Prospective study on quantitative and qualitative antimicrobial and anti-inflammatory drug use in white veal calves

OBJECTIVES To document and quantify drug use in white veal calves, an intensive livestock production system where multidrug resistance is abundantly present. METHODS Drug consumption data were prospectively collected on 15 white veal production cohorts (n = 5853 calves) in Belgium (2007-09). Treatment incidences (TIs) based on animal defined daily dose (ADD), prescribed daily dose (PDD) and used daily dose (UDD) were calculated. Risk factors were identified by linear regression. RESULTS The average TI(ADD) of antimicrobial treatments was 416.8 ADD per 1000 animals at risk. Predominantly, oral group antimicrobial treatments were used (95.8%). Of the oral group antimicrobial treatments, 12% and 88% were used for prophylactic or metaphylactic indications, respectively. The main indication for group and individual drug use was respiratory disease. The most frequently used antimicrobials (group treatments) were oxytetracycline (23.7%), amoxicillin (18.5%), tylosin (17.2%) and colistin (15.2%). Deviations from the leaflet dosage recommendations were frequently encountered, with 43.7% of the group treatments underdosed (often oxytetracycline and tylosin to treat dysbacteriosis). In 33.3% of the oral antimicrobial group treatments a combination of two antimicrobial preparations was used. Smaller integrations used more antimicrobials in group treatments than larger ones (P < 0.05); an integration is defined as a company that combines all steps of the production chain by having its own feed plant and slaughterhouse and by placing its calves in veal herds owned by producers that fatten these calves for this integration on contract. Producers used higher dosages than prescribed by the veterinarian in cohorts with a single caretaker (P < 0.01). CONCLUSIONS The present study provided detailed information on the intensive antimicrobial use in the white veal industry. Reduction can only be achieved by reducing the number of oral group treatments.

Longitudinal study on morbidity and mortality in white veal calves in Belgium

BackgroundMortality and morbidity are hardly documented in the white veal industry, despite high levels of antimicrobial drug use and resistance. The objective of the present study was to determine the causes and epidemiology of morbidity and mortality in dairy, beef and crossbred white veal production. A total of 5853 calves, housed in 15 production cohorts, were followed during one production cycle. Causes of mortality were determined by necropsy. Morbidity was daily recorded by the producers.ResultsThe total mortality risk was 5,3% and was significantly higher in beef veal production compared to dairy or crossbreds. The main causes of mortality were pneumonia (1.3% of the calves at risk), ruminal disorders (0.7%), idiopathic peritonitis (0.5%), enterotoxaemia (0.5%) and enteritis (0.4%). Belgian Blue beef calves were more likely to die from pneumonia, enterotoxaemia and arthritis. Detection of bovine viral diarrhea virus at necropsy was associated with chronic pneumonia and pleuritis. Of the calves, 25.4% was treated individually and the morbidity rate was 1.66 cases per 1000 calf days at risk. The incidence rate of respiratory disease, diarrhea, arthritis and otitis was 0.95, 0.30, 0.11 and 0.07 cases per 1000 calf days at risk respectively. Morbidity peaked in the first three weeks after arrival and gradually declined towards the end of the production cycle.ConclusionsThe present study provided insights into the causes and epidemiology of morbidity and mortality in white veal calves in Belgium, housed in the most frequent housing system in Europe. The necropsy findings, identified risk periods and differences between production systems can guide both veterinarians and producers towards the most profitable and ethical preventive and therapeutic protocols.

Natural antibodies related to metabolic and mammary health in dairy cows

Natural antibodies (NAb) are defined as antibodies that circulate in normal healthy individuals under the absence of deliberate antigenic stimulation. Two types of NAb are distinguished: NAb towards exogenous antigens and NAb towards autoantigens (N(A)Ab). The objectives of the current study were threefold. First, we studied the relation between metabolic health and concentrations of NAb binding keyhole limpet hemocyanin (KLH) or lipopolysaccharide (LPS) in milk and plasma of dairy cows in early lactation. Second, we determined the presence of N(A)Ab binding transferrin, myosin and thyroglobulin in bovine milk. Third, we studied the relation between N(A)Ab in bovine milk and mammary health. For the first objective, dairy cows were either fed a control (C) (n=8) or a diet where 2 kg of concentrates were replaced by an iso-energetic concentrate containing marine algae (ALG) from week -3 till 8 postpartum (experiment 1). Plasma and milk samples were analyzed weekly for NAb binding either KLH or LPS. Plasma was analyzed for glucose, non-esterified fatty acids (NEFA) and β-hydroxybutyrate (BHBA). For the second and third objective, milk samples were collected weekly from 96 dairy cows from week 2 till 9 postpartum and analyzed for milk composition and N(A)Ab binding myosin, transferrin and thyroglobulin (experiment 2). For both datasets, N(A)Ab titers are expressed as (2)log values of the highest dilution giving a positive reaction. Data are expressed as means ± SEM. Repeated observations were analyzed in a mixed model. In experiment 1, no diet effect (P>0.05) was observed on NAb binding LPS in plasma or milk, NAb binding KLH in milk was greater (P=0.05) for cows fed the control diet. Concentration of NAb binding KLH and LPS in plasma was negatively related to plasma NEFA concentration (P<0.05). In experiment 2, NAb binding myosin (5.66 ± 0.06), thyroglobulin (4.85 ± 0.06), and transferrin (5.76 ± 0.07) were identified in milk. Clinical mastitis incidence (9%) tended to be positively related to concentration of NAb binding myosin (P=0.06) and negatively related to Nab binding transferrin (P=0.08). In conclusion, NAb binding KLH and LPS in plasma and milk are related to metabolic health, as indicated by plasma NEFA concentration. Furthermore, this study demonstrates the presence of N(A)Ab in bovine milk and shows trends for a relation between N(A)Ab binding auto-antigens and mastitis. Future studies should confirm these trends and shed light on the predictive value of N(A)Ab in bovine milk for mammary health.

The fatty acid profile of subcutaneous and abdominal fat in dairy cows with left displacement of the abomasum

The objective of this study was to determine the fatty acid (FA) profile and assess desaturase indices of nonesterified fatty acids (NEFA) in the blood, as well as in the abdominal (ABD) and subcutaneous (SUBC) fat stores, in dairy cows with left displacement of the abomasum (LDA). Blood, ABD, and SUBC samples were taken from 50 Holstein cows offered for surgery to correct LDA. The FA profile of the 3 compartments was determined by gas chromatography after lipid extraction, methylation, and, in the case of blood plasma, separation of lipid classes. The most abundant FA in all 3 compartments were 16:0, 18:0, and 18:1 cis-9, with a total proportion of 82.5, 68.0, and 74.1g/100 g of FA in ABD, NEFA, and SUBC, respectively. A principal component analysis was performed on the entire FA profile as well as on the Δ(9)-desaturase indices (14:1 cis-9/14:0, 16:1 cis-9/16:0, 18:1 cis-9/18:0). The principal component analysis extracted 2 principal components (PC), representing 51.6% (PC1) and 21.1% (PC2) of the total variance in FA composition of the 3 compartments. The loading plot for the regression factors revealed a strong positive correlation between PC1 with the Δ(9)-desaturase indices and the proportions of 14:1 cis-9 and 16:1 cis-9, and revealed a negative correlation with the proportion of 18:0 and saturated FA. The correlation with PC2 was positive for the proportion of unsaturated FA, 18:2n-6, and 18:3n-3, and negative for the proportion of 14:0, 16:0, and saturated FA. The SUBC could be distinguished from the NEFA and ABD by a positive score for PC1, whereas differentiation among the latter 2 compartments could be made by a positive (NEFA) or negative (ABD) score for PC2. The Δ(9)-desaturase indices for C14 and C16 differed between all compartments but were numerically closer for NEFA and ABD versus NEFA and SUBC. The desaturase indices of the main FA (18:1 cis-9 and 18:0) did not differ between NEFA and ABD. These results support the existence of a different FA composition in ABD compared with SUBC. The greater similarity between the FA profiles of ABD and NEFA compared with SUBC and NEFA and the closer desaturase indices of ABD and NEFA support the hypothesis of a preferential mobilization of ABD fat in dairy cows with LDA.

Integrating fasciolosis control in the dry cow management: the effect of closantel treatment on milk production.

The liver fluke Fasciola hepatica is a parasite of ruminants with a worldwide distribution and an apparent increasing incidence in EU member states. Effective control in dairy cattle is hampered by the lack of flukicides with a zero-withdrawal time for milk, leaving the dry period as the only time that preventive treatment can be applied. Here, we present the results of a blinded, randomized and placebo-controlled trial on 11 dairy herds (402 animals) exposed to F. hepatica to 1) assess the effect of closantel treatment at dry-off (or 80–42 days before calving in first-calving heifers) on milk production parameters and 2) evaluate if a number of easy-to-use animal parameters is related to the milk production response after treatment. Closantel treatment resulted in a noticeable decrease of anti-F. hepatica antibody levels from 3–6 months after treatment onwards, a higher peak production (1.06 kg) and a slightly higher persistence (9%) of the lactation, resulting in a 305-day milk production increase of 303 kg. No effects of anthelmintic treatment were found on the average protein and fat content of the milk. Milk production responses after treatment were poor in meagre animals and clinically relevant higher milk production responses were observed in first-lactation animals and in cows with a high (0.3–0.5 optical density ratio (ODR)), but not a very high (≥0.5 ODR) F. hepatica ELISA result on a milk sample from the previous lactation. We conclude that in dairy herds exposed to F. hepatica, flukicide treatment at dry-off is a useful strategy to reduce levels of exposure and increase milk production in the subsequent lactation. Moreover, the results suggest that treatment approaches that only target selected animals within a herd can be developed based on easy-to-use parameters.

Influence of a marine algae supplementation on the oxidative status of plasma in dairy cows during the periparturient period.

This study was part of a larger study that addressed the effects of marine algae (ALG) supplementation in the ration of high yielding periparturient dairy cows. The objectives were to induce milk fat depression (MFD) in early lactation by feeding docosahexaenoic acid (DHA) from ALG and to determine the effects on milk production, milk components and metabolic status early post partum. This study focuses on the oxidative status in the plasma during the ALG supplementation. Plasma samples were collected from 16 Holstein Friesian cows at the day of parturition and at -1, 2, 4 and 6 weeks relative to calving with half of the cows receiving the ALG supplement (44gDHA/d) from 3 weeks pre partum on. The following parameters were measured in plasma: ferric reducing ability of plasma (FRAP), α-tocopherol level, glutathione peroxidase activity (GSH-Px) and thiobarbituric acid reactive substances (TBARS) concentration. There was a significant effect of time for FRAP and α-tocopherol indicating changes in the plasma oxidative status around parturition. The ALG supplementation was successful in creating a milk fat depression (MFD) but could not improve the energy balance. Feeding of ALG significantly increased lipid peroxidation as measured by TBARS, probably through their high content of n-3 polyunsaturated fatty acids.

Insulin response of the glucose and fatty acid metabolism in dry dairy cows across a range of body condition scores

The objective of the present research was to determine the insulin response of the glucose and fatty acid metabolism in dry dairy cows with a variable body condition score (BCS). Ten pregnant Holstein Friesian dairy cows (upcoming parity 2 to 5) were selected based on BCS at the beginning of the study (2mo before expected parturition date). During the study, animals were monitored weekly for BCS and backfat thickness and in the last 2wk, blood samples were taken for determination of serum nonesterified fatty acid (NEFA) concentration. Animals underwent a hyperinsulinemic euglycemic clamp test in the third week before the expected parturition date. The hyperinsulinemic euglycemic clamp test consisted of 4 consecutive insulin infusions with increasing insulin doses: 0.1, 0.5, 2, and 5mIU/kg per minute. For each insulin infusion period, a steady state was defined as a period of 30min where no or minor changes of the glucose infusion were necessary to keep the blood glucose concentration constant and near basal levels. During the steady state, the glucose infusion rate [steady state glucose infusion rate (SSGIR) in µmol/kg per minute] and NEFA concentration [steady state NEFA concentration (SSNEFA) in mmol/L] were determined and reflect the insulin response of the glucose and fatty acid metabolism. Dose response curves were created based on the insulin concentrations during the steady state and the SSGIR or SSNEFA. The shape of the dose response curves is determined by the concentration of insulin needed to elicit the half maximal effect (EC50) and the maximal SSGIR or the minimal SSNEFA for the glucose or fatty acid metabolism, respectively. The maximal SSGIR was negatively associated with variables reflecting adiposity of the cows (BCS, backfat thickness, NEFA concentration during the dry period, and absolute weight of the different adipose depots determined after euthanasia and dissection of the different depots), whereas the EC50 of the glucose metabolism was positively associated with these variables. These results reflect a decreased insulin sensitivity and a decreased insulin responsiveness of the glucose metabolism in overconditioned dry dairy cows. The minimal SSNEFA and the EC50 of the fatty acid metabolism were not associated with variables reflecting adiposity of the cows, meaning that the insulin response of the fatty acid metabolism was not associated with the level of fat accumulation in dry dairy cows. Additionally, within individual cows, the EC50 of the glucose metabolism was higher than the EC50 of the fatty acid metabolism, meaning that the response of the fatty acid metabolism occurs at lower insulin concentrations compared with the response of the glucose metabolism. It can be concluded that a negative association exists between the level of fat accumulation in pregnant dairy cows at the end of the dry period and the insulin response of the glucose metabolism.

Age at calving in heifers and level of milk production during gestation in cows are associated with the birth size of Holstein calves

The objective of the present study was to evaluate environmental and dam factors associated with birth size of Holstein calves. Data of 1,594 births from dairy herds in Belgium and Germany were analyzed in a retrospective cross-sectional study. Immediately after birth, the birth weight of the calves was measured. On the next day, the heart girth (HG), withers height, and diagonal length of the dams and calves were measured. Parity, body condition score, gestation length (GL), and age at calving were recorded for all dams. For the cows, days open, lactation length, length of the dry period, and calving interval were also calculated. The magnitude and shape of the lactation that took place during gestation was quantified using the MilkBot model based on monthly milk weights. Using the same procedure, cumulative milk production from conception to drying off (MGEST) was calculated. After descriptive analyses, mixed models were used to identify factors that are significantly associated with the birth weight (most consistent measure of size at birth) of the calves born to both heifers and cows. Of the variables offered to the offspring birth weight model in heifers (n=540), calf sex, season of calving, GL, HG, withers height, diagonal length, and age at calving were significant. The mean birth weight of the calves born to heifers was estimated to be 41.3±1.01kg. In comparison to calves born to old (25.5 to 37.3mo; n=99) heifers, the birth weight was estimated to be 2.75, 3.29, and 2.35kg heavier when the calves were born to very young (20.3 to <22mo; n=98), young (22 to <23.5mo; n=145), and standard aged (23.5 to <25.5mo; n=198) heifers, respectively. Of the variables offered to the offspring birth weight model in cows (n=1,054), calf sex, season of calving, GL, parity, dry period, and MGEST were significant. The mean birth weight of the calves born to cows was estimated to be 44.1±0.99kg. For cows having an identical HG, the birth weight of the calves was estimated to be 0.97 and 1.11kg higher in cows with low (1,400 to <5,400kg) and high (6,500 to <7,200) MGEST, respectively, compared with cows with very high (7,200 to 11,600kg) MGEST. The decisive effects of age at calving in heifers and of high milk production levels during gestation in cows on the birth weight of their calves may provide a basis for developing managerial interventions to improve long-term health and productivity of the offspring.

Influence of adipocyte size and adipose depot on the in vitro lipolytic activity and insulin sensitivity of adipose tissue in dairy cows at the end of the dry period

The aim of the present research was to describe characteristics of adipose tissue lipolysis in dairy cows with a variable body condition score (BCS). Ten clinically healthy Holstein Friesian cows were selected based on BCS and euthanized 10 to 13 d before the expected parturition date. Immediately after euthanasia, adipose tissue samples were collected from subcutaneous and omental fat depots. In both depots, we observed an increase in adipocyte size with increasing BCS. Using an in vitro explant culture of subcutaneous and omental adipose tissue, we aimed to determine the influence of adipocyte size and localization of adipose depot on the lipolytic activity in basal conditions and after addition of isoproterenol (nonselective β-agonist) and insulin in different concentrations. Glycerol release in the medium was used as a measure for lipolytic activity. We observed that the basal lipolytic activity of subcutaneous and omental adipose tissue increased with adipocyte volume, meaning that larger fat cells have higher basal lipolytic activity independent of the location of the adipose depot. Dose-response curves were created between the concentration of isoproterenol or insulin and the amount of glycerol released. The shape of the dose-response curves is determined by the concentration of isoproterenol and insulin needed to elicit the half-maximal effect and the maximal amount of stimulated glycerol release or the maximal inhibitory effect of insulin. We observed that larger fat cells released more glycerol upon maximal stimulation with isoproterenol and this was more pronounced in subcutaneous adipose tissue. Additionally, larger fat cells had a higher sensitivity toward lipolytic signals. We observed a trend for larger adipocytes to be more resistant to the maximal antilipolytic effect of insulin. The insulin concentration needed to elicit the half-maximal inhibitory effect of insulin was within the physiological range of insulin and was not influenced by adipocyte size or adipose depot. We conclude that overconditioned cows have larger adipocytes and are predisposed to excessive mobilization of body fat due to a higher basal and stimulated lipolytic activity of large adipocytes while the antilipolytic effect of insulin is preserved.

Effect of uterine lavage on neutrophil counts in postpartum dairy cows

Subclinical endometritis affects approximately 30% of lactating dairy cows, causing significant economic losses to the dairy industry. Yet, there is no efficient treatment available for this condition. The present study examines the effect of uterine lavage in clinically normal cows with sterile saline solution at 30 days in milk (DIM) on the percentage of polymorphonuclear cells (PMNs) detected with endometrial cytology as an indicator of subclinical endometritis. It was hypothesized uterine lavage would be a technique to reduce the number of PMNs in the uterus, and hence be beneficial for cows affected by subclinical endometritis. Cytology samples were taken by low-volume flushing from 50 Holstein Friesian cows on 30 and 40 DIM. On Day 30, cows were clinically examined and randomly assigned into a treatment and control group. In the treatment group, the cytology sampling on Day 30 was immediately followed by uterine lavage with 500-600 mL of sterile physiological saline (35-40°C). Cytology sampling was repeated in all cows at 40 DIM. Lactation numbers >2, peripheral progesterone concentrations >1 ng/mL and uterine lavage at 30 DIM all were significantly associated with lesser PMN percentages at 40 DIM (P=0.0041; 0.0187 and 0.0043, respectively). Uterine lavage might, therefore, be a useful and practical method to decrease the number of PMNs in the uterus of cattle. Results from the current study can be used as preliminary data for designing in depth therapeutic protocols for treatment of subclinical endometritis in cattle.
