Effects of dry-rolled or steam-flaked corn finishing diets with or without twenty-five percent dried distillers grains on ruminal fermentation and apparent total tract digestion

A metabolism study was conducted to evaluate ruminal fermentation and apparent total tract digestibilities of cattle finishing diets. Holstein steers (n = 16, 351 kg of BW) with ruminal cannulas were fed diets consisting of 0 or 25% dried corn distillers grains (DDG), using dry-rolled corn (DRC) or steam-flaked corn (SFC) as the principal energy source (2 x 2 factorial arrangement). The study was conducted in 2 periods, with 4 steers per treatment in each period. Periods consisted of a 12-d adaptation phase and a 3-d collection phase. Compared with DRC, feeding SFC decreased intakes of DM, OM, starch, NDF, and ether extract (P < 0.01), and steers fed SFC excreted less DM, OM, starch, NDF, and ether extract (P < 0.01). Compared with SFC, feeding DRC decreased ruminal concentrations of acetate, butyrate, isobutyrate, and isovalerate, and decreased the acetate-to-propionate ratio (P < 0.01). Compared with SFC, DRC decreased ruminal propionate, valerate, and lactate concentrations (P < 0.01). When compared with cattle fed SFC, ruminal pH of cattle fed DRC was less at 0 h and greater at 6 h postfeeding (P < 0.01). Ruminal ammonia concentrations were greater for DRC vs. SFC at h 0, 6, 10, 12, 14, 16, 18, 20, and 22 postfeeding (P < 0.05). Feeding DDG decreased consumption of starch and ether extract, but increased NDF intake (P < 0.01). Fecal excretion of ether extract was increased by adding DDG compared with diets without DDG (P < 0.05), resulting in less apparent total tract digestibility of ether extract for cattle fed DDG (P < 0.01). Ruminal lactate concentrations were increased with addition of DDG compared with diets without DDG (P = 0.01). Ruminal ammonia concentrations were less for steers fed 25 vs. 0% DDG at 2, 4, 6, 8, and 10 h postfeeding (P < 0.05). We conclude, based on these results, that ruminal fermentation and apparent total tract digestibility of DDG are affected by grain processing.

Long-term spatial and temporal trends in frost indices in Kansas, USA

Frost indices such as number of frost days (nFDs), number of frost-free days (nFFDs), last spring freeze (LSF), first fall freeze (FFF), and growing-season length (GSL) were calculated using daily minimum air temperature (Tmin) from 23 centennial weather stations across Kansas during four time periods (through 1919, 1920–1949, 1950–1979, and 1980–2009). A frost day is defined as a day with Tminu2009<u20090xa0°C. The long- and short-term trends in frost indices were analyzed at monthly, seasonal, and annual timescales. Probability of occurrence of the indices was analyzed at 5xa0%, 25xa0%, 50xa0%, 75xa0%, and 95xa0%. Results indicated a general increase in Tmin from 1900 through 2009 causing a decrease in nFDs. LSF and FFF occurred earlier and later than normal in the year, respectively, thereby resulting in an increase in GSL. In general, northwest Kansas recorded the greatest nFD and lowest Tmin, whereas southeast Kansas had the lowest nFD and highest Tmin; however, the magnitude of the trends in these indices varied with location, time period, and time scales. Based on the long-term records in most stations, LSF occurred earlier by 0.1–1.9xa0days/decade, FFF occurred later by 0.2–0.9xa0day/decade, and GSL was longer by 0.1–2.5xa0day/decade. At the 50xa0% probability level, Independence in the south-eastern part of Kansas had the earliest LSF (6 April), latest FFF (29 October) and longest GSL (207xa0days). Oberlin (north-western Kansas) recorded the shortest GSL (156xa0days) and earliest FFF (7 October) had the latest LSF (2 May) at the 50xa0% probability level. A positive correlation was observed for combinations of indices (LSF and GSL) and elevation, whereas a negative correlation was found between FFF and elevation.

Effects of supplementing feedlot steers and heifers with zilpaterol hydrochloride on Warner–Bratzler shear force interrelationships of steer and heifer longissimus lumborum and heifer triceps brachii and gluteus medius muscles aged for 7, 14 and 21 d

Longissimus lumborum (LL) muscles from 117 steers plus LL, gluteus medius (GM), and triceps brachii (TB) muscles from 132 heifers were evaluated for effects of feeding duration of zilpaterol hydrochloride (Zilmax(R); ZH; 7.56g/907kg on a dry matter basis) and aging time on tenderness. Both genders were blocked by initial weight into six blocks of four pens. Pens were assigned to treatments of control (C), or 20, 30 or 40days on ZH, with a 3day withdrawal. Steaks from each subprimal were vacuum aged individually for 7, 14 or 21days, frozen, thawed, and cooked to 71 degrees C for Warner-Bratzler shear force (WBSF). All muscles from steers and heifers from ZH30 and ZH40 treatments had higher (P<0.05) WBSF than those of C. The WBSF of steer LL and heifer TB from the ZH20 treatment was higher (P<0.05) than C. There was a treatment by aging interaction (P>0.05) for WBSF of GM steaks from heifers. Percentage of intramuscular fat had little effect on tenderness. Percentages of steer LL and heifer TB steaks with WBSF values below thresholds of either 5.0 or 4.6kg from the ZH20 treatment were quite high, whereas percentages of heifer LL and GM muscles below 5.0kg (67%) and 4.6kg (57%) were low. Feeding ZH20days generally increased WBSF values, but mean WBSF values for steer LL and heifer TB were below 4.6kg. Feeding ZH 20days resulted in>40% of GM steaks with WBSF values above 4.6kg.

Cooked yields, color, tenderness, and sensory traits of beef roasts cooked in an oven with steam generation versus a commercial convection oven to different endpoint temperatures

A CVap steam generation oven was compared with a Blodgett convection oven to examine effects on yields, cooked color, tenderness, and sensory traits of beef Longissimus lumborum (LL), Deep pectoralis (DP), and Biceps femoris (BF) muscles cooked to 1 of 3 temperatures (65.6, 71.1, or 76.7°C). Four roasts were cooked in the CVap for a constant time, and 2 roasts were cooked in the Blodgett until they reached target temperatures (3 replications). Cooking yields were higher (P<0.05) for BF and LL roasts in the CVap. Slice shear force (SSF) for BF roasts was lowest (P<0.05) in the CVap but lowest (P<0.05) for DP roasts in the Blodgett. No oven effect (P>0.05) was found for LL roasts. Sensory tenderness for BF roasts in the CVap was higher (P<0.05) than those in the Blodgett. Juiciness was higher (P<0.05) for LL roasts in the Blodgett. The CVap oven offers some tenderization (BF) and cooking yield advantages (BF and DP) over forced-air convection cooking.

A dynamic role for sterols in embryogenesis of Pisum sativum

Molecular roles of sterols in plant development remain to be elucidated. To investigate sterol composition during embryogenesis, the occurrence of 25 steroid compounds in stages of developing seeds and pods of Pisum sativum was examined by GC-MS analysis. Immature seeds containing very young embryos exhibited the greatest concentrations of sterols. Regression models indicated that the natural log of seed or pod fr. wt was a consistent predictor of declining sterol content during embryonic development. Although total sterol levels were reduced in mature embryos, the composition of major sterols sitosterol and campesterol remained relatively constant in all 12 seed stages examined. In mature seeds, a significant decrease in isofucosterol was observed, as well as minor changes such as increases in cycloartenol branch sterols and campesterol derivatives. In comparison to seeds and pods, striking differences in composition were observed in sterol profiles of stems, shoots, leaves, flowers and flower buds, as well as cotyledons versus radicles. The highest levels of isofucosterol, a precursor to sitosterol, occurred in young seeds and flower buds, tissues that contain rapidly dividing cells and cells undergoing differentiation. Conversely, the highest levels of stigmasterol, a derivative of sitosterol, were found in fully-differentiated leaves while all seed stages exhibited low levels of stigmasterol. The observed differences in sterol content were correlated to mRNA expression data for sterol biosynthesis genes from Arabidopsis. These findings implicate the coordinated expression of sterol biosynthesis enzymes in gene regulatory networks underlying the embryonic development of flowering plants.

Effects of feeding a single or sequence of beta-adrenergic agonists on cull cow meat quality

Sixty cull cows were implanted and assigned to four treatments: C = concentrate ration only; RH = supplemented with ractopmaine-HCl (8.33 mg/kg of feed) for 25 d; ZH = supplemented with zilpaterol-HCl (ZH) (200mg head (-1)d(-1)) for the last 20 d; and RH + ZH = supplemented with RH for 25 d followed by ZH for 20 d. All cows were fed a concentrate ration for 74 d. Infraspinatus steaks from cows supplemented with RH and/or ZH had lower (P<0.05) shear force than steaks from C cows. Longissimus (LM) steaks from the 6-8th rib section of ZH and RH+ZH cows had decreased (P<0.0001) desmin degradation at 10 and 21 d postmortem compared to steaks from C and RH cows. Collagen solubility of the LM was increased (P<0.05) by ZH and RH+ZH compared to C. There were no treatment differences in 12th rib LM tenderness when enhanced with calcium lactate. Color and sensory traits of meat from RH+ZH cows were not different from C but flavor intensity was greater and off-flavor less than for C cows.

Melanism in a Chinese Population of Harmonia axyridis (Coleoptera: Coccinellidae): A Criterion for Male Investment with Pleiotropic Effects on Behavior and Fertility

In Beijing, China, females of Harmonia axyridis are promiscuous but prefer typical (succinea form) males to melanic ones in the spring generation, ostensibly due to the thermal disadvantages of melanism during summer. We used laboratory observations to test whether males invested differentially in females according to their elytral color, and whether male behavior was phenotype-dependent. Video-recording was used to monitor no-choice mating tests between virgin adults in all phenotype combinations and females were isolated post-copula to observe their egg retention times and reproduction over 5xa0days. Females tended to wait longer before using the sperm of melanic males, and melanic females delayed longer than succinic females. Melanic males spent longer in copula with succinic than melanic females and the latter received fewer bouts of male abdominal shaking that correlate with sperm transfer, regardless of the phenotype of their mate. Although melanic males abandoned melanic females faster than did succinic males, they remained in copula with females of both phenotypes for a longer period after shaking, suggesting a larger investment in mate guarding by the less-preferred male phenotype. Although female fecundity did not vary among phenotype combinations, egg fertility was lower for females mated to melanic males, suggesting a pleiotropic effect of melanism on male fertility in addition to its effects on male mating behavior.

Effects of sequential feeding of β-adrenergic agonists on cull cow performance, carcass characteristics, and mRNA relative abundance

The objectives of this study were to determine the effects of supplementation with a single β-adrenergic agonist (β-AA) or a sequence of β-AA on cow performance, carcass characteristics, and mRNA relative abundance of cull cows implanted and fed a concentrate diet. Sixty cull cows were implanted with Revalor-200 (200 mg of trenbolone acetate and 20 mg of estradiol) and assigned to 1 of 4 treatments (n = 15/treatment): CON = fed a concentrate diet only; RH = supplemented with ractopamine-HCl for the last 25 d before slaughter; ZH = supplemented with zilpaterol-HCl for 20 d before a 3-d withdrawal before slaughter; RH + ZH = supplemented with RH for 25 d, followed by ZH for 20 d before a 3-d withdrawal before slaughter. Ractopamine-HCl was supplemented at a dose of 200 mg·animal(-1)·d(-1), and ZH was supplemented at 8.33 mg/kg (100% DM basis) of feed. All cows were fed a concentrate diet for 74 d. Each treatment had 5 cows per pen and 3 replicate pens. Body weights were collected on d 1, 24, 51, and 72. Muscle biopsies from the LM were collected on d 24, 51, and at slaughter from a subsample of 3 cows per pen. Carcass traits were evaluated postslaughter. The 2 ZH treatments averaged 15.3 kg more BW gain, 0.20 kg greater ADG, and 7.8 cm(2) larger LM area than CON and RH treatments, and 21 kg more HCW than CON, but these differences were not significant (P > 0.10), likely due to a sample size of n = 15/treatment. The sequence of RH followed by ZH tended to optimize the combination of HCW, LM area, percent intramuscular fat, and lean color and maturity compared with the ZH treatment. Abundance of β(2)-adrenergic receptor (AR) mRNA was not altered in the RH + ZH treatment during RH supplementation from d 24 to 51 of feeding. However, the abundance of β(2)-AR mRNA increased (P < 0.05) the last 23 d of feeding for the RH treatment and tended (P = 0.10) to increase in ZH cows during ZH supplementation. For all cows, abundance of type IIa myosin heavy chain (MHC-IIa) mRNA decreased (P < 0.05) after 24 d of feeding. Abundance of MHC-IIx mRNA increased (P < 0.05) for ZH and RH + ZH treatments the last 23 d of feeding during ZH supplementation. Although few significant differences were observed in performance or carcass traits, mRNA quantification indicated that β-AA supplementation elicited a cellular response in cull cows. Implanting and feeding cull cows for 74 d, regardless of β-AA supplementation, added economic value by transitioning cows from a cull cow to what is referred to in industry as a white cow market in which cows have white fat resulting from grain feeding.

Effect of mouse antisera targeting the Phlebotomus papatasi midgut chitinase PpChit1 on sandfly physiology and fitness

In sandflies, the absence of the peritrophic matrix (PM) affects the rate of blood digestion. Also, the kinetics of PM secretion varies according to species. We previously characterised PpChit1, a midgut-specific chitinase secreted in Phlebotomus papatasi (PPIS) that is involved in the maturation of the PM and showed that antibodies against PpChit1 reduce the chitinolytic activity in the midgut of several sandfly species. Here, sandflies were fed on red blood cells reconstituted with naïve or anti-PpChit1 sera and assessed for fitness parameters that included blood digestion, oviposition onset, number of eggs laid, egg bouts, average number of eggs per bout and survival. In PPIS, anti-PpChit1 led to a one-day delay in the onset of egg laying, with flies surviving three days longer compared to the control group. Anti-PpChit1 also had a negative effect on overall ability of flies to lay eggs, as several gravid females from all three species were unable to lay any eggs despite having lived longer than control flies. Whereas the longer survival might be associated with improved haeme scavenging ability by the PM, the inability of females to lay eggs is possibly linked to changes in PM permeability affecting nutrient absorption.

Digestibility of dried distiller’s grains with solubles in steam-flaked or dry-rolled corn diets

In previous experiments, we observed that the nutritional value of dried distiller’s grains is less when added to finishing diets made of steam-flaked corn than when added to diets of dry-rolled corn. We hypothesized that effects of grain processing on value of distiller’s grains are attributable to differences in the digestion characteristics of grains processed via flaking or dry rolling. In this study, our objective was to evaluate differences in ruminal metabolism and total tract digestion of diets made from dry-rolled or steam-flaked corn with and without dried distiller’s grains.
