Inheritance of (1–3)(1–4)-beta-D-glucan content in barley ( Hordeum vulgare L.)

Abstractβ-glucan is the soluble dietary fiber component and occurs at its highest in barley. This study aims to evaluate the inheritance of β-glucan content in barley grains and to map quantitative trait loci (QTL) associated with this trait. F5-derived 107 lines from the cross of the six-rowed waxy hulless barley, ‘Yonezawa Mochi’ and the six rowed non-waxy hulless barley,’ Neulssalbori’ were measured for their agronomic traits and β-glucan level at four different environments. These recombinant lines showed significant genotypic variation (P < 0.01) and normal distribution for β-glucan content with a range of 43.6–62.1 g kg−1 across environments. A significant genotype-by-environment interaction was also found. The broad-sense heritability estimates for β-glucan content ranged from 0.42 to 0.82 across environments. Using one-factor analysis and composite interval mapping, a main effect of QTL associated with β-glucan content was identified in the genomic region near waxy gene (wx) and HVM4 on chromosome 7H. The major QTL at this region explained on average 44.4% of the variation for the mean of β-glucan content across environments with LOD values that ranged from 5.7 in Suwon in 2001 to 13.9 in Suwon in 2003. Two minor QTLs were identified but their significance of association with β-glucan content was inconsistent across environments.

Mineral Compositions of Korean Wheat Cultivars

Twenty-nine Korean wheat cultivars were analyzed for 8 important minerals (Cu, Fe, Mn, Zn, Ca, K, Mg and P) using Inductively Coupled Plasma Atomic Emission Spectrometry (ICP-AES). A hierarchical cluster analysis (HCA) was applied to classify wheat cultivars, which has a similarity in mineral compositions. The concentration ranges of the micro-minerals Cu, Fe, Mn, and Zn: 0.12~0.71 mg/100 g, 2.89~5.89 mg/100 g, 1.65~4.48 mg/100 g, and 2.58~6.68 mg/100 g, respectively. The content ranges of the macro-minerals Ca, K, Mg and P: 31.3~46.3 mg/100 g, 288.2~383.3 mg/100 g, 113.6~168.6 mg/100 g, and 286.2~416.5 mg/100 g, respectively. The HCA grouped 6 clusters from all wheat samples and a significant variance was observed in the mineral composition of each group. Among the 6 clusters, the second group was high in Fe and Ca, whereas the fourth group had high Cu, Mn and K concentrations; the fifth cluster was high in Zn, Mg and P. The variation in mineral compositions in Korean wheat cultivars can be used in the wheat breeding program to develop a new wheat cultivar with high mineral content, thus to improve the nutritional profile of wheat grains.

Forage Productivity and Feed Value in Triple Cropping Systems with Winter Forage Crops-Silage Corn-Summer Oat Cultivation at Paddy Field in Southern Region of Korea

Multiple cropping system is an important agricultural system in Korea, which is significant to increase forage yield and promote agricultural economic development. This experiment was carried out to develop triple cropping systems (winter cereals crop for forage - silage corn - summer oats) for maximum year-around forage production at paddy field in southern region. We also to select a winter forage crop to determine corn planting time, and to examine the forage productivity and feed value of summer cultivated oat, which was planted after corn cultivation. When winter cereal crops for forages are harvested in accordance with and corn planter, the fresh yield and dry matter yield of rye were 32.0 ton/ha and 5.8 ton/ha, respectively, showing higher yielding compared to other winter crops. Corn silage as summer crop was showed the fresh yield and dry matter yield of 72.1 ton/ha and 20.2 ton/ha, respectively. In the feed value, crude protein and TDN contents were 7.4% and 69.3%, respectively. They was showed higher productivity and feed value. The heading date of High-speed oat was October 10, and its fresh yield and dry matter yield were 37.6 ton/ha and 8.0 ton/ha, respectively. As a feed value, crude protein and TDN content was 11.4% and 59.1%, respectively. Therefore, triple cropping systems for the production of forage all throughout the year are possible with the introduction of summer oats in the southern region, and rye could be the most suitable winter forage crops for triple cropping systems.

ISSR-derived Molecular Markers for Korean Wheat Cultivar Identification

Inter simple sequence repeat (ISSR) is a technique that leads to the development of novel specific molecular markers and relationship analysis between species. In this study, 54 ISSRs were used to develop cultivar-specific DNA sequence characterized amplified regions (SCARs) for Korean wheat cultivars (Triticum aestivum). Using nine of 54 UBC ISSR primers, 16 cultivar-specific fragments were detected among 32 wheat cultivars. Of the 16 primers, 10 successfully amplified targets in 32 cultivars and six novel cultivar-specific markers were finally selected. A total of 13 Korean wheat cultivars (Ol, Dahong, Namhae, Jinpum, Anbaek, Jopoom, Jokyung, Sinmichal 1, Jeokjoong, Dajung, Goso and Joah) were classified by newly developed six SCAR markers. Cultivar-specific markers can be used not only for cultivar fingerprinting but also for marker-assisted selection in wheat breeding program.

A New High Grain Yielding Forage Rye Cultivar, “Seedgreen”

“Seedgreen” (Secale cereal L.), a new rye cultivar was developed by National Institute of Crop Science (NICS), RDA in 2013. It was developed from an open pollination from within 10 rye varieties or lines including “Chochun” in 1995. The line “SR95POP-S1-140-9-1-3-7-5-3” was selected for its excellent agronomic appearance, and was placed in yield trials for three years from 2008 to 2010. The new cultivar was designated “Homil50” and was placed in regional yield trials at the five locations around Korea from 2011 to 2013, during which time the name “Seedgreen” was given. This cultivar is an erect plant type and of a long size, with a dark-green leaf color, a yellowish-white colored, medium-diameter culm, and a brown-colored, medium-size grain. The heading and maturation dates of Seedgreen were April 22 and June 16, which were 3 days and 2 days earlier than that of “Gogu”, respectively. Seedgreen also showed better winter hardiness and a greater resistance to lodging and wet injury compared to those of the check cultivar. Over three years, the average dry matter yield of Seedgreen was 8.3 ton ha (fresh yield = 39.8 ton ha), which was harvested in late April and was lower than that of the check cultivar Gogu. The seed productivity of Seedgreen was approximately 4 ton ha, which was 16 % more than that of the check. Seedgreen was higher to than Gogu in term of protein content (10.5% and 9.7%, respectively), total digestible nutrients (TDN) (58.3% and 57%, respectively), and TDN yield ha (4.81 ton and 4.77 ton, respectively). This cultivar is recommended as a fall sowing crop in areas where the average daily minimum-mean temperatures are higher than -12°C in January, and as a winter crop for whole-crop forage before the planting of rice or green manure around Korea.
