Prophylactic effects of duloxetine on post-stroke depression symptoms: an open single-blind trial.

Post-stroke depression (PSD) is a common yet severe sequela of stroke, and is often accompanied with somatic symptoms. Duloxetine, a new serotonin-norepinephrine reuptake inhibitor, may help to prevent depression after stroke. 95 ischemic stroke patients without depression were randomly divided into two groups: duloxetine group (n = 47) and control group (n = 48). Patients in the control group received routine ischemic stroke therapy, whereas patients in the duloxetine group received duloxetine (dose range 30–90 mg) for 12 weeks in addition to routine therapy. Follow-up observations lasted for 24 weeks. The Hamilton Depression Scale was used to measure depression, and the National Institute of Stroke Scale, Mini-Mental State Examination, Activities of Daily Living Scale (Chinese version) and Short Form 36 Health Survey Questionnaire were used to assess neurological function, cognitive function, rehabilitation from stroke and quality of life. Results showed that in general, duloxetine spared ischemic stroke patients from both minor and major depression by 16%. In addition, duloxetine helped patients to rehabilitate more rapidly from stroke, and was associated with better cognitive function and quality of life. In conclusion, the prophylactic use of duloxetine not only decreased the incidence of PSD, but also promoted rehabilitation, cognitive function and quality of life.

Caspase-8 Mediates Amyloid-β-induced Apoptosis in Differentiated PC12 Cells

The pathogenesis of Alzheimer’s disease (AD) is very complex and there are currently no significant treatments for the disease. Caspase-8 is known to be involved in neuronal apoptosis. To explore a possible molecular mechanisms involved in AD pathology, this study investigated the effect of caspase-8 knockdown on amyloid-β 1–40 (Aβ1–40)-induced apoptosis in PC12 cells. The proliferation of PC12 cells was significantly inhibited in Aβ-treated cells, and a high fraction of the cells underwent apoptosis in a dose- and time-dependent manner. Transfection of caspase-8 small interfering RNA (siRNA) resulted in reduced apoptosis following Aβ1–40 treatment. The activation of caspase-3, caspase-8, and caspase-9 was stimulated by Aβ1–40, an effect that was also significantly reduced by caspase-8 siRNA. Knockdown of caspase-8 increased the phosphorylation of the signaling molecules AKT and ERK1/2 relative to cells treated with Aβ1–40 alone. Caspase-8 is an important effector molecule involved in apoptosis induced by Aβ1–40 and is likely involved in AD pathology. This study suggests that targeted inhibition of caspase-8 may be a new therapeutic for preventing neuronal apoptosis and inhibiting the progression of AD.

Neuroinflammation has always been of concern in the pathogenesis of Alzheimer’s disease (AD). As a major inflammatory mediator, prostaglandin E2(PGE2) plays an important role in the inflammatory process of AD. Up to now, there is still controversy on the neuroprotective or neurotoxic role of PGE2. However, the role of PGE2 in neurodegeneration may be far more complex, due to the 4 EP receptor subtypes. This article aims to summarize the relationship between PGE2 receptor EP subtypes and AD. Evaluation of the risk factors of depressive disorders comorbid with obstructive sleep apnea.

Objective Overlap of obstructive sleep apnea (OSA) complicates diagnosis of depressive disorder and renders antidepressant treatment challenging. Previous studies have reported that the incidence of OSA is higher in patients with depression than in the general population. The purpose of this article was to investigate clinical risk factors to predict OSA in depression disorders. Methods A total of 115 patients diagnosed with major depressive disorder (MDD) and bipolar disorder (in a major depressive episode), who underwent overnight polysomnography, were studied retrospectively. They were divided into two groups: non-OSA and OSA. The patients who had apnea–hypopnea index (AHI) <5 were defined as the non-OSA group, whereas the OSA group was defined as those with an AHI ≥5. Logistic regression was used to analyze the association among AHI and clinical factors, including sex, age, body mass index (BMI), Hamilton Depression Rating Scale (HAMD), Hamilton Anxiety Rating Scale, Pittsburgh Sleep Quality Index (PSQI), and diagnosis (MDD or bipolar disorder [in a major depressive episode]). Results In 115 patients, 51.3% had OSA. Logistic regression analysis showed significant associations between AHI and diagnosis (MDD or bipolar disorder [in a major depressive episode]), BMI, HAMD, and PSQI (P<0.05). Conclusion The findings of our study suggested that the rate of depression being comorbid with OSA is remarkably high and revealed that there is a high rate of undetected OSA among depressive disorder patients and untreated OSA among mood disorder patients. The clinical risk factors (diagnosis [MDD or bipolar disorder {in a major depressive episode}], BMI, HAMD, and PSQI) could predict AHI or OSA diagnosis and contribute to OSA screening in depressive disorder patients.

Personality traits and perception of Müller-Lyer illusion in male Chinese military soldiers and university students

Abstract Background In military men, performance such as gun-shooting precision relies on factors such as the ability to resist visual illusion, and this misperception of visual stimulus might be linked with sensation seeking related personality. Methods We have invited 103 male military men and 104 age-matched university male students to undergo the experiment of the Brentano version of the Müller-Lyer illusion and the Zuckerman-Kuhlman Personality Questionnaire (ZKPQ) tests. Results The military men scored significantly lower than students did on the ZKPQ Impulsive Sensation Seeking test but higher on Aggression-Hostility and Sociality test, and displayed less misperception magnitude to the illusion. The Impulsive Sensation Seeking, Neuroticism-Anxiety and Aggression-Hostility traits in military men, and the Activity in students were respectively correlated with the misperception magnitudes of the illusion in different manners. Conclusion Limited results in our study have indicated that the military men had pronounced personality traits which were correlated with the misperception magnitude of the Müller-Lyer illusion.
