and training, but it is also argued that application of psychological knowledge about driver perception and cognition can contribute to optimal road and vehicle design.

Anger-congruent behaviour transfers across driving situations

Anger and aggression on the road may sometimes appear unprovoked and unrelated to current driving circumstances. It is unclear whether such anger and aggression arises because of events prior to those circumstances in which anger is experienced and aggression is exhibited. In this study, time pressure and enforced following of a slowly moving vehicle were used to increase drivers’ anger in order to assess whether affect and behaviour during a subsequent, non-provocative, drive would change accordingly. Ninety-six drivers drove twice in a simulated urban environment. During the first drive, oncoming traffic and a slowly moving lead vehicle required that half of the drivers travelled far slower than they would choose. During the second drive, drivers again followed slower vehicles and were required to respond to traffic events not encountered in the manipulation drive. Mood (Profile of Mood States) was assessed before and after each drive, and anger evaluations, arousal (heart rate) and behaviour (speed, lane position and collisions) were measured during drives. Anger increased and both mood and driving behaviour deteriorated in drivers exposed to slower lead vehicles, compared with control group drivers. These behavioural differences of speed and lane positioning carried over into the subsequent drive even to driving situations unlike those where provocation had previously occurred. Drivers who had previously been impeded later approached hazards with less caution, and attempted more dangerous overtaking manoeuvres. It is concluded that sometimes dangerous driving may result from anger provoked by circumstances other than those in which the behaviour is exhibited.

Anticipating the content and circumstances of skill transfer: Unrealistic expectations of driver training and graduated licensing?

There is substantial evidence that driving skills improve during driver training, but the long-term safety benefit of such formal training remains unproven. Restricting the exposure of newly licensed drivers to more hazardous driving circumstances, as in graduated driver licensing (GDL) regimes, demonstrably reduces crash risk, but drivers remain at risk after the restrictions are eased. GDL and most other licensing regimes advocate increased basic training and practice, but thereafter require neither advanced training nor systematic increase in exposure to risk. This assumes that basic skills acquired during formal training will transfer positively to new and more demanding traffic circumstances. This paper reviews the theoretical basis for these assumptions and offers a way of systematically identifying the extent of transfer desired. It is concluded that there is little theoretical or empirical foundation for the supposition that what is learned during or after training will have a safety benefit in later driving.
