This research aimed to investigate the effect of thermal processing on carotenoid profile, quantity and stability in 22 colour-fleshed potato cultivars grown in the Czech Republic. The total of nine carotenoids was analysed by HPLC using a C30 column and PDA detection. The total carotenoid content for all cultivars ranged from 1.44 to 40.13 μg/g DM. Yellow cultivars showed a much higher average total carotenoid content (26.22 μg/g DM) when compared to red/purple-fleshed potatoes (5.69 μg/g DM). Yellow cultivars were dominated by antheraxanthin, whereas neoxanthin was the main carotenoid in red/purple cultivars. Thermal processing significantly impacted all potato cultivars. Boiling decreased the total carotenoids by 92% compared to baking (88%). Lutein was the most stable carotenoid against thermal processing (decreased by 24-43%) followed by β-carotene (decreased by 78-83%); other carotenoids were degraded nearly completely. Increased formation of (Z)-isomers by thermal processing has not been confirmed.

Hydrogen sulfide, one of three known gasotransmitters, is involved in physiological processes, including reproductive functions. Oocyte maturation and surrounding cumulus cell expansion play an essential role in female reproduction and subsequent embryonic development. Although the positive effects of exogenous hydrogen sulfide on maturing oocytes are well known, the role of endogenous hydrogen sulfide, which is physiologically released by enzymes, has not yet been described in oocytes. In this study, we observed the presence of Cystathionine β-Synthase (CBS), Cystathionine γ-Lyase (CTH) and 3-Mercaptopyruvate Sulfurtransferase (3-MPST), hydrogen sulfide-releasing enzymes, in porcine oocytes. Endogenous hydrogen sulfide production was detected in immature and matured oocytes as well as its requirement for meiotic maturation. Individual hydrogen sulfide-releasing enzymes seem to be capable of substituting for each other in hydrogen sulfide production. However, meiosis suppression by inhibition of all hydrogen sulfide-releasing enzymes is not irreversible and this effect is a result of M-Phase/Maturation Promoting Factor (MPF) and Mitogen-Activated Protein Kinase (MAPK) activity inhibition. Futhermore, cumulus expansion expressed by hyaluronic acid (HA) production is affected by the inhibition of hydrogen sulfide production. Moreover, quality changes of the expanded cumuli are indicated. These results demonstrate hydrogen sulfide involvement in oocyte maturation as well as cumulus expansion. As such, hydrogen sulfide appears to be an important cell messenger during mammalian oocyte meiosis and adequate cumulus expansion.

Colored-grain spring and winter wheat, spring tritordeum and barley (blue aleurone, purple pericarp, and yellow endosperm) from the harvests 2014 and 2015 were evaluated for tocol contents by HPLC-FD. Higher content of total tocols was found in spring wheat varieties compared with winter varieties. Four tocols (β-tocotrienol, α-tocotrienol, β-tocopherol, and α-tocopherol) were identified in wheat and tritordeum varieties. Dominant tocols in purple- and blue-grained wheat and yellow-grained tritordeum were α-tocopherol and β-tocotrienol, whereas spring barley varieties differed from wheat and tritordeum by high α-tocotrienol content. Tocol content was significantly affected by genotype and in a lesser extent in some varieties and lines also by rainfall and temperatures during crop year. Higher rainfall and lower temperatures caused in most varieties higher tocol contents. Purple- and blue-grained wheat lines with higher tocol, anthocyanin and phenolic acids with health benefits may be useful for breeding new varieties.

Carotenoids are important phytonutrients responsible for the yellow endosperm color in cereal grains. Five carotenoids, namely lutein, zeaxanthin, antheraxanthin, α- and β-carotene, were quantified by HPLC-DAD-MS in fourteen genotypes of wheat, barley and tritordeum harvested in Czechia in 2014 and 2015. The highest carotenoid contents were found in yellow-grained tritordeum HT 439 (12.16μg/gDW), followed by blue-grained wheat V1-131-15 (7.46μg/gDW), and yellow-grained wheat TA 4024 (7.04μg/gDW). Comparing carotenoid contents, blue varieties had lower whereas purple ones had the same or higher levels than conventional bread wheat. Lutein was the main carotenoid found in wheat and tritordeum while zeaxanthin dominated in barley. The majority of cereals contained considerable levels of esterified forms (up to 61%) of which lutein esters prevailed. It was assessed that cereal genotype determines the proportion of free and esterified forms. High temperatures and drought during the growing season promoted carotenoid biosynthesis.

The cumulus expansion of cumulus-oocyte complex (COC) is an essential regulating process of oocyte maturation and as such it is a possible biomarker of the in vitro maturing oocytes quality. Cumulus expansion is usually assessed by non-invasive methods based on visual evaluation with many inaccuracies. On the other hand, analytical measurement of the quantity of hyaluronic acid (HA), the most abundant compound of expanded cumuli, is one of possible methods to evaluate cumulus expansion precisely. Therefore, this study aimed to verify the applicability of HA analysis for evaluating the cumulus expansion and testing oocyte maturation. The COCs were cultured in modified M199 medium for 8–48 h. The samples for the HA analysis were prepared on an 8-hour time scale, and HA retained in COCs was measured using a spectrophotometric method adapted for this purpose. We observed an increasing quantity of HA during the in vitro cultivation. A comparison with expanded COCs’ classification or expansion area proved the proposed method of HA analysis suitable for the evaluation of cumulus expansion in vitro. Our findings consider the quantity of HA-expressed cumulus expansion to be a valuable marker of COC quality enabling an adequate oocyte meiotic stage estimation.

Abstract Cumulus expansion of the cumulus-oocyte complex is necessary for meiotic maturation and acquiring developmental competence. Cumulus expansion is based on extracellular matrix synthesis by cumulus cells. Hyaluronic acid is the most abundant component of this extracellular matrix. Cumulus expansion takes place during meiotic oocyte maturation under in vivo and in vitro conditions. Quantification and measurement of cumulus expansion intensity is one possible method of determining oocyte quality and optimizing conditions for in vitro cultivation. Currently, subjective methods of expanded area and more exact cumulus expansion measurement by hyaluronic acid assessment are available. Among the methods of hyaluronic acid measurement is the use of radioactively labelled synthesis precursors. Alternatively, immunological and analytical methods, including enzyme-linked immunosorbent assay (ELISA), spectrophotometry, and high-performance liquid chromatography (HPLC) in UV light, could be utilized. The high sensitivity of these methods could provide a precise analysis of cumulus expansion without the use of radioisotopes. Therefore, the aim of this review is to summarize and compare available approaches of cumulus expansion measurement, respecting special biological features of expanded cumuli, and to suggest possible solutions for exact cumulus expansion analysis.

Heavy metals are among the most serious environmental contaminants in mining districts. Soil, as one of the main components of the environment, is the place of heavy metal entry into plants and consequently into the food chain, too. Potatoes grown in the region of Middle Spis (Slovakia) may be a source of increased content of heavy metals and pose a health risk to the consumer. The contents of heavy metals (Cd, Pb, and Ni) in potato and soil samples were determined using the AAS method and compared with limit values set by the Slovak Republic and the European Union. The content of heavy metals was determined in 12 potato cultivars with different length of vegetation period (mid-early, very early, and early, resp.), which were grown in three localities with a highly disturbed environment. Total contents and mobile forms of heavy metals as well as physical and chemical properties were determined in soil samples which were collected from the same sampling sites. Only Pb content in potato tubers was higher than the hygienic limit value (0.1 mg kg−1 FM) in 15 sampling sites (interval was n.d. –0.2298 mg kg−1 FM). The contents of exchangeable forms (total content) of heavy metals in soil were ranged between the intervals: Cd 0.004–0.055 (0.94–1 56), Pb 0.023–0.295 (17.00–26.80), and Ni 0.019–0.475 (30.80–71.00) mg kg−1. At current average consumption levels of potatoes, tolerable weekly intake (TWI) or tolerable daily intake (TDI) for observed heavy metals was not exceeded.

Carotenoids are one of major lipophilic constituents contributing to total antioxidant activity and provitamin content of potato, a major non-cereal staple food. The review briefly discusses health promoting properties of carotenoids and especially their contents and composition in different potato cultivars affected by flesh colour (white-, yellow-, purpleand red-fleshed) and the effect of selected factors on carotenoid total and individual levels, such as genotype, breeding, tuber development, heat processing – cooking, storage, effect of year, locality, etc. The aim of the recent research is obtaining potatoes with higher levels of beneficial carotenoids to improve one the most popular vegetables in the world.

Sulphur is one of the most important elements in plants. Sulphur also positive influences on growth functions (nitrogen metabolism, enzyme activity and protein and oil synthesis), yield and quality bulbs of onion. In this experiment was watched the progress of making the total polyphenols content in different levels of fertilizer sulphur in onion during vegetation. In the work we watched also the influence of sulphur fertilization on the antioxidant activity. Sulphur was added in amounts 7.3 mg.kg -1 S, 11 mg.kg -1 S and 14,6 mg.kg -1 S. Samples of fresh onion we collected at the beginning, in the middle and in the end of vegetation period and we prepared an extract: 50 g cut onion extracted by 100 ml 80% ethanol according sixteen hours. These extracts were used for experiment. The content of total polyphenols was estimated by using Folin-Ciocalteau reagent. The absorbance was measured at 765 nm of wave length against blank. Antioxidant activity was measured using a compound DPPH˙ (2.2-diphenyl-1-picrylhydrazyl) at 515.6 nm in the spectrophotometer. Our values of total polyphenolic content during vegetation period were in range from 508.16 ±27.59 mg.kg -1 to 638.2 ±12.84 mg.kg -1 . The highest content of total polyphenolics was recorded at the end of the vegetation period (638.32 ±12.84 mg.kg -1 ) in III. sampling (incorporation of sulphur in quantity of 14.6 mg S.kg -1 soil). This increase was statistically significant (P-value = 2.10 -3 ). The lowest content of total polyphenols was measured in the middle of vegetation period (415.41 ±13.32 mg.kg -1 ) in II. sampling (incorporation of sulphur in quantity of 11 mg S.kg -1 soil). Another indicator that has been evaluated and compared was the influence of sulphur fertilization on the antioxidant activity. The highest values of antioxidant activity were recorded at the end of vegetation period in all variants. The lowest value of antioxidant activity (25.41 ±7.67%) was measured in three sampling in II.variant.
