Effect of a Successive Feed of Melt Indole on the Production of L-Tryptophan by Tryptophan Synthase.

トリプトファンシンターゼを強化したEscherichia coli形質転換体によるインドール(lnd)とL-セリンからのL-トリプトファン(Trp)の合成経路を研究した-本酵素はIndとTrpにより著しい阻害を受ける。そこで,溶融Indを反応系に連続的に添加する方法を初めて構築した。本方法によるTrp生成能は,Indの一括添加法のそれを大きく上回り,高濃度Trpの生成が実証された。1L反応槽中での3時間の反応により,生成Trpの濃度92gL-1,菌体当たりのTrp生成量9.2g(g-cell)-1が達成された。菌体当たりのInd供給速度には上限があり,それを越えるとTrpの生成が途中で停止した。酵素とIndの反応により不活性な酵素に変化するモデルを取り込んだ速度式を考案し,上記のTrp合成反応の正確なシミュレーションが可能となった。
