Ausgehend von der Beobachtung, dass 1-Methyl-2-benzyl-hydrazin tumorhemmend wirkt, wurden durch systematische Variationen die für diese Wirkung essentiellen Strukturmerkmale festgelegt. Aus einer grösseren Anzahl von Verbindungen wurden 1-Methyl-2-p-(isopropylcarbamoyl)benzyl-hydrazin-hydrochlorid und 1-Methyl-2-p-allophanoylbenzyl-hydrazin-hydrobromid für ausgedehnte experimentelle und klinische Versuche ausgewählt.

The identification, synthesis and activity of a novel class of piperidine renin inhibitors is presented. The most active compounds show activities in the picomolar range and are among the most potent renin inhibitors ever identified.

Piperidine renin inhibitors with heterocyclic core modifications or hydrophilic attachments show improved physical properties (lower lipophilicity, improved solubility). Tetrahydroquinoline derivative rac-30 with a molecular weight of 517 and a log D(pH 7.4) of 1.9 displays potent and long lasting blood pressure lowering effects after oral administration to sodium depleted conscious marmosets.

The synthesis of 3β-acetoxy-16β, 20β-dihydroxy-4′-methyl-18-nor-5β, 14β-pregnano[13,14-f]hexahydro-1′, 4′-oxazepin-3′-one (20a) is described.

Abstract Oxidation of secondary alcohols of different structures with chromic acid in aqueous acetic acid or chromic anhydride in acetic anhydride has been found to be accompanied by partial epimerization at the adjacent quaternary C-atom. In the case of compound 24, this epimerization amounts to 45%. Reaction conditions and the structure of the alcohol influencing the extent of epimerization have been investigated. With compound 10 it has been proved that epimerization occurs prior to oxidation to the ketone. A possible reaction mechanism is discussed.
