The aim of this study was to determine the allele and genotype frequencies of the Insulin like growth factor 1 gene (IGF-1) and Insulin like growth factor 1 receptor gene (IGF-1R) polymorphisms in East Anatolian (EAR) and South Anatolian Red cattle (SAR). Polymorphisms in both genes are claimed to affect economic parameters like body weight and subcutaneous back fat. For the study, blood samples were collected from 50 SAR and 50 EAR cattle. In both breeds, high frequencies of allele B of IGF-1, which was supposed to have positive effect on carcass traits and allele A of IGF-1R, related to milk traits, were observed. Therefore, no relationship of the polymorphisms studied with economic traits was observed, as both breeds have respectively low carcass and milk parameters. However, distribution of allele frequencies of IGF-1 and IGF-1R in SAR and EAR cattle was similar with Zebu cattle that support Bos indicus introgression to Anatolian breeds.

Abstract The objective of the present study was to determine the genotype and allele frequencies for osteopontin (OPN), prolactin (PRL) and pituitary-specific transcription factor-1 (PIT-1) that are claimed to be associated with milk production traits in cattle in South Anatolian Red (SAR) and East Anatolian Red (EAR) cattle. In this study, 40 cattle for each of SAR and EAR were used. OPN in intron 4, PRL in exon 3 and PIT-1 in exon 6 were amplified by polymerase chain reaction (PCR) and the resulting PCR products were subjected to digestion using BseNI, RsaI and HinfI enzymes, respectively, to determine the single nucleotide polymorphisms (SNPs). Results showed that frequencies of the OPN allele C, PRL allele B, and PIT-1 allele A which are associated with milk production tarits were found low in SAR and EAR. As a result, SAR and EAR cattle breeds have the lower allel frequencies that influence the milk production traits for three genes than high-yielding European dairy cattle breeds.

Anatolia has been an important region for civilizations and agricultural revolution as a major domestication centre for livestock species. Goats (Capra hircus) were among the earliest domesticated animals in this region. In this study, genetic diversity of Anatolian goat breeds was characterized by comparison of mitochondrial DNA hypervariable region 1. A total of 295 individuals, including 99 Anatolian Black goats, 96 Angora goats and 100 Kilis goats, were used. Haplogroup A was found to be the dominant haplogroup in all three breeds. The highest haplogroup diversity, including haplogroups A, B2, C and G, was observed in the Anatolian Black breed. Haplogroup D was only observed in Kilis and Angora goats. Haplogroup G was found in Angora and Anatolian Black breeds. The Anatolian goat breeds had high genetic diversity values and a weak phylogeographical structure. The nucleotide diversity values were found to be higher than those in previously studied goat breeds. The fact that Anatolia is a domestication centre and its geographical position as a junction of trade routes may have caused the higher genetic diversity of Anatolian goat breeds.

Abstract This study was carried out to determine polymorphisms of four genes in South Anatolian Red (SAR) and East Anatolian Red (EAR) indigenous cattle breeds in Turkey. Single nucleotide polymorphisms (SNPs) monitored in this study are Y581S in ATP binding cassette sub family G member 2 (ABCG2) gene, c.1892T>C and c.3359A>C in peroxisome proliferator-activated receptor gamma coactivator 1 alpha (PPARGC1A) gene and g.8232C>A in oxidized low-density lipoprotein receptor 1 (OLR1) gene. The frequency of the ancestral allele A of the ABCG2 gene Y581S polymorphism was found to be very high (SAR: 0.63; EAR: 0.64) in both cattle breeds. The CC genotypes of PPARGC1A gene c.1892T>C (SAR: 0.65; EAR: 0.80) and OLR1 gene g.8232C>A polymorphisms (SAR: 0.82; EAR: 0.86), which are associated with high milk fat percentage, had higher frequencies than those of the other genotypes. In conclusion, we might suggest that the allele distribution of the ABCG2 gene Y581S polymorphism can be the evidence indicating autosomal gene flow from zebu cattle to SAR and EAR cattle breeds.

Kivircik sheep is an important local Turkish sheep according to its meat quality and milk productivity. The aim of this study was to analyze gene expression profiles of both prenatal and postnatal stages for the Kivircik sheep. Therefore, two different cDNA libraries, which were taken from the same Kivircik sheep mammary gland tissue at prenatal and postnatal stages, were constructed. Total 3072 colonies which were randomly selected from the two libraries were sequenced for developing a sheep ESTs collection. We used Phred/Phrap computer programs for analysis of the raw EST and readable EST sequences were assembled with the CAP3 software. Putative functions of all unique sequences and statistical analysis were determined by Geneious software. Total 422 ESTs have over 80% similarity to known sequences of other organisms in NCBI classified by Panther database for the Gene Ontology (GO) category. By comparing gene expression profiles, we observed some putative genes that may be relative to reproductive performance or play important roles in milk synthesis and secretion. A total of 2414 ESTs have been deposited to the NCBI GenBank database (GW996847–GW999260). EST data in this study have provided a new source of information to functional genome studies of sheep.
