Substrate specificity of human prolyl-4-hydroxylase

Proline analogs (3-F, 3-Cl, 3-Br, 3,3-cyclopropyl, 3,3-methylene, 3-Me, and 4-Me) were synthesized, incorporated into CbzGlyPheXGlyOEt, and tested as substrate analogues/mechanistic probes for the human prolyl-4-hydroxylase catalyzed hydroxylation reaction. With the exception of the 3-fluoroproline containing peptide, none of these peptides were substrates for the enzyme.

Selective Deprotection Of tert-Butyldimethylsilyl Ether With Lithium Bromide And 18-Crown-6

Abstract Lithium bromide, in the presence of a crown ether, has been found to selectively remove primary tert-butyldimethylsilyl ethers under controlled conditions. This selectivity has been utilized to synthesize 3′-TBDMS-2′-deoxyuridine in one step from the diprotected compound.

Cofactor biosynthesis : a mechanistic perspective

The chemistry of the cofactors has provided a fertile area of overlap between organic chemistry and biochemistry, and the organic chemistry of the cofactors is now a thoroughly studied area. In contrast, the chemistry of cofactor biosynthesis is still relatively underdeveloped. In this review the biosynthesis of nicotinamide adenine dinucleotide, riboflavin, folate, molyb-dopterin, thiamin, biotin, lipoic acid, pantothenic acid, coenzyme A, S-adenosylmethionine, pyridoxal phosphate, ubiquinone and menaquinone in E. coli will be described with a focus on unsolved mechanistic problems.
