Isolation and identification of selenite reducing archaea from Tuz (salt) Lake In Turkey

In this study, Tuz lake brine samples were investigated for isolation and identification of selenite resistant halophilic prokaryotes. Among the 20 strains of extremely halophilic Bacteria and Archaea, a Gram negative rod designated as strain 106, showed high capacity in the resistance to selenite (25 mM) under aerobic conditions. Phenotypic characterizations and phylogenetic analyses based on 16S rDNA sequence comparison indicated that strain 106 was Halorubrum xinjiangense. The ability of strain 106 to deposite selenium‐containing particles were investigated by Transmission Electron Microscopy (TEM). Electron micrographs shows intact cells after selenite reduction and large amounts of selenium‐containing particles are present in the culture medium indicating that strain 106 is able to efficiently transport elemental selenium out of the cell.

Diversity of acidophilic prokaryotes at two acid mine drainage sites in Turkey

The biodiversity of acidophilic prokaryotes in two acidic (pH 2.8–3.05) mine drainage (AMD) sites (Balya and Çan) in Turkey was examined using a combined cultivation-based and cultivation-independent approach. The latter included analyzing microbial diversity using fluorescent in situ hybridization (FISH), terminal restriction enzyme fragment length polymorphism (`T-RFLP), and quantitative PCR (qPCR). Numbers of cultivatable heterotrophic acidophilic bacteria were over an order of magnitude greater than those of chemolithotrophic acidophiles in both AMD ponds examined. Isolates identified as strains of Acidithiobacillus ferrivorans, Acidiphilium organovorum, and Ferrimicrobium acidiphilum were isolated from the Balya AMD pond, and others identified as strains of Leptospirillum ferriphilum, Acidicapsa ligni, and Acidiphilium rubrum from Çan AMD. Other isolates were too distantly related (from analysis of their 16S rRNA genes) to be identified at the species level. Archaeal diversity in the two ponds appeared to be far more limited. T-RFLP and qPCR confirmed the presence of Ferroplasma-like prokaryotes, but no archaea were isolated from the two sites. qPCR generated semiquantitative data for genera of some of the iron-oxidizing acidophiles isolated and/or detected, suggesting the order of abundance was Leptospirillum > Ferroplasma > Acidithiobacillus (Balya AMD) and Ferroplasma > Leptospirillum > Acidithiobacillus (Çan AMD).

SİVAS’IN SOLAR TUZLALARINDAKİ MİKROBİYAL TOPLULUKLARIN KÜLTÜR-BAĞIMLI KARAKTERİZASYONU

Bu calismada Sivas’ta bulunan farkli solar tuzlalardan halofilik mikroorganizmalarin izolasyonu yapilmistir ve elde edilen izolatlarin 16S rRNA gen analizine dayali karakterizasyonu gerceklestirilmistir. Izolasyon islemi farkli tuz konsantrasyonlari ile hazirlanmis 5 farkli besiyeri ile gerceklestirilmistir. Morfolojik olarak farklilik gosteren kolonilerin 16S rRNA genleri ARDRA ile analiz edilmistir ve kesim profilleri karsilastirilmistir. 47 adet sus tanimlama amaciyla secilmistir. Bacteria domaini icerisinde yer alan izolatlarin Idiomarina , Salinivibrio , Halomonas , Salinibacter , Gracilibacillus , Thalassobacillus , Aliifodinibius , Chromohalobacter , Planococcus , Marinobacter , Bacillus , Aquisalimonas , Marinococcus , Alkalibacillus , Kangiella ve Microbulbifer cinsleri ile filogenetik olarak iliskili oldugu saptanmistir. Halofilik arke izolatlari ise Haloferax , Natrinema , Haloterrigena , Halopelagius , Halorhabdus ve Haloarcula cinsleri icerisinde dagilim gostermistir.

Determination Of Prokaryotic Diversity Of Salda Lake By Next Generation Sequencing Method

In our study, water samples were taken from Salda Lake and the pH and salinity data for the sample were determined. After that, samples were filtered and DNA extraction was performed. The obtained DNA was sequenced by the Illumina MiSeq sequencing system and data was analyzed by the QIIME program. When the data obtained after the analysis were evaluated, it was determined that the most dominant bacterial phyla are Proteobacteria and Actinobacteria. At class level, Gammaproteobacteria, Acidimicrobia and Actinobacteria were dominant. In order level Enterobacteriales, Acidimicrobiales, Actinobacteriales were abundant. When the Archaea domain was evaluated it was determined that the Euryarchaeota phylum was dominant. Halobacteria members were abundant at the class level. In addition Halobacteria class, Halobacteriales order, Halobacteriaceae family includes Haloarcula, Halobacterium, Halonotius, Halorhabdus, Halorubrum and Haloplanus genus members.

ATIK SUDAN İZOLE EDİLEN FENOL DEGRADE EDİCİ BAKTERİLERİN ENZİMATİK VE ANTİBİYOTİK DUYARLILIK AKTİVİTELERİ

Calisma kapsaminda iki farkli atik su aritma tesisinin cesitli unitelerinden ornek alinmis ve bu orneklerin pH ve element icerikleri belirlenmistir. Ozellikleri belirlenen orneklerden fenollu besiyerine inokulasyon yapilmis ve 19 izolat elde edilmistir. Bu izole edilen 19 izolattan morfolojik olarak farkli oldugu dusunulen 9 izolatin tanimlamasi yapilmistir. Izolatlar arasinda yogun olarak Pseudomonas turleri belirlenmistir. Tanimlanan bu 9 izolatin atik sulardaki aritim potansiyellerini degerlendirebilmek icin antibiyotik duyarlilik testi gerceklestirilmistir. Ayrica izolatlarin amilaz ve proteaz enzim uretim aktiviteleri degerlendirilmistir. Izolatlarin ofloksasin, azitromisin ve streptomisine duyarli olduklari belirlenmistir. Enzim uretim yetenekleri degerlendirildiginde ise yalnizca 2 izolat zayif proteaz aktivitesi gosterirken izolatlarin cogu amilaz aktivitesi gostermistir.
