Comparison of the effects of various β-adrenergic blocking agents with known antiarrhythmic drugs on aconitine-arrhythmia produced by the cup method

Abstract A controllable atrial arrhythmia continued as long as aconitine solution, applied by the cup method, was present in a restricted area of atrial surface of the exposed dog heart. Comparative studies of antiarrhythmic activity of β-adrenergic blocking agents with known antiarrhythmic compounds were carried out by intravenous infusion for a period from 10 to 30 min. Antiarrhythmic activities of these compounds were in the following order; pro propranolol > MJ 1999 = ajmaline > LB 46 > quinidine = diphenylhydantoin, the potencies of which roughly corresponded to 10:1: 1 2 : 1 5 . Alprenolol, ICI 50172, DCI and procainamide blocked atrial arrhythmia only in a very limited number of experiments, because prominent hypotension or marked depression in atrioventricular conduction occurred before the heterotopic pacemaker was suppressed. No relationship was found between β-adrenergic blocking and antiarrhythmic activities.

Der Effekt von sechsβ-adrenergischen Blockierungsmitteln auf Akonitin-Arrhythmia bzw. Ouabain-Arrhythmia wurde mittels i.v. Verabreichung vergleichend untersucht. Reihenfolge der Wirksamkeit auf Akonitin-Arrhythmia: Propranolol>MJ 1999>LB 46>H 56/28, ICI 50172 und DCI, und dieselbe auf Ouabain-Arrhythmia: Propranolol, LB 46 und H 56/28 gleichwertig, MJ 1999 und ICI 50172 wirkungslos.
