Ecotoxicological impacts of clofibric acid and diclofenac in common carp (Cyprinus carpio) fingerlings: hematological, biochemical, ionoregulatory and enzymological responses.

Investigation on the toxic effects of pharmaceutical drugs namely clofibric acid (CA) and diclofenac (DCF) were studied in a common carp Cyprinus carpio at different concentrations such as 1, 10 and 100 μg L(-1) for a short-term period of 96 h under static bioassay method. At all concentrations, red blood cell (RBC), plasma sodium (Na(+)), potassium (K(+)), and glutamate oxaloacetate transaminase (GOT) levels were decreased in fish treated with CA and DCF. Contrastingly, white blood cell (WBC), plasma glucose, protein, lactate dehydrogenase (LDH) and gill Na(+)/K(+)-ATPase level were increased. However, a mixed trend was observed in hemoglobin (Hb), hematocrit (Hct), plasma chloride (Cl(-)), mean cellular volume (MCV), mean cellular hemoglobin (MCH), mean cellular hemoglobin concentration (MCHC) and glutamate pyruvate transaminase (GPT) levels. There was a significant (P<0.01 and P<0.05) change in all parameters measured in fish exposed to different concentrations of CA and DCF. In summary, the alterations in hematological, biochemical, ionoregulatory and enzymological parameters can be used as biomarkers in monitoring the toxicity of CA and DCF in aquatic environment. However, more detailed studies on using of specific biomarkers to monitor the human pharmaceuticals are needed.

Effects of Ibuprofen on hematological, biochemical and enzymological parameters of blood in an Indian major carp, Cirrhinus mrigala

In the present investigation, the most commonly used pharmaceutical drug Ibuprofen (IB) was exposed to an Indian major carp Cirrhinus mrigala under static bioassay method to estimate its toxicological effects for a period of 35 days. The median lethal concentration (LC 50) of IB to the fish C. mrigala for 24h was found to be 142 ppm. In sublethal treatment (1/10th of LC 50 24h value, 14.2 ppm), a significant decrease in erythrocyte (RBC), mean cellular hemoglobin concentration (MCHC) and plasma protein levels were observed throughout the study period when compare to that of their respective controls. In contrast, haemoglobin (Hb), hematocrit (Hct), mean cellular volume (MCV), mean cellular hemoglobin (MCH), leucocyte (WBC), plasma glucose and alanine transaminase (ALT) levels were increased in this study period. On the other hand, a mixed trend was noticed in aspartate aminotransaminase (AST) enzyme activity. Alterations of these parameters can be effectively used as potential biomarkers in monitoring of IB toxicity in the aquatic organisms. However, more detailed studies on these specific biomarkers are needed to assess the impacts of human pharmaceutical drugs in the field of pharmacotoxicology and aquatic toxicology.

Influence of cypermethrin toxicity on ionic regulation and gill Na+/K+-ATPase activity of a freshwater teleost fish Cyprinus carpio

The effects of acute and sublethal toxicity of cypermethrin, a synthetic pyrethroid insecticide on plasma electrolytes (Na(+), K(+) and Cl(-)) levels and gill Na(+)/K(+)-ATPase activity in a common carp Cyprinus carpio were examined. The 24h LC(50) value of cypermethrin for C. carpio was 1.86ppm. During acute exposure, cypermethrin caused adverse effects in the form of hyponatreima, hypokalemia and hypochloremia and inhibition of gill Na(+)/K(+)-ATPase activity. In sublethal exposure to cypermethrin (0.186ppm for 35 days), plasma Na(+) was decreased throughout the exposure period except 7th day whereas plasma K(+) level was increased up to 28th day, then declined. However, plasma Cl(-) level was decreased. Likewise, gill Na(+)/K(+)-ATPase activity was decreased as the exposure period extended. The present study indicates that cypermethrin was highly toxic to freshwater fish and ion levels in blood as measured by specific ion concentrations (Na(+), K(+) and Cl(-)) and changes in gill Na(+)/K(+)-ATPase activity may represent a sensitive and useful nonspecific biomarkers of chemical exposure and effects.

Toxicological effects of clofibric acid and diclofenac on plasma thyroid hormones of an Indian major carp, Cirrhinus mrigala during short and long-term exposures.

In the present investigation, the toxicity of most commonly detected pharmaceuticals in the aquatic environment namely clofibric acid (CA) and diclofenac (DCF) was investigated in an Indian major carp Cirrhinus mrigala. Fingerlings of C. mrigala were exposed to different concentrations (1, 10 and 100μgL(-1)) of CA and DCF for a period of 96h (short term) and 35 days (long term). The toxic effects of CA and DCF on thyroid hormones (THs) such as thyroid stimulating hormone (TSH), thyroxine (T4) and triiodothyronine (T3) levels were evaluated. During the short and long-term exposure period TSH level was found to be decreased at all concentrations of CA (except at the end of 14(th) day in 1 and 10μgL(-l) and 21(st) day in 1μgL(-l)) whereas in DCF exposed fish TSH level was found to be increased when compared to control groups. T4 level was found to be decreased at 1 and 100μgL(-l) of CA exposure at the end of 96h. However, T4 level was decreased at all concentrations of CA and DCF during long-term (35 days) exposure period. Fish exposed to all concentrations of CA and DCF had lower level of T3 in both the treatments. These results suggest that both CA and DCF drugs induced significant changes (P<0.01 and P<0.05) on thyroid hormonal levels of C. mrigala. The alterations of these hormonal levels can be used as potential biomarkers in monitoring of pharmaceutical drugs in aquatic organisms.

Impact of endosulfan on certain hematological and biochemical parameters of catfish Labeo fimbriatus: Sublethal study

Hematological and biochemical parameters are routinely used as indicators of the physiological or sublethal stress response to endogenous or exogenous changes in fish. In this work, the sublethal toxic effects of endosulfan (0.002 mg l-1) on certain hematological and biochemical parameters of Labeo fimbriatus were examined for 28 days and the analyses were made on 7, 14, 21, and 28 days. Endosulfan caused significant lower value of red blood corpuscles (RBC), hemoglobin, plasma glucose, and protein levels when compared to the control groups (p < 0.05). However, white blood corpuscles (WBC) increased throughout the study period. The results indicate that a low amount of endosulfan alters the hematological and biochemical parameters of fish, which can be useful in diagnosing the structural and functional status of fish exposed to toxicants.

Responses of the Indian major carp Labeo rohita to deltamethrin at acute and sublethal concentrations

The effects of deltamethrin at acute and sublethal concentrations on some hematological and biochemical profiles in the Indian major carp Labeo rohita have been examined. The lethal concentrations (LC50) of deltamethrin for 24 and 96 h were found to be 0.44 and 0.38 mg L−1, respectively. During acute treatment (24 h), hemoglobin (Hb), hematocrit (Hct), red blood cell (RBC), white blood cell (WBC), mean cellular volume (MCV), mean cellular hemoglobin (MCH), and mean cellular hemoglobin concentration (MCHC) values were significantly increased compared to control group. Plasma glucose, protein, cholesterol, albumin, globulin, triglyceride, and glycogen in gill and muscle were significantly decreased. Upon sublethal treatment (0.044 mg L−1, 10th of 24 h LC50), RBC and the biochemical parameters, except glucose and cholesterol, decreased significantly, while MCV, MCH, and MCHC values increased up to the 28th day and then declined. Cholesterol level was significantly increased throughout the study period. A biphasic response in the levels of Hb, Hct, WBC, and glucose were observed during sublethal treatment.

Establishment of Pre-Harvest Residue Limits (PHRLs) of Fluopicolide and Metrafenone in Cherry Tomato ( Lycopersicon esculentum Mill.)

BACKGROUND: The present investigation was aimed to predict the pre-harvest residue limits (PHRLs) of the fluopicolide and metrafenone on cherry tomato and to estimate their half-life and characteristics of the residues.METHODS AND RESULTS: Pesticides were treated once on cherry tomato in field 1 and 2 under the standard application rate. The samples were collected 7 times at the end of 0(2 hours after pesticides spaying), 1, 2, 3, 5, 7 and 10 days before harvest. Residues of fluopicolide and metrafenone were analyzed by the LC-MS/MS. In this study, the method limit of quantification (MLOQ) for both fluopicolide and metrafenone in cherry tomato was found to be 0.005 mg kg-1. Their recovery levels were 92.7∼94.8% and 82.6∼88.0%, shown with coefficient of variation of less than 10%. Half-life of fluopicolide and metrafenone in field 1 and 2 were found to be 15.0 days and 12.8 days, 18.9 days and 21.5 days, respectively.CONCLUSION: Based on the results, this study shows the level of PHRL on cherry tomato is 0.27 mg/kg for fluopicolide and 2.29 mg/kg for metrafenone at 10 days before harvesting. The present study indicates the residues of both pesticides on cherry tomato will be below maximum residue limit (MRL) at harvest.
