Objective measurement of the freshness of fish is important from the viewpoint of food sanitation. The most widely used method of testing for freshness is the K value method, and the K value is defined as the percentage of inosine and hypoxanthine among six kinds of adenosine triphosphate-(ATP)- related compounds. The method is based on chemical analysis of breakdown products of ATP in the fish muscle, employing complicated pretreatment procedures and expensive instruments. Thus, the method is destructive analysis and requires much effort and time. The amounts of volatile bases such as trimethylamine (TMA), dimethylamine (DMA), and ammonia in the fish muscle are also often used as the freshness index, but this method again is destructive and complicated.
