A polyhydric phenol sulfokinase and chronic liver injury

SummaryThe sulfokinase, that transfers activated sulfate from PAPS (3′-phosphoadenosine-5′phosphosulfate) to 4nitrocatechol sulfate was studied.1.It is thought that the enzyme plays an important role in detoxication by the sulfation of polyhydric phenols in liver.2.The enzyme had an optimum at pH 8.0 in Tris-acetate buffer. Km was 0.105 x 10-3M. The rate of conjugation was linear within 1 min.3.The main distribution of the sulfokinase was found in the liver, lungs, spleen and various other organs in the rat. The activity of the enzyme in rat liver was the highest in soluble fraction. In chronic hepatic injury caused by carbon tetrachloride injection, the activity of the enzyme gradually decreased to below about onehalf of the control value.

Carrier state and infectivity of HBs-Ag

It has recently been clarified by Magnius and others that e antigen and antibody system is closely related to HBV infectivity, i.e., HBsAg-positive sera detected e antigen (e-Ag) have an intensive infectivity, and those detected e antibody are no infective. Therefore, the authors tested e-Ag and e-Ab by the Micro-Ochteronys method in sera from 127 carriers, and, from the data, discussed as to intrafamilial infections with HBV and the clustering of the carriers. The results are as follows. 1. In eleven children of 6 e-Ag-positive carrier mothers, 10 children were positive for HBsAg, and 3 out of 6 children of 2 e-Ag-positive carrier fathers also were HBsAg-positive. Moreover, in one of 3 children living with their e-Ag-positive grandfather, HBsAg was detected in the serum. Accordingly, it can be said that an e-Ag-positive carrier produces new carriers at a high rate in the following generations, and the rate is extremely high in mother-to-child transmission. 2. In 103 asymptomatic carriers, the frequency of e-Ag-positive result was higher in the younger ages (75% in ages of below 10 years and 12% in ages of more than 41 years). On the contrary, the rate of e-Ab-positive carriers was higher in the older ages (17% in ages of below 10 years and 44% in ages of more than 41 years). This result implies that, in the majority of HBsAg carriers, although there are wide differences in the individuals, HBV infectivity declines with years and finally vanishes.

A Case of intrahepatic aneurysm diagnosed by selective celiacography

In t rahepat ic aneurysm is an uncommon lesion which is very ra re ly diagnosed preoperatively and only a smal l number of cases have been succesfully treated. A 34-years-old male admitted in our hospital because of bleeding gas t r ic ulcer, and gas t ro je junos tomy was performed on 15. March, 1966. The bleeding was pointed out rup tured a r te ry on the bot tom of the ulcer by histologic examinat ion. After this operat ion serum hepati t is was developed and SGT elevated to 260 u., SGPT was 420 unit . For the purpose of making accurate diagnosis needle biopsy of liver was done under control ing by laparoscopy, and no serious bleeding was noted on liver surface. However following this procedure severe r igh t upper quodrant pain occured and vigorous t a r ry stool wi th hematoemesis came out continiously dur ing several weeks. Then selective cel iacography was performed by Seldinger s method and the X-ray film revealed two round aneurysm in the area of lobus quadora tus of the liver, which had small flngr top size and belong to the 3rd b ranch of hepatic ar tery. On the next day, par t ia l hepatectomy was performed and no more aneurysm was found by direct hepatic a r te r iography dur ing surgery. Now, pa t ien t has no hematoemesis or t a r ry stool af ter hepateetomy and has been very well. Numerous factor have been implicated in the etiology of this disease, bu t in our case needle biopsy tha t has been performed on severe stageof serum hepat i t i s was chief role of making this aneurysm.
