Tail docking in pigs: acute physiological and behavioural responses

Tail docking of piglets is a routine procedure on farms to control tail-biting behaviour; however, docking can cause an acute stress response. The objectives of this research were to determine the stress responses to tail docking in piglets and to compare two methods of tail docking; cautery iron (CAUT) and the more commonly used blunt trauma cutters (BT). At approximately 6 days of age, piglets were tail docked using CAUT (n = 20), BT (n = 20) or sham tail docked with their tails remaining intact (CON; n = 40). Blood samples were taken prior to tail docking and at 30, 60 and 90 min after tail docking to evaluate the effect of tail docking on white blood cell (WBC) measures and cortisol concentrations. The above experiment was repeated to observe behaviour without the periodic blood sampling, so as not to confound the effects of blood sampling on piglet behaviour. Piglet behaviour was recorded in the farrowing crate using 1 min scan-samples via live observations for 60 min prior to and 90 min after tail docking. Total WBC counts were reduced (P > 0.05) among BT and CAUT compared with CON piglets 30 min after tail docking. Cortisol concentrations were higher (P < 0.01) among BT compared with CON and CAUT piglets 60 min after tail docking. Cautery and BT-docked piglets spent more (P < 0.05) time posterior scooting compared with CON piglets between 0 and 15 min, and 31 and 45 min after tail docking. Piglets tail docked using CAUT and BT tended to spend more (P < 0.07) time sitting than CON piglets between 0 and 15 min post tail docking. Elevated blood cortisol can be reduced by the use of the CAUT rather than the BT method of tail docking. Although the tail docking-induced rise in cortisol was prevented by using CAUT, the behavioural response to BT and CAUT docking methods was similar.

Effects of variations in the environment, length of journey and type of trailer on the mortality and morbidity of pigs being transported to slaughter

The month, average air temperature, relative humidity, number of pigs of each sex per trailer load, journey time, type of trailer and bedding, and waiting time at the processing plant were recorded from 16,323 trailers transporting 2,730,754 pigs to a packing plant in the USA during a year, together with the numbers of pigs that were dead on arrival, injured or unable to walk. The percentage of pigs dead on arrival increased at temperatures above 20°C, and the percentage of pigs unable to walk increased at temperatures of 5°C or below. The journey time and waiting time at the processing plant influenced the percentages of dead and injured pigs and pigs unable to walk.

Cortisol responses to dehorning of calves given a 5-h local anaesthetic regimen plus phenylbutazone, ketoprofen, or adrenocorticotropic hormone prior to dehorning

The purpose of this work was to assess whether the non-steroidal anti-inflammatory drugs (NSAIDs) phenylbutazone and ketoprofen, and an adrenocorticotropic hormone (ACTH) induced cortisol surge, reduce the cortisol response which occurs when the local anaesthetic wears off in calves following dehorning. There were four control groups and one dehorned group; also four groups were given local anaesthetic lasting 5h and were dehorned without or with phenylbutazone, ketoprofen or an ACTH injection, one group was injected with ACTH twice (at 0 and 6h) and another received ACTH and 6h later was dehorned. Blood samples were taken before and after dehorning and plasma cortisol concentrations were determined by radio-immunoassay. Dehorning increased the mean plasma cortisol concentrations [max 137 (11)nmoll(-1)] above control values [38 (5)nmoll(-1)] for about 7h, whereas local anaesthesia maintained concentrations at control values until about 5h after dehorning, and then they became elevated until about 10h. The maximum rise in mean concentration which occurred when the local anaesthetic wore off [128 (32)nmoll(-1)] was not affected when phenylbutazone was given before dehorning [141 (28)nmoll(-1)], but was reduced significantly when ketoprofen [65 (17)nmoll(-1)] or ACTH [61 (19)nmoll(-1)] were injected before or at the time of dehorning, respectively. Marked cortisol responses to ACTH injected at 0 and 6h were similar, but the early part of the cortisol response to dehorning 6h after an ACTH injection was reduced. It is suggested that the delayed cortisol response, which began 5h after dehorning, arose both from ketoprofen-sensitive and cortisol-sensitive sensory input as well as from other factors. Phenylbutazone did not affect the sensory input from the amputation wounds in the present calves.

The effect of temperament and responsiveness towards humans on the behavior, physiology and milk production of multi-parous dairy cows in a familiar and novel milking environment.

The objectives of this study were to investigate whether; 1) temperament or 2) behavioral responsiveness to humans, can affect the behavior, physiology and productivity of dairy cows being milked in a familiar and novel milking environment. Temperament of multi-parous cows was defined based on exit time from a restraint device, as High Responders (HR; n=10), Medium Responders (MR; n=10) or Low Responders (LR; n=10). The behavioral response of cows to humans was assessed using four tests: restraint, exit speed, avoidance distance test and a voluntary approach test. Cows were milked according to their established routines in a rotary (familiar) milking parlor and behavioral, physiological and production data were collected over five consecutive days, including heart rate, cortisol and oxytocin concentrations and milk yield. The following week, cows were milked in a novel environment (herringbone parlor within the same farm facility) over five consecutive days, and the data and sample collection program was repeated. Cows were then given an exogenous adrenocorticotropic hormone (ACTH) challenge to measure adrenal responsiveness. Exit time was negatively correlated with the behavioral responses of cows to restraint and human avoidance distance (HAD) in the paddock and arena. The behavioral response of cows to the milking process was greater in MR than LR and HR cows in the familiar and novel milking environments. Milk yields were greater in LR than HR cows in the novel but not the familiar milking parlor. Oxytocin concentrations increased during milking in the novel environment, regardless of cow temperament. In the familiar and novel environments, heart rates were higher in HR than LR cows before and during milking and rMSSD was lower in HR cows during milking in a novel environment. There was no difference in cortisol concentrations between LR and HR cows in response to an ACTH challenge, but HR cows had higher baseline cortisol levels than LR cows. The number of leg lifts cows performed when restrained in the crush was associated with several physiological and milk production measures. These results indicate that cows with different temperaments vary in their basal physiology as well as their behavioral and physiological responses to milking and stress associated with being milked in a novel environment.

The physiological and behavioral response of pigs castrated with and without anesthesia or analgesia

Surgical castration is a common management practice performed on male pigs to prevent the occurrence of boar taint. Surgical castration is known to cause physiological and behavioral changes in pigs indicative of pain-induced distress; however, it is commonly performed without pain relief. The objective of this study was to evaluate the effectiveness of carbon dioxide gas (CO(2)) anesthesia and a non-steroidal anti-inflammatory drug (NSAID) to alleviate the pain caused by castration. At 3 d of age, male pigs were either control handled (CON), castrated without pain relief (CAS), given an NSAID and then immediately castrated (CAS+NSAID), anesthetized with CO(2) and then castrated (CAS+CO2), or anesthetized with CO(2) and given an NSAID at the time of castration (CAS+BOTH). Blood samples were collected before castration, and at 30, 60, 120, and 180 min, 24 h, and 3 d after castration or handling for analysis of cortisol, C-Reactive protein (CRP), and substance-P (SP) concentrations. This study was then repeated using the same treatment groups, and the behavioral response to castration and handling were measured using a 1-min scan sampling procedure. The percentage of stress vocalizations was recorded during the administration of all treatments. Anesthesia and analgesia did not effectively reduce (P > 0.05) the cortisol response to surgical castration. Overall, CRP concentrations were greater (P < 0.05) in CAS+CO2 pigs as compared with CON pigs. Sixty minutes after castration or handling, SP concentrations were greater (P < 0.08) in pigs given CO(2) anesthesia (CO2, CAS+CO2, and CAS+BOTH) than CON, CAS, and CAS+NSAID pigs. Pigs castrated without pain relief spent more (P < 0.001) time lying without contact than all other treatments during the first 30 min after castration, but thereafter CAS+CO2 pigs spent more (P < 0.001) time lying without contact than other treatments. During the first 30 min after the treatments were applied, CAS+CO2 pigs spent more (P < 0.01) time displaying pain-like behaviors than CON, CAS, CAS+NSAID, and CAS+BOTH pigs. The percentage of stress vocalizations was greater (P < 0.05) in CAS and CAS+NSAID pigs than all other treatments. Neither CO(2) anesthesia nor a NSAID, given separately or combined, markedly reduced the pain-induced distress caused by castration in pigs. More research is needed to evaluate practical methods of on-farm pain relief for pigs.

Physiology and behavior of pigs before and after castration: effects of two topical anesthetics

Surgical castration of male piglets is a common management practice conducted on commercial swine farms to prevent the occurrence of boar taint and aggressive behavior. However, the procedure of surgical castration causes acute pain-induced distress, which is an animal welfare concern. The objective of this study was to evaluate the use of two topical anesthetics to alleviate the pain caused by castration in piglets as measured by physiological and behavior indices of stress. At 3 days of age, 40 weight-matched piglets were allocated to one of four treatment groups. Treatments included: (i) sham castration (CON), (ii) surgical castration (CAS), (iii) castration and short-acting local anesthetic applied topically to the castration wound (SHORT) and (iv) castration and long-acting local anesthetic applied topically to the castration wound (LONG). Blood samples were collected from piglets before and 30, 60, 120 and 180 min after castration to measure leukocyte and differential counts and cortisol concentrations. The above experiment was repeated without blood collection and behavior was recorded for 30 min before and 180 min after castration or handling. Stress vocalizations were recorded during castration and handling. All piglets were weighed before and 24 h after castration and wound healing was recorded daily for the first 14 days after castration. Leukocyte counts and differentials did not differ (P > 0.05) among any of the treatments. Cortisol concentrations were elevated (P < 0.06) in CAS, SHORT and LONG piglets compared with controls 30 and 60 min after castration. The percentage of stress vocalizations was greater (P < 0.05) among castrated piglets compared with CON piglets, regardless of anesthetic treatment. Piglets that were castrated and not given a topical anesthetic spent more time (P < 0.05) lying without contact compared with piglets castrated and given a topical anesthetic, regardless of the topical anesthetic administered. Body weight change did not differ (P > 0.05) among treatments 24 h after castration or control handling and wound healing scores were greater (P < 0.05) in SHORT compared with CAS and LONG piglets 9 to 14 days after castration. In this study, the use of a short- or long-acting topical anesthetic was not effective in reducing the pain-induced distress caused by castration in piglets. Further research is needed to evaluate alternative practical methods to reduce the pain caused by the on-farm castration of piglets.

Administration of anesthetic and analgesic prevent the suppression of many leukocyte responses following surgical castration and physical dehorning.

The objectives of the current research were to determine the physiological effects and responses of many leukocytes following surgical castration and/(or) physical dehorning and the influence of anesthetics and analgesics in 3-month-old calves. Eighty 3-month-old Holstein bull calves were completely randomized to treatments in a 2 × 2 × 2 factorial arrangement with castration, dehorning, and anesthetic/analgesic as the main effects. Peripheral blood samples were collected just before (0) and 0.5, 1.5, 2.5, 4, 6, 24, and 72 h after the respective procedure(s) and analyzed for total leukocyte and differential counts, as well as plasma cortisol and haptoglobin concentrations. Blood from the 0, 0.5 and 24h collections were analyzed for many ex vivo leukocyte responses. Data were analyzed using a repeated measures analysis of variance with the fixed effects of treatment, time, and the interaction of treatment × time. Pre-planned contrasts were performed to determine the effect of (1) management procedure (castration and/(or) dehorning), (2) anesthetic/analgesic, and (3) were the management procedures additive. There were treatment × time interactions (P<0.05) on plasma cortisol and haptoglobin concentrations as well as for total leukocyte and neutrophil concentrations in blood. Castration and dehorning increased cortisol concentrations and the effect of the procedures was additive (P<0.02). Dehorning alone elicited a greater (P<0.05) cortisol response than castration alone. In contrast, the leukocytosis and neutrophilia was greater (P<0.01) among castrated calves. In addition, haptoglobin concentrations at 24h after castration were elevated (P<0.01) in calves that were castrated. Both castration and dehorning suppressed (P=0.04) many leukocyte responses including the secretion of tumor necrosis factor-α when whole blood cultures were stimulated with lipopolysaccharide, surface expression of L-selectin on peripheral blood neutrophils, and the oxidative burst intensity of peripheral blood neutrophils when co-cultured with an Escherichia coli. The effects of castration and dehorning on blood leukocyte counts or any of the leukocyte responses were not additive (P>0.23). Castration and dehorning effects of plasma haptoglobin concentrations tended (P=0.10) to be additive at 72 h after the procedure(s). Prior administration of local anesthetic and a systemic analgesic attenuated (P<0.001) the cortisol response and prevented (P=0.03) the observed leukocytosis, neutrophilia, and leukocyte suppression. These data suggest that calves should be castrated and dehorned on the same day rather than spreading them out across two days and calves should be administered pain relief prior to performing either procedure.

Effect of castration and dehorning singularly or combined on the behavior and physiology of Holstein calves

The objectives of this study were to determine i) the effect of castration, dehorning, or both on the physiology and behavior of 3-mo-old Holstein calves, and ii) the effectiveness of pain relief to alleviate the pain caused by castration and/or dehorning. Holstein calves (n = 80) were assigned randomly to 1 of 8 treatments (10 calves/treatment): i) control handling (SHAM); ii) surgical castration (CAS); iii) dehorning (DH); iv) surgical castration and dehorning (CD); v) control handling plus pain relief (ANA); vi) surgical castration plus pain relief (CAS+A); vii) dehorning plus pain relief (DH+A); or viii) surgical castration and dehorning plus pain relief (CD+A). Pain relief consisted of administering local anesthetic and a nonsteroidal anti-inflammatory drug (NSAID) immediately before castration, dehorning, or both. Sequential blood samples were collected to measure leukocyte counts and cortisol concentrations. Behavior was recorded using 5-min scan samples during the first 3 h after application of the treatments. Calves were weighed before and 24 h after treatment application. Calves dehorned spent more time head shaking (p < 0.001) and ear flicking (p < 0.05), and CD calves spent more time ear flicking (p < 0.05) and foot stamping (p < 0.01) than SHAM handled calves. Calves castrated, dehorned, or both spent less (p < 0.01) time eating compared with sham handled calves. Giving calves pain relief before castration and/or dehorning increased (p < 0.05) the time spent eating compared with CAS, DH, and CD calves. At 6 h posttreatment, neutrophil to lymphocyte ratio was greater (p < 0.01) in castrated and/or dehorned calves compared with SHAM-handled calves. Castration and/or dehorning also increased (p < 0.05) cortisol concentrations for at least 4 h after these procedures were performed; however, administering pain relief before castration and/or dehorning markedly reduced (p < 0.05) this response. Behavioral and physiological changes caused by castration, dehorning, or both are indicative of calves experiencing pain for at least 4 h after application of these procedures, and these responses were additive when performed together. Therefore, providing calves with pain relief, in the form of local anesthetic and an NSAID, can markedly reduce both the behavioral and physiological response to these procedures.

Space requirements of weaned pigs during a sixty-minute transport in summer

Currently, there are no trucking quality assurance recommendations for space allowance of weaned pigs during transport in the United States. The objective of this research was to establish a first estimate of the space requirements of weaned pigs during transport in summer based on measures of animal well-being. A commercial semi-trailer was fitted with compartments that provided 0.05, 0.06, and 0.07 m(2)/pig, which were replicated on the upper and lower deck, with a constant 100 pigs per compartment. Cameras were placed in each experimental compartment to record behaviors and postures of pigs during transport. The frequencies of standing, lying, sitting, standing/rearing on another pig, and lying/huddling on top of another pig were recorded using 1-min scan samples during the entire duration of transport. Blood samples were collected and BW and lesion scores recorded from 32 pigs per space allowance for physiological and immune measures before and after transport (n = 32 pigs/treatment). Pigs were transported for 60 +/- 5 min to the wean-to-finishing site using the same route for each replicate during summer (temperature: 28.4 +/- 1.2 degrees C and relative humidity: 59.8 +/- 4.4% within the trailer). Data were analyzed using the MIXED procedure (SAS Institute Inc., Cary, NC). Cortisol, hematocrit, blood urea nitrogen, total protein, albumin, aspartate aminotransferase, creatine kinase, and gamma-glutamyl transferase increased (P < 0.05) after transport regardless of space allowance. Plasma glucose and BW decreased (P < 0.05) after transport regardless of space allowance. Lesion scores increased (P < 0.001) after transport and were greater (P < 0.05) for barrows compared with gilts. The neutrophil to lymphocyte ratio was greater (P < 0.005) for pigs transported at 0.05 m(2)/pig compared with pigs transported at 0.06 and 0.07 m(2)/pig. Pigs transported at 0.05 m(2)/pig lay down less (P < 0.05) than pigs transported at 0.06 and 0.07 m(2)/pig between 30 and 60 min of transport. Greater neutrophil to lymphocyte ratio and less lying behavior performed by pigs transported at 0.05 m(2)/pig suggest that a minimum space allowance of 0.06 m(2)/pig was preferable when transporting weaned pigs for 60 min during summer in this study.

The effect of training first-lactation heifers to the milking parlor on the behavioral reactivity to humans and the physiological and behavioral responses to milking and productivity

The objectives of this study were (1) to determine whether training heifers before calving would modulate the behavioral reactivity to humans and the behavioral and physiological responses to milking routines after calving, and (2) whether heifer temperament would affect this response. The behavioral reactivity of heifers to humans was assessed prepartum using 4 behavioral tests (restraint, exit speed, avoidance distance in the paddock and arena, and a voluntary approach test). These behavioral tests were repeated immediately after and 12 wk after training. Temperament was defined based on exit time from a restraint device, and heifers were denoted as either low (LR; n=20) or high (HR; n=20) responders. Two weeks before calving, half the LR and HR heifers were selected randomly for training to the milking parlor, whereas the other heifers were left undisturbed in the paddock. Training consisted of 4 sessions conducted over 2 consecutive days and involved introducing the heifers to the milking platform, associated noises, and human contact. During the first 5d of lactation, behavioral and physiological data were collected from all heifers, including behavior during cup attachment, plasma cortisol concentrations, and residual milk volumes. Milk production data were collected over the entire lactation period. Training reduced the avoidance distance of LR but not HR heifers, and trained HR heifers stepped more than trained LR heifers during a restraint test. The behavioral response of heifers to the restraint test was positively correlated with milk yield, milking duration, and residual milk volume. Trained LR heifers flinched, stepped, and kicked more during attachment of milking clusters than did untrained LR heifers. During the first week of lactation, total milk yield was lower in trained than untrained HR heifers, and maximum flow rates were higher and residual milk volumes were lower in trained than untrained heifers, irrespective of heifer temperament. Plasma cortisol concentrations were lower on d 5 of lactation than on d 1 and were higher in HR heifers after milking than before milking. Over the first 8 mo of lactation, milking duration was lower and average flow rates were higher in trained than untrained heifers. These results suggest that trained heifers may have experienced less distress during the first week of lactation, but the effect of training on the behavioral and physiological responses to milking appeared to be influenced by heifer temperament.
