Canine faecal contamination and parasitic risk in the city of Naples (southern Italy)

BackgroundDogs are associated with more than 60 zoonotic diseases among which, parasitosis and, in particular, helminthosis, can pose serious public-health concerns worldwide. Many canine gastrointestinal parasites eliminate their dispersion elements (eggs, larvae, oocysts) by the faecal route. The quantity of canine faeces deposited on public and private property in cities worldwide is both a perennial nuisance and an important health issue. Public sites such as playgrounds, parks, gardens, public squares and sandpits may be an important source of human infection. The aim of this study was to evaluate the extent of both canine faecal contamination in the city of Naples (southern Italy), and presence of canine parasitic elements, with particular regard to those which are potential agents of zoonosis. A regular grid of sub-areas (1 km × 700 m) was overlaid on the city map using a Geographical Information System (GIS). In each sub-area the straightest 1 km transect was drawn and digitalized on-screen in the GIS. Between February and May 2005 canine faeces were counted along the 1 km transects in 143 sub-areas, and 415 canine faecal samples were collected and submitted to coprological examinations. Negative binomial regression models and Gaussian random effects models were used to analyze the association between faeces count and human population density taking into account for extraPoisson variability. Logistic regression model was used to evaluate the association between positivity to parasitic elements and number of canine faeces.ResultsOut of the 143 studied sub-areas, 141 (98.6%) contained canine faeces. There was a strong spatial gradient with 48% of the total variability accounted by between neighbourhood variability; a positive association between the number of faeces and the human population density was found. Seventy (over 415, 16.9%) canine faecal samples were positive for parasitic elements. There was no association between positivity to parasitic elements and the number of canine faeces. Eggs of Toxocara canis, Toxascaris leonina, Ancylostoma caninum and Trichuris vulpis were found, as well as oocysts of Isospora canis.ConclusionIn conclusion, the results of the present study, conducted using GIS both for planning and sampling and for evaluation and presentation of findings, showed the presence of canine faecal contamination in the city of Naples, and the presence of canine parasitic elements, some of which are potential agents of zoonosis.

Use of the FLOTAC technique for the diagnosis of Aelurostrongylus abstrusus in the cat

Aelurostrongylus abstrusus is a lungworm of the domestic cats distributed worldwide. Its presence, however, is often underestimated due to limitation of common coprological diagnostic techniques. The present study reports, for the first time, the use of the FLOTAC technique for A. abstrusus diagnosis in feline faecal samples with results compared to the following three standard copromicroscopic techniques: the Baermann technique, the McMaster technique and the Wisconsin technique. The comparison study showed that the FLOTAC technique produced mean larvae per gram of faeces significantly (P < 0.05) greater than that produced by the other more widely used diagnostic tools. Thus, the FLOTAC technique can be utilised for quantifying lungworm larva burdens in feline faecal samples because of its high sensitivity, as already shown for various parasites of veterinary and human importance.

Resistance of trichostrongyles to benzimidazoles in Italy: a first report in a goat farm with multiple and repeated introductions

Anthelmintic resistance is widely distributed in small ruminants throughout the world. The extension of resistance seems lower in southern European countries and has not been reported previously in Italy. In the present study, resistance to benzimidazoles, levamisole and ivermectin was evaluated in a multi-breed goat farm of southern Italy. The farm had a history of repeated goat introductions from other flocks and a moderate regimen of anthelmintic treatments using alternatively the three above-mentioned drugs. Resistance of gastrointestinal strongyles was studied on the basis of faecal egg counts, egg hatch assay and necropsies. Resistance to anthelmintics was evidenced for benzimidazoles only, and Trichostrongylus colubriformis was the only resistant strongyle species. Single drug and single species resistance suggest that resistance is on its beginning and that measures for reducing the spread of resistance are of interest and should be promoted.

Efficacy and pharmacokinetics of OZ78 and MT04 against a natural infection with Fasciola hepatica in sheep

Fasciolosis is a parasitosis caused by the food-borne trematode Fasciola spp. of major veterinary significance. Triclabendazole is the first line treatment in humans and animals but cases of resistance are spreading worldwide. The synthetic peroxides OZ78 and MT04 are lead compounds for the treatment of fasciolosis. In the present study we investigated the efficacy and drug disposition following a single intramuscular dose of 100 mg/kg OZ78 and MT04 in sheep harbouring a natural Fasciola hepatica infection. A liquid chromatography and tandem mass spectrometry (LC-MS/MS) method was developed and validated to quantify plasma and bile concentrations of both compounds. Plasma samples were analysed with an accuracy for OZ78 and MT04 from 91 to 115% and a precision lower than 8.9%. Bile samples displayed an accuracy between 92 and 101% and a precision up to 12.7%. Bile samples were collected at 0 and 6h post-administration. Plasma mean peak concentration was 11.1 μg/ml at 1.5 h for OZ78 and 4.8 μg/ml at 4.2 h for MT04. Mean AUC of OZ78 and MT04 was 6698 and 3567 μg min/ml, respectively. Bile concentration at 6h post-treatment was 1.0 μg/ml for OZ78 and 1.4 μg/ml for MT04. Treatment with OZ78 showed no effect on egg burden and adult worm counts in vivo, whereas MT04 displayed a significant egg count reduction of 98.5% and a worm burden reduction of 92%. In conclusion, our study reveals an excellent activity of MT04 against F. hepatica in naturally infected sheep and a first insight into its PK behaviour.

An updating on Cryptosporidium parvum in the water buffalo

Abstract A cross-sectional survey of Cryptosporidium parvum infection in the water buffalo was carried out in central Italy. The survey was carried out on a sample of 90 farms, selected using a grid approach within a Geographical Information System, followed by proportional allocation. On each farm, faecal samples were collected from three to five asymptomatic buffalo calves, aged from 1 to 9 weeks (total number = 347). Each sample was tested for the presence of copro-antigens of C. parvum using a commercially available ELISA. Out of the 90 farms, 22 (24.4%) resulted positive. With respect to animals, out of the 347 faecal samples, 51 (14.7%) were found to have antigens of C. parvum. The results of the logistic regression model showed a positive association between the positivity to C. parvum and the high number of buffaloes on farms.

Distribution of helminths in buffalo farms from central Italy

Abstract The aim of the present study was to obtain up-to-date information on the distribution of helminths in buffalo farms of central Italy. The survey was carried out on a sample of 127 farms (epidemiological units), selected using a grid approach within a Geographical Information System, followed by proportional allocation. In each farm the number of buffaloes tested (adults, heifer/steers and calves) ranged from 9 to 21 based on the number of animals on the farm. The total number of faecal samples collected from the 127 farms was 1883. Copromicroscopic examinations were performed using the FLOTACtechnique. The following helminths were detected in the examined farms: gastrointestinal strongyles (33.1%), Strongyloides spp. (3.1%), Fasciola hepatica (7.1%), Dicrocoelium dendriticum (2.4%), Paramphistomidae (7.1%), and Moniezia spp. (2.4%).

Cystic echinococcosis in water buffaloes (Bubalus bubalis)

Abstract An epidemiological and molecular survey of cystic echinococcosis (CE) caused by Echinococcus granulosus in the water buffalo (Bubalus bubalis) of the Italian Mediterranean breed was carried out in the Campania region of southern Italy. Out of a total of 799 water buffaloes examined at slaughterhouses, 80 (10.0%) were found infected. The molecular study was performed on 58 hydatid cysts in order to determine the E. granulosus strain(s) present in this host. A region of cytocrome c oxydase 1 gene (CO1) was amplified by polymerase chain reaction and the PCR products were then purified and sequenced. DNA amplification of the partial CO1 gene gave a 446 bp fragment for all isolates examined. After sequencing, a region of 419 bp was identified for each sample. Thirty-two isolates were identified as the common sheep strain G1, 15 as the buffalo strain G3, 3 as the Tasmanian sheep strain G2, and 3 as the G1 c genotype (GenBank AF458873). In addition, 5 isolates presented 99% identity with the G2 genotype (Tasmanian sheep strain).

First studies on Giardia duodenalis in the water buffalo

Each faecal sample was tested for the presence of copro-antigens of G. duodenalis using a commercially available ELISA. Out of the 90 farms, 27 (30.0%) resulted positive. With respect to animals, out of the 347 faecal samples, 63 (18.1%) were found to have antigens of G. duodenalis. The results of the logistic regression model showed a positive association between the positivity to G. duodenalis and the presence of sheep on farm.
