Genetic diversity analysis of traditional and improved cultivars of Pakistani rice (Oryza sativa L.) using RAPD markers

The molecular marker is a useful tool for assessing genetic variations and resolving cultivar identities. Information on genetic diversity and relationships among rice genotypes from Pakistan is currently very limited. The objective of this study was to evaluate the genetic polymorphisms and identities of 10 traditional, 28 improved and 2 Japanese cultivars of rice using the random amplified polymorphic DNA technique. Twenty-five decamer-primers could generate a total of 208 RAPD fragments, of which 186 or 89.4% were polymorphic. The number of amplification products produced by each primer varied from 4 to 16 with an average of 8.3 bands primer -1 . The size of amplified fragments were ranged from 200 to 4000 bp. Pair-wise Nei and Lis similarity had estimated the range of 0.50 to 0.96 between rice cultivars. Based on analysis performed on a similarity matrix using UPGMA, 40 cultivars were grouped into 3 main clusters corresponding to aromatic, non-aromatic and japonica group. There were a few of independent cultivars. The cluster analysis had placed most of the aromatic cultivars into a close relation showing a high level of genetic relatedness. However, the clusters formed by the aromatic cultivars were distinct from those of non-aromatic and japonica types. Interestingly, a number of improved and traditional cultivars originating from diverse sources did not form well defined groups and were interspersed, indicating no association between the RAPD patterns and the geographic origin of the cultivars. The information generated from this study can be used to maximize selection of diverse parents and broaden the germplasm base in the future of rice breeding programs.

Determination of genetic variability of Asian rice ( Oryza sativa L. ) varieties using microsatellite markers

The microsatellite or simple sequence repeat (SSR) marker analysis was done to determine the allelic diversity and relationship among thirty five Asian cultivars of rice including 19 aromatic, 13 non-aromatic and 3 japonica type cultivars. A total of 144 alleles were detected at the 32 SSR loci, of which 141 (98%) were polymorphic. The number of alleles generated by each marker ranged from 2 to 13 with an anverage of 4.5 alleles per marker The size of smallest and largest allele ranged from 8(RM122) to as high as 71 (RM302). Polymorphism information content (PIC) values ranged between 0.157 (RM19, RM55) and 0.897 (RM70), with an average of 0.603 per marker. Basmati rice varieties amplified different alleles at 15 of the SSR loci than those in the japonica and/ or indica rice varieties. A number of SSRs were identified that could be utilized to differentiate between basmati and other non-basmati rice varieties. The RM252 and RM310 showed a clear differentiation of japonica cultivars from other ones. Pair-wise Nei and Lis similarity coefficients ranged from 0.19 to 0.90. The dendrogram based on the cluster analysis by microsatellite polymorphism, grouped 35 rice cultivars into two major groups effectively differentiating the tall, late maturing and slender aromatic cultivars from the short statured, early, short bold and long bold non-aromatic cultivars. These results could be useful for monitoring purity, genotype identification and for plant variety protection.

Morpho- biochemical evaluation of Brassica rapa sub-species for salt tolerance

Salt stress is one of the key abiotic stresses that affect both the qualitative and quantitative characters of many Brassica rapa sub-species by disturbing its normal morphobiochemical processes. Therefore, the present research work was designed to study the effect of different NaCl events (0, 50,100 and 150 mmol) on morphological and biochemical characters and to screen salt tolerant genotypes among brown, yellow and toria types of B. rapa sub-species. The plants were grown in test tubes with addition of four level of NaCl (0, 50,100 and 150 mmol). The effect of salinity on shoot and root length, shoot/ root fresh and dry weight, relative water content (RWC), proline and chlorophyll a, b, a+b contents was recorded after 4 weeks of sowing. The genotype 22861 (brown type) showed excellent morphological and biochemical performance at all stress levels followed by Toria-Sathi and Toria-A respectively as compared to Check variety TS-1. The genotype 26158 (yellow type) gave very poor performance and retard growth. The %RWC values and chlorophyll a, b and a+b contents were decreased several folds with the increase of salt concentration. While, the proline contents was increased with raising of salt stress. The brown and toria types showed maximum tolerance to salt stress at early germination stages as compare to yellows one. The present study will serve as model to develop quick salt tolerant genotypes among different plant sub-species against salt stress.
