Seroprevalence of Toxoplasma gondii and Neospora caninum infections in goats in Poland

In 2007 a survey on herd-level seroprevalence of Toxoplasma gondii and Neospora caninum infections in goats in Poland was carried out. Sera were collected from all 49 breeding goat herds, scattered over the entire country, with the vast majority of them located in the western, central and northern provinces. Only adult females (≥12 months of age) were included in the study. A herd was recorded as infected if at least one seropositive female was detected. In each herd, simple random sampling was applied and sample size was determined in a way, which allowed to evaluate serological status of a herd at expected individual-level seroprevalence 10% and level of confidence 95%. In total, 1060 sera were tested using two commercial indirect ELISA kits. Sera positive to N. caninum were subsequently confirmed with IFAT. The true herd-level seroprevalence was 100% for T. gondii and 9.0% for N. caninum infection. Three herds positive to T. gondii infection were randomly selected and all adult goats were tested with an ELISA. Individual-level true seroprevalence in these herds ranged from 30.2% to 100%. This is the first time that antibodies to N. caninum have been detected in goats in Poland.

Schmallenberg virus antibodies detected in Poland.

Between 24 and 30 July 2012 230 adult goats from three western provinces of Poland bordering on Germany (Western Pomerania, Lubuskie and Lower Silesia) were blood-sampled and tested for antibodies to Schmallenberg virus (SBV) using indirect immunoenzymatic test (ID Screen® Schmallenberg virus indirect, IDvet Innovative Diagnostics). The ELISA test identified 21 seropositive goats - 15 in Western Pomerania (16% of all goats tested in this province), five in Lubuskie (6%) and one in Lower Silesia (2%). Our study demonstrates for the first time the presence of antibodies to SBV in Poland.

Seroprevalence of Toxoplasma gondii in wild boars, red deer and roe deer in Poland

Little is known about the prevalence of Toxoplasma gondii in wild life, particularly game animals in Poland. Meat juice collected during the 2009/2010 and 2010/2011 hunting seasons from 552 red deer (Cervus elaphus), 367 wild boars (Sus scrofa) and 92 roe deer (Capreolus capreolus) was tested for T. gondii antibodies using the multi-species ID Screen Toxoplasmosis Indirect kit (IDvet, Montpellier, France). Antibodies to T. gondii were detected in 24.1% of red deer (95% CI: 20.7%, 27.8%), 37.6% of wild boar (95% CI: 32.8%, 42.7%) and 30.4% of roe deer (95% CI: 22.0%, 40.5%). To the authors’ best knowledge, this is the first epidemiological report of T. gondii prevalence in red deer, roe deer and wild boars in Poland. T. gondii is present in wildlife animal tissues and consumption of the game may be a potential source of infection for humans.

Risk factors associated with seropositivity to small ruminant lentiviruses in goat herds

Cross-sectional studies based on serological testing and questionnaires were conducted at 5-yr intervals (1996, 2002 and 2007) in goat breeding herds from Poland to determine true herd-level seroprevalence of caprine arthritis-encephalitis and the herd-level risk factors for seropositivity. Multivariable logistic regression models were developed for data from 1996 and the combined data from 2002 and 2007, separately. True herd-level seroprevalences in 1996, 2002 and 2007 were 30.8% (CI 95%: 20.5-41.0%), 65.7% and 71.9%, respectively. The import of goats from abroad was a risk factor only in 1996 (OR 13.6, CI 95%: 1.14-162). The presence of seropositive bucks in a herd was a risk factor in 1996 (OR 21, CI 95%: 1.89-233) and in 2002-2007 (OR 2.9, CI 95%: 1.04-8.4). Moreover, large herds (>30 does in 1996 or >100 does in 2002-2007) were more likely to be seropositive than smaller herds (OR=10.1, CI 95%: 2.17-46 in 1996 and OR 5.4, CI 95%: 1.11-26 in 2002-2007).

Serological evidence for BVDV-1 infection in goats in Poland — Short communication

A serological survey was conducted in 2007 in the breeding goat population in Poland to gain insights into the epidemiology of pestivirus infection. All breeding herds were included in the study and representative serum samples were taken in each herd to evaluate herd-level seroprevalence at 10% expected individual-level prevalence and 95% level of confidence. Altogether 1060 serum samples from 49 herds were tested with blocking ELISA and then the positive and inconclusive results were confirmed in a serum neutralisation test, which also allowed us to determine the pestivirus species responsible for seroconversion. Herd-level seroprevalence proved to be 10.2% and bovine viral diarrhoea virus type 1 (BVDV-1) was responsible for the seroconversion in seven out of eight cases. In the remaining serum sample the causative virus could not be identified due to a pronounced cross-neutralising activity possibly derived from multiple infections. This is the first report on the diagnosis of BVDV-1 infection in Polish goats.

Detection of porcine reproductive and respiratory syndrome virus (PRRSV) and influenza A virus (IAV) in oral fluid of pigs

Recently oral fluid has become a novel sample type for pathogen nucleic acid and antibody detection, as it is easy to obtain with non-invasive procedures. The objective of the study was to analyze porcine reproductive and respiratory syndrome virus (PRRSV) and influenza A virus (IAV) circulation in growing pigs from three Polish production farms, using Real Time PCR and ELISA testing of oral fluid and serum. Oral fluids were collected every 2weeks, in the same 3-4 pens of pigs aged between 5 and 17weeks. Additionally, blood samples were collected every 4weeks from 4 pigs corresponding to the same pens as oral fluid and tested for the presence of PRRSV nucleic acid (pooled by 4) and antibodies. In farm A no PRRSV circulation was detected and only maternal antibodies were present. In farm B and farm C antibodies to PRRSV in serum and oral fluid were detected in most samples. In farm B PRRSV Type 1 was detected in 80.9% of oral fluid samples and in 58.3% of serum pools, and in farm C in 92.8% of oral fluid samples and 75% serum pools. Striking differences were observed between different pens in PRRSV detection patterns. In farms B and C ORF5 sequence analysis showed the presence of wild type strains which were about 84-85% identical to the modified live vaccine used. In all three farms two waves of IAV shedding with oral fluid were detected, in weaners and fatteners.

Leptospiral antibodies in the breeding goat population of Poland

In 2007, a survey was conducted on the prevalence of antibodies to 19 Leptospira serovars in goats in Poland . Sera were collected from adult females of all 49 breeding goat herds in Poland by applying simple random sampling. In total, 736 sera were tested by the microscopic agglutination test. A herd was considered seropositive if at least one goat with a titre of 100 or more was detected. Herd-level seroprevalence of Leptospira was 89.8 per cent and individual-level seroprevalence varied from less than 1.0 to 85.0 per cent among the herds. Antibodies to Leptospira serovars Zanoni, Bratislava, Autumnalis, Australis and Javanica were most frequently detected. Although 40.3 per cent of seropositive goats had high antibody titres (≥400), suggesting recent infection, no relationship with abortions or other clinical manifestation of leptospirosis in goats was detected.

Zolpidem poisoning in a cat

Zolpidem (Stilnox) is a non-benzodiazepine hypnotic drug of the imidazopyridine class intended for treatment of insomnia in humans. A 16-year-old neutered cat, weighing 3.8 kg, was presented with sudden onset of stupor, disorientation, severe ataxia, vomiting and hypersalivation. Symptomatic treatment was given when ingestion of 1.25 mg/kg zolpidem (half of a 10-mg tablet) was confirmed, because no information on the efficacy and safety of the use of flumazenil in the treatment of zolpidem poisoning in cats has been published to date. As zolpidem is prescribed with increasing frequency in humans, the occurrence of accidental poisonings of pets is likely to increase.

Multidrug resistance in Escherichia coli strains isolated from infections in dogs and cats in Poland (2007-2013).

The antimicrobial susceptibility of Escherichia coli isolates associated with various types of infections in dogs and cats was determined. The studied isolates were most frequently susceptible to fluoroquinolones and the extended-spectrum cephalosporins (ESCs), antimicrobials commonly used in treatment of infections in companion animals. However, an increase in the percentage of strains resistant to β-lactam antibiotics including ESCs was noted between January 2007 and December 2013. The frequency of multidrug-resistant (MDR) E. coli isolation (66.8% of isolates) is alarming. Moreover, the statistically significant increase of the percentage of MDR isolates was observed during the study period. No difference in the prevalence of multidrug resistance was found between bacteria causing intestinal and extraintestinal infections and between canine and feline isolates. Nonhemolytic E. coli isolates were MDR more often than hemolytic ones. Our study showed the companion animals in Poland as an important reservoir of MDR bacteria. These results indicate that continuous monitoring of canine and feline E. coli antimicrobial susceptibility is required. Furthermore, introduction and application of recommendations for appropriate use of antimicrobials in small animal practice should be essential to minimize the emergence of multidrug resistance among E. coli in companion animals.

Influence of small ruminant lentivirus infection on cheese yield in goats

Three-year cohort study was carried out to investigate the influence of small ruminant lentivirus (SRLV) infection on cheese yield in goats. For this purpose records of milk yield, milk composition and cheese yield were collected in a dairy goat herd. Cheese yield was recorded as the amount of fresh cheese obtained from 1 kg milk. All goats were serologically tested for SRLV infection twice a year. The analysis included 247 records in total (71 for seropositive and 176 from seronegative individuals) and was carried out with the use of the four-level hierarchical linear model (α = 0·05). SRLV infection proved to be a statistically significant independent factor reducing cheese yield (P = 0·013)--when other covariates were held constant cheese yield was reduced by 4·6 g per each 1 kg milk in an infected goat compared with an uninfected goat. Other statistically significant covariates positively associated with cheese yield were protein contents, fat contents and the 3rd stage of lactation (P < 0·001 for all).
