Optimum Feeding Rates in Juvenile Olive Flounder Paralichthys olivaceus Fed Practical Expanded Pellet at Low and High Water Temperatures

Two feeding trials were conducted to investigate the optimum feeding rates in juvenile olive founder Paralichthys olivaceus fed practical expanded pellet(EP) containing 58.1% crude protein, 10.7% crude lipid, and 10.8% ash at low and high water temperatures. In the first experiment, triplicate groups of 15 fish with average weight of 7.7 g were fed at one of seven feeding rates(0, 1, 1.5, 2, 2.25, or 2.5% of body weight/day or to satiation) for 4 weeks at low water temperature. In the second experiment, quadruplicate groups of 20 fish with average weight of 5.5 g were fed at one of eight feeding rates(0, 3, 3.5, 4, 4.25, 4.5, or 4.75% of body weight/day or to satiation) for 4 weeks at high water temperature. Based on growth performance, we estimated that the optimum feeding rates for juvenile olive flounder were 1.97-2.51% and 4.82-6.36% of body weight/day at low and high water temperatures, respectively. RNA-seq transcriptome analysis of the olive flounder (Paralichthys olivaceus) kidney response to vaccination with heat-inactivated viral hemorrhagic septicemia virus.

ABSTRACT Viral hemorrhagic septicemia (VHS) is a highly contagious disease of cultured flounder caused by VHS virus (VHSV). To develop effective VHSV vaccines, it is essential to understand the molecular mechanisms underlying the hosts protective response against VHSV. The purpose of this study is to clarify which genes are involved in the protective response of olive flounder after VHSV vaccination. We first injected olive flounder intraperitoneally with 107 TCID50 heat‐inactivated VHSV vaccine and evaluated the vaccine efficacy at 20 °C. Fish vaccinated with heat‐inactivated VHSV were significantly protected compared to non‐vaccinated fish, with a relative percentage survival of 83%. To analyze the vaccination‐induced changes in the expression profiles of genes, kidneys were collected from control and vaccinated fish at days 1, 3, and 7 after vaccination and global gene expression profiling was carried out by RNA sequencing. The analysis revealed that 15,001 genes were differentially expressed by at least 2‐fold between vaccinated fish and non‐vaccinated controls. Of these, 58 genes clustered into the acute phase response, Toll‐like receptor, interferon‐inducible/regulatory proteins, and apoptosis pathways. These data provided insights into the molecular mechanisms underlying the protective immune response of olive flounder against heat‐inactivated VHSV vaccine and might aid future studies to develop a highly immunogenic vaccine against VHSV in flounder. HighlightsViral hemorrhagic septicemia virus (VHSV) is a major pathogen of cultured flounder.We analyzed the transcriptomic response to heat‐inactivated VHSV in olive flounder.In total, 15,001 genes were differentially expressed after vaccination.Data analysis delineated 4 functional pathways responsive to VHSV vaccination.These findings can aid the future development of highly immunogenic VHSV vaccines.

An Economic Analysis of Black Rockfish, Sebastes schlegeli in the Marine Floating Cage Culture

This study is aimed to analyze the economic performance of black rockfish aquaculture by standardizing cost structures by region and farming size. The result of survey on farming and sale condition in each region, stocking density, survival rate, juvenile price, and unit production was the highest at Yeosu and Tongyeong, Heuksando showed the lowest. While rearing period was the longest at Tongyeong, the shortest region was Yeosu and Wando. In farming cost structure by region, amount of feeing was the highest at Tongyeong and Yeosu, and the lowest was Heuksando. Cost of medicine was high in Wando and Taean region, Yeosu and Hecksando was low. In case of farm size, feed cost ratio was high in the order of medium(0.75ha), small(0.25ha) and large(1.25ha) size. Standard production cost at every farm size of Heuksando showed the lowest among these regions. Taean and Yeosu was middle, and Tongyeong and Wando was the highest. According to the income, profit rate and investment return of farm size in all regions, as the bigger farm size, the higher income and profit rate was revealed. However, in case of Wando, Taean, and Heuksando which regions has high investment return, medium farm size was higher than large size. The result of economic analysis according to various factors, economic feasibility of black rockfish aquaculture in marine floating cage was showed significant changes by rearing and market condition.

Complete Genome Sequence of Nervous Necrosis Virus Isolated from Sevenband Grouper (Epinephelus septemfasciatus) in South Korea

ABSTRACT The draft genome sequence of the nervous necrosis virus (NNV) SGYeosu08, isolated from sevenband grouper (Epinephelus septemfasciatus) in Yeosu, South Korea, was cloned and analyzed. The full-length RNA1 was a 3,103-nucleotide-encoding region of RNA-dependent RNA polymerase, and the RNA2 encoding a coat protein was 1,433 nucleotides in length. This genome sequence might be useful in the development of an accurate diagnostic tool.

A Study on the Comparative Analysis of Business Performance of Abalone Seed, Haliotis discus hannai by Region and Farming Size in the Land-based System

This study is aimed comparative analysis for business performance of land-based ezo abalone (Haliotis discus hannai) seed farms by standardizing cost structures by region and farming size. The result of survey on average farming incomes by region showed that farming incomes in Haenam and Jindo regions were much higher than those in other regions. Followed by Wando region, incomes in other regions were analyzed to be relatively lower. It is shown that farming incomes became higher as farm size increased. Farming incomes per unit size which were modified from farming incomes by region were highest in Jindo region, followed by East sea region, Wando and Haenam regions. Incomes in Jeju and Yeosu regions were analyzed to be relatively lower. Results on farming incomes per unit size () showed that both farming incomes and profits became higher as size increased. It implies that a scale effectiveness might exist in case of land-based abalone culture system. Impacts of major factors on farming profits and returns on investment in abalone seed aquaculture are summarized as follow. First, only if the survival rate increased by 10% with improvement of component ratio, variable effects became largest. In variable effects of other factors, a variation in Jindo region was largest and on the other hand, a variation in Yeosu region was shown to be smallest.

Effects of Estradiol-17β on the Feminization of Japanase Eel, Anguilla japonica

The effects of oral administration of estradiol- (E2) on glass eels (Body weight: , Total length: ) and young eels (Body weight: , Total length: ) on gonadal sex and growth were examined, respectively. Glass eels were fed a diet containing E2 at a dose of 10 mg/kg or 25 mg/kg, respectively, for five months. The female ratio significantly increased in all E2-treated groups (10 mg/kg diet group: 70%; 25 mg/kg diet group: 90%) when compared to the control group (10%). Young eels were fed a diet containing E2 at a dose of 25 mg/kg for four months. The female ratio also significantly increased in the E2-treated groups (60%) compared to the control group (20%). The highest female ratio was observed in the stage of glass eels rather than young eels. In all experiments, however, the growth of eels treated with E2 was similar to that of controls to the end of the experiment. Thus, oral administration of E2 could be a good approach to controlling sex differentiation.

Effects of a Commercial Extruded Pellet on Growth Performance and Water Quality in Growing Olive Flounder Paralichthys olivaceus

This study was conducted to evaluate the effect of commercial extruded pellet (EP) diet on the growth and water quality compared with a raw fish moist pellet (MP) diet for flounder on field feeding experiments. Three replicate groups of 130 fish per each tank (initial weight of 380 g) were fed one of the EP and Moist pellet (MP) for 7 months. In field feeding experiment, no significant difference was observed in final mean weight of fish fed the experimental EP (1,233 g) and MP (1,237 g), and any noticeable problem caused by feeding the experimental EP was not found during the whole period of feeding trial. Feed efficiency of fish fed the experimental EP was higher than that of MP, and there was no significant difference in survival between the fish fed experimental EP and MP. The concentrations of SS, COD, TN and TP of seawater in tanks of MP groups were 1-5 times higher than those of EP groups. Therefore, these results strongly suggest that diet EP could be developed to replace MP diet for the marketable size of production for flounder without adverse effects on growth performance and water quality.

Montanide IMS 1312 VG adjuvant enhances the efficacy of immersion vaccine of inactivated viral hemorrhagic septicemia virus (VHSV) in olive flounder, Paralichthys olivaceus

ABSTRACT Vaccination by immersion is suitable for mass vaccination of small size fish. However, no viral vaccine has been developed for immersion applications, because of low efficacy. In this study, we evaluated the efficacy and safety of immersion vaccine against viral hemorrhagic septicemia (VHS) containing Montanide IMS 1312 VG adjuvant in olive flounder (Paralichthys olivaceus). Healthy fish were vaccinated by an immersion method with a heat‐inactivated FP‐VHS2010–1 strain of VHS virus (VHSV) in combination with Montanide IMS 1312 VG for 5 min at 20 ± 2 °C. The control group was vaccinated with sterile PBS. No toxicity of immersion vaccine with Montanide IMS 1312 VG adjuvant was observed by hematological and histopathological analysis. Immersion vaccine with adjuvant enhanced gene expression of immune‐associated genes, i.e., genes encoding interleukin (IL)‐1&bgr;, IL‐6, IL‐8, and Toll‐like receptor (TLR) 3. Relative percent survival (RPS) of fish was measured on weeks 4 and 8 post vaccination. In fish vaccinated with adjuvant, RPS was significantly higher than that of fish vaccinated without adjuvant. The results of the present study provide evidence that the VHSV immersion vaccine with Montanide IMS 1312 VG induces protective immunity in olive flounder against VHS. HIGHLIGHTSImmersion vaccination of heat‐inactivated VHSV with Montanide 1312 VG protected olive flounder from VHS.Immersion vaccination inceased the levels of several immune genes.Addition of Montanide 1312 VG further increased the levels of immune genes.Duration of protective effect of immersion vaccination was short.

Early Life History of the Sevenband Grouper, Epinephelus septemfasciatus from Korea

This study is conducted to monitor the morphological developmental features of the egg development, larvae and juvenile of Epinephelus septemfasciatus, the fertilized eggs were gotton using artificial insemination. Matured parents are collected from marine caged fish farms in Geomun-ri, Samsan-myeon, Yeosu-si, Jeollanamdo Korea in June 2012. The fertilized eggs were pelagic eggs containing one oil globule, and measured 0.81~0.89 mm (0.85±0.04 mm, n=50) in diameter. In regard to rearing environment, the water temperature is 21.0~23.0°C and the salinity is 32.0~33.2‰. Hatching was observed from 48 hours after fertilization, the mouth and anus of prelarvae was not opened but had egg yolk at newly hatched. 4 days after hatching, the mouth and anus of postlarvae was opened and began to eat Rotifer and was measured 2.40~2.49 mm (2.45±0.03 mm n=10) in total length. 12 days after hatching, postlarvae was measured 3.77~4.67 mm (4.27±0.33 mm) in total length, its the second pole tide of dorsal fin and the first pole tide of pelvic fin was extended longitudinally. 71 days after hatching, juvenile was measured 40.5~45.4 mm (42.6±2.04 mm) in total length. Seven bands were observed in body, and pole tides of dorsal and pelvic fins were shortened.

The Histo-Pathological Change of Rockfish, Sebastes Schlegeli fed the Extruded Pellet and a Raw Fish-based Moist Pellet in Marine Net Cage

A histo-pathogical examination was carried out to evaluate the effect of commercial extruded pellet (EP) and a raw fish moist pellet (MP) diet on the health of juvenile rockfish cultured in marine net-cage for 7 months. Fish were distributed randomly to each net cage as a group of 76,000 fish (initial mean body weight 5.9 g). After 2 months, the hypertrophy or swelling of liver parenchymal cells was identified in most individuals and lasted until 7 months. Livers in EP fed group frequently showed hypertrophic parenchyma and fatty change with occasional atrophic cells. However, after 4 months, lymphocytic infiltration in splenic parenchyma was seen in a number of individuals. In addition, the gastric glandular epithelium was atrophied and in the lumen of renal tubules protozoan parasites were frequently identified but there was no correlation with the type of feed. Moreover, juvenile rockfish on EP diet showed gross and microscopic hypertrophy of the liver which would be due to oversupply of feed. Severe hepatic cellular hypertrophy or swelling could lead to the damage of microcirculation. Especially fatty change and atrophic change of liver could be the result from the damage, which could be responsible for immunological problem. Lymphocytic infiltration of spleen on the MP diet suggests that juvenile rockfish could be frequently exposed to infectious antigens.
