A genome-wide scan for QTL affecting economically important traits in beef production was performed using an F(2) resource family from a Japanese Black x Limousin cross, where 186 F(2) animals were measured for growth, carcass, and meat-quality traits. All family members were genotyped for 313 informative microsatellite markers that spanned 2,382 cM of bovine autosomes. The centromeric region of BTA2 contained significant QTL (i.e., exceeding the genome-wide 5% threshold) for 5 carcass grading traits [LM area, beef marbling standards (BMS) number, luster, quality grade, and firmness), 8 computer image analysis (CIA) traits [LM lean area, ratio of fat area (RFA) to LM area, LM area, RFA to musculus (M.) trapezius area, M. trapezius lean area, M. semispinalis lean area, RFA to M. semispinalis area, and RFA to M. semispinalis capitis area], and 5 meat quality traits (contents of CP, crude fat, moisture, C16:1, and C18:2 of LM). A significant QTL for withers height was detected at 80.3 cM on BTA5. We detected significant QTL for the C14:0 content in backfat and C14:0 and C14:1 content in intermuscular fat around the 62.3 to 71.0 cM region on BTA19 and for C14:0, C14:1, C18:1, and C16:0 content and ratio of total unsaturated fatty acid content to total SFA content in intramuscular fat at 2 different regions on BTA19 (41.1 cM for C14:1 and 62.3 cM for the other 4 traits). Overall, we identified 9 significant QTL regions controlling 27 traits with genome-wide significance of 5%; of these, 22 traits exceeded the 1% genome-wide threshold. Some of the QTL affecting meat quality traits detected in this study might be the same QTL as previously reported. The QTL we identified need to be validated in commercial Japanese Black cattle populations.

We have adapted the enzymatic method [Biochemical and Biophysical Research Communications 176 (3) (1991) 1617] for the safe and rapid assay of L-carnitine (L-CA) in skeletal muscle using a microplate reader. The concentration of L-CA in fresh semitendinosus muscle from broiler chicken, pig, beef cattle, deer, horse and goat muscle were 0.69, 1.09, 1.86-3.57, 4.57, 4.95 and 11.36 μmol/g wet weight, respectively. The animals which had higher concentration of L-CA, also had the highest amounts of myoglobin as an index to the redness of the muscle. Furthermore, we investigated this relationship between white muscle, M. pectoralis profundus, and red muscle, M. soleus, in laying hens. The L-CA and myoglobin concentration in red muscle were significantly higher than those in white muscle (p<0.01). These findings suggest that L-CA concentration in muscle is related to oxygen metabolism and to myofiber types.

The percentage of MUFA to total fatty acids of beef differs among intermuscular, intramuscular, and subcutaneous fat even within an individual cow. Our objective was to investigate the variation of the percentage of MUFA by geometric and sectional change of marbling flecks in rib eye. Longissimus muscles of 8 Japanese Black steers from a common sire and a common maternal grand sire were used. Three slices (1 from rib roast and 2 from sirloin) from each animal were selected for analysis. Five marbling flecks from each slice were randomly taken to obtain the percentage of MUFA using gas chromatography. High-quality digital images of all slices were taken with a mirror-type camera. The area and location of each marbling fleck were calculated by image analysis. The marbling flecks were categorized by area [small <0.4 cm(2), medium 0.4 to 2.0 cm(2), large >2.0 cm(2)], by location (dorsal and ventral), and by slice section through the LM (front, middle, and back). The effects of classification according to the area, location, and slice section were statistically significant (P < 0.05) for the percentage of MUFA. Least squares means of the percentage of MUFA for marbling flecks of sizes small, medium, and large were 56.8, 58.4, and 60.2%, respectively, indicating that larger marbling flecks had greater MUFA (P < 0.05). Those of dorsal, ventral, front, middle, and back were 59.1, 57.8, 55.4, 59.9, and 60.1%, respectively. The percentages of MUFA of the marbling flecks located in the dorsal part were greater than those in the ventral part (P < 0.05). The percentages of MUFA from middle and back were greater than those from front (P < 0.01). We suggest that the area, location, and slice section of marbling would be the determining factors for the percentage of MUFA of marbling.

The aim of the present study was to evaluate the application of image analysis for Japanese Black cattle in Australia (Australian JB). Therefore, we assessed meat quality using an image analysis method to estimate the heritability of this trait in Australian JB. We photographed the cross-section of the 5th–6th ribs and calculated image analysis traits of 473 and 539 head of Australian JB and Australian JB sire crosses with other breeds (F1), respectively. Least square means of grading and image analysis traits were calculated. We further estimated the heritability of grading and image analysis traits of 414 head of Australian JB. The Australian Meat Industry Classification System (AUS-MEAT) marbling score (6.8) and percentage marbling area (29.2%) for Australian JB were significantly (P < 0.01) higher than those for F1 (4.7% and 19.3%, respectively). Percentage marbling area strongly correlated with the AUS-MEAT marbling score (r = 0.88), indicating that marbling can be improved using percentage marbling area as a substitute for AUS-MEAT marbling score. The head counts of AUS-MEAT marbling score increased in the Australian JB (mode value = 9). The result indicated that the AUS-MEAT marbling score lacks a sufficient range of values to evaluate a high marbling beef breed such as the Australian JB. Further, the heritability of percentage marbling area was 0.54, which is higher than the heritability of AUS-MEAT marbling score (0.23). Therefore, we conclude that determining percentage marbling area using image analysis may prove to be an effective method for improving the marbling of the Australian JB.

The objective of this study was to estimate genetic parameters for various fertility traits on Holstein upgraded dairy heifers and cows in a smallholder system under tropical conditions using data sets from the Thailand national recording scheme. The investigated traits were age at first service (AFS), age at first calving (AFC), days from calving to first service (DTFS), days between first and last service (DFLS), days open (DO), calving interval (CI), number of services per conception (NSPC), and conception at first service (FSC). The data consisted of 68,555, 34,401, and 54,004 records on heifers, primiparous, and multiparous cows, respectively, calving between 1996 and 2011. Gibbs sampling was employed to obtain (co)variance components using both univariate and bivariate analyses with linear and threshold animal models. Virgin heifers had better fertility performance than primiparous and multiparous cows. The reproductive performance in primiparous cows was inferior compared with multiparous cows. Cows with higher Holstein-Friesian blood showed lower reproductive efficiency. Estimated heritabilities for most of the fertility traits were 0.04 or less except for AFS (0.26) and AFC (0.25). The estimated genetic correlations among fertility traits within parity indicated that selection for cows with high conception rate could lead to shortened DO and CI, as well as DTFS. The FSC and NSPC could be used as the best indicators for heifer and cow fertility and could be complemented by other traits, which were genetically considered as different traits such as DTFS and DFLS in terms of a fertility index. This would enable efficient selection for better fertility. Genetic correlations for fertility traits in primiparous and multiparous cows were very high (>0.90), but those between heifers and cows were lower (0.03 to 0.83). The latter results indicated that fertility traits of heifers and cows should be considered as different traits.

This study was designed to: (i) estimate genetic parameters and breeding values for conception rates (CR) using the repeatability threshold model (RP-THM) and random regression threshold models (RR-THM); and (ii) compare covariance functions for modeling the additive genetic (AG) and permanent environmental (PE) effects in the RR-THM. The CR was defined as the outcome of an insemination. A data set of 130 592 first-lactation insemination records of 55 789 Thai dairy cows, calving between 1996 and 2011, was used in the analyses. All models included fixed effects of year × month of insemination, breed × day in milk to insemination class and age at calving. The random effects consisted of herd × year interaction, service sire, PE, AG and residual. Variance components were estimated using a Bayesian method via Gibbs sampling. Heritability estimates of CR ranged from 0.032 to 0.067, 0.037 to 0.165 and 0.045 to 0.218 for RR-THM with the second, third and fourth-order of Legendre polynomials, respectively. The heritability estimated from RP-THM was 0.056. Model comparisons based on goodness of fit, predictive abilities, predicted service results of animal, and pattern of genetic parameter estimates, indicated that the model which fit the desired outcome of insemination was the RR-THM with two regression coefficients.

The test-day milk fat-to-protein ratio (TD-FPR) could serve as a measure of energy balance status and might be used as a criterion to improve metabolic stability and fertility through genetic selection. Therefore, genetic parameters for fertility traits, test-day milk yield (TD-MY) and TD-FPR, as well as, their relationships during different stages of lactation, were estimated on data collected from 25 968 primiparous Thai dairy crossbred cows. Gibbs sampling algorithms were implemented to obtain (co)variance components using both univariate linear and threshold animal models and bivariate linear-linear and linear-threshold animal models with random regression. Average TD-MY and TD-FPR were 12.60 and 1.15. Heritability estimates for TD-MY, TD-FPR and selected fertility traits ranged from 0.31 to 0.58, 0.17 to 0.19 and 0.02 to 0.05, respectively. Genetic correlations among TD-FPR and TD-MY, TD-FPR and fertility traits, and TD-MY and fertility traits ranged from 0.05 to -0.44, from -0.98 to 0.98 and -0.22 to 0.79, respectively. Selection for lower TD-FPR would decrease numbers of inseminations per conception and increase conception at first service and pregnancy within 90 days. In addition, cow selection based only on high milk production has strong effects to prolong days to first service, days open and calving interval.

In Japan, Wagyu cattle include four Japanese breeds; Black, Brown, Shorthorn, and Polled. Today, the renowned brand name Wagyu includes not only cattle produced in Japan, but also cattle produced in countries such as Australia and the United States. In recent years, the intramuscular fat percentage in beef (longissimus muscle) from Japanese Black cattle has increased to be greater than 30%. The Japanese Black breed is genetically predisposed to producing carcass lipids containing higher concentrations of monounsaturated fatty acids than other breeds. However, there are numerous problems with the management of this breed including high production costs, disposal of untreated excrement, the requirement for imported feed, and food security risks resulting from various viral diseases introduced by imported feed. The feeding system needs to shift to one that is more efficient, and improves management for farmers, food security for consumers, and the health environment for residents of Japan. Currently, we are developing a metabolic programming and an information and communications technology (ICT, or Interne of Things) management system for Wagyu beef production as future systems. If successful, we will produce safe, high-quality Wagyu beef using domestic pasture resources while solving the problems of how to utilize increasing areas of abandoned agricultural land and to make use of the plant-based feed resources in Japan’s mountainous areas.

As marbling is a principal input into many grading systems it is important to have an accurate and reliable measurement procedure. This paper compares three approaches to measuring marbling: trained personnel, near infrared spectroscopy (NIR) and image analysis. One 25mm slice of meat was utilised from up to 12 cuts from 48 carcasses processed in Poland and France. Each slice was frozen to enable a consistent post-slaughter period then thawed for image analysis. The images were appraised by experienced beef graders and the sample used to determine fat content by NIR. We find that image analysis based marbling measures are capturing something different to trained personnel and that there is a strong relationship between near infrared spectroscopy and trained personnel. Finally, we demonstrate that marbling measures taken on one muscle can be predictive of marbling in other muscles in the same carcase. This is particularly important for cut based models such as the Meat Standards Australia system.

40頭の黒毛和種去勢牛に対する3方向からの画像(後,横および上方向)が,ビデオカメラにより記録され,コンピュータのメモリー上に取り入れられた.生体の輪郭線の抽出および生体の部分体積および部分密度の推定は,著者らにより作成されたコンピュータプログラムで行われた.生体に関する12形質,15の枝肉形質および超音波による皮下脂肪厚も測定された.それらの体測定値と画像解析形質との関連性が検討された,生体部分体積と生体部分密度の平均値はそれぞれ0.35m3,1.608g/cm3であった.生体部分体積は,体のサイズに関連する形質に対して有意かつ正に影響していた.生体部分密度と超音波による皮下脂肪厚との間に有意かつ負の相関が認められた.枝肉形質を推定するための重回帰分析に,生体部分体積および密度を取り入れることで,歩留基準値のR2値に有意な改善が認められた.
