Seroprevalence of equine piroplasms in the Republic of Korea

Equine piroplasms include two tick-borne protozoan parasites, Babesia caballi and Theileria equi. Although no clinical equine piroplasmosis has been reported in the Republic of Korea, the possible existence of the disease has been proposed due to a nationwide distribution of the vector ticks. To determine if the antibodies against B. caballi and T. equi were present, 184 sera of horses (Equus caballus) raised in the Republic of Korea from 2007 to 2010 were assessed using cELISA kits. Two (1.1%) out of 184 sera were positive for T. equi, but none were seropositive for B. caballi. Both samples tested positive came from one region (Gyeonggi province). The accuracy of the cELISA was confirmed by PCR using primers specific to the 18S rRNA of T. equi. This study presents for the first time horses infected by T. equi in the Republic of Korea. Since the infection of T. equi occurred in horses raised in the Republic of Korea, further studies with continuous monitoring of the vector ticks for equine piroplasms and appropriate control programs need to be established.

Molecular and serologic detection of Coxiella burnetii in native Korean goats (Capra hircus coreanae).

The occurrence of Q fever in native Korean goats (Capra hircus coreanae) was investigated for the first time in the country using ELISA and PCR. A total of 597 blood samples were collected from goats belonging to five different provinces of Korea. To detect Coxiella burnetii, sera were separated from the whole blood and analysed by ELISA; DNA was extracted directly from the whole blood and analysed by PCR. Overall, 114 (19.1%, 95% C.I.=16.1-22.4) and 57 goats (9.5%, 95% C.I.=7.5-12.2) tested positive for C. burnetii in the ELISA- and PCR-based screening, respectively, while 18 goats (3.0%, 95% C.I.=1.9-4.7) tested positive in both the assays. There was a significant difference between the number of ELISA- and PCR-positive goats (P<0.05). The seroprevalence of Q fever was significantly higher among the adult goats (≥1y, 22.0%) than among the young goats (<1y, 13.8%) (P<0.05). While the results of the serologic analysis showed no seasonal variation, data from the PCR-based assay indicated that there were a higher number of positive cases during the cold seasons. Because Q fever infection has high rates of prevalence in native Korean goats, further studies on humans at a high risk of contracting this disease should be conducted. The PCR-based assay used in this study is a useful method for the direct detection of C. burnetii in blood samples from small ruminants.

Serological responses after vaccination of growing pigs with foot-and-mouth disease trivalent (type O, A and Asia1) vaccine.

Korea experienced its fifth Foot-and-Mouth Disease (FMD) outbreak (type O) since 1934 from November 2010 to April 2011. The Korean government initiated emergency vaccination for all FMD-susceptible domestic animals in December 2010 using type O FMD vaccines. Starting in September 2011, trivalent FMD vaccines (types O, A, and Asia1) were used for the vaccination of all animals. This study was performed to identify the appropriate time for FMD vaccination in growing pigs when vaccination is applied only once (at either 8 weeks or 12 weeks of age). Seroprevalences from growing pigs under different vaccination regimens (once or twice) were also studied. A total of 526 growing pigs on 7 farms were used in this study. This study showed that the vaccination of growing pigs at both 8 and 12 weeks of age resulted in higher seroprevalences (97.5% in type O, 92.3% in type A and 99.4% in type Asia1) than did a single vaccination at 8 weeks of age (86.7% in type O, 88.0% in type A and 93.0% in type Asia1) (P<0.05). Pigs vaccinated once at 8 weeks of age showed much higher seroprevalences than pigs vaccinated once at 12 weeks of age (60.9% in type O, 62.8% in type A and 77.6% in Asia1, respectively) (P<0.05).

Novel foot-and-mouth disease virus in Korea, July-August 2014

Despite nation-wide immunization with O, A, and Asia 1 type vaccines in Republic of Korea, foot-and-mouth disease type O occurred again in July 2014 after three years and three months. This virus was a Mya-98 strain of the Southeast Asian topotype and was most similar to the identified type that circulated in East Asia in 2014. This was new virus with the deletion of 23 amino acids in 3A/3B1 region and low pathogenic property.

Robust Protection against Highly Virulent Foot-and-Mouth Disease Virus in Swine by Combination Treatment with Recombinant Adenoviruses Expressing Porcine Alpha and Gamma Interferons and Multiple Small Interfering RNAs.

ABSTRACT Because the currently available vaccines against foot-and-mouth disease (FMD) provide no protection until 4 to 7 days postvaccination, the only alternative method to halt the spread of the FMD virus (FMDV) during outbreaks is the application of antiviral agents. Combination treatment strategies have been used to enhance the efficacy of antiviral agents, and such strategies may be advantageous in overcoming viral mechanisms of resistance to antiviral treatments. We have developed recombinant adenoviruses (Ads) for the simultaneous expression of porcine alpha and gamma interferons (Ad-porcine IFN-αγ) as well as 3 small interfering RNAs (Ad-3siRNA) targeting FMDV mRNAs encoding nonstructural proteins. The antiviral effects of Ad-porcine IFN-αγ and Ad-3siRNA expression were tested in combination in porcine cells, suckling mice, and swine. We observed enhanced antiviral effects in porcine cells and mice as well as robust protection against the highly pathogenic strain O/Andong/SKR/2010 and increased expression of cytokines in swine following combination treatment. In addition, we showed that combination treatment was effective against all serotypes of FMDV. Therefore, we suggest that the combined treatment with Ad-porcine IFN-αγ and Ad-3siRNA may offer fast-acting antiviral protection and be used with a vaccine during the period that the vaccine does not provide protection against FMD. IMPORTANCE The use of current foot-and-mouth disease (FMD) vaccines to induce rapid protection provides limited effectiveness because the protection does not become effective until a minimum of 4 days after vaccination. Therefore, during outbreaks antiviral agents remain the only available treatment to confer rapid protection and reduce the spread of foot-and-mouth disease virus (FMDV) in livestock until vaccine-induced protective immunity can become effective. Interferons (IFNs) and small interfering RNAs (siRNAs) have been reported to be effective antiviral agents against FMDV, although the virus has associated mechanisms of resistance to type I interferons and siRNAs. We have developed recombinant adenoviruses for the simultaneous expression of porcine alpha and gamma interferons (Ad-porcine IFN-αγ) as well as 3 small interfering RNAs (Ad-3siRNA) to enhance the inhibitory effects of these antiviral agents observed in previous studies. Here, we show enhanced antiviral effects against FMDV by combination treatment with Ad-porcine IFN-αγ and Ad-3siRNA to overcome the mechanisms of resistance of FMDV in swine.

A recombinant adenovirus bicistronically expressing porcine interferon-α and interferon-γ enhances antiviral effects against foot-and-mouth disease virus

Foot-and-mouth disease (FMD) is a virulent and economically costly disease in domestic livestock. Since the current vaccine available against FMD provides no protection until 7days postvaccination, the only alternative method to halt the spread of the FMD virus (FMDV) during outbreaks is by the application of anti-viral agents. The combination of recombinant adenovirus expressing type I interferon (IFN-α) and adenovirus expressing type II IFN (IFN-γ) has been reported to be an effective anti-viral treatment strategy against FMDV. Nevertheless, the recombinant adenovirus mixture may be inefficient because of the low anti-viral efficiency of IFN-γ compared to that of IFN-α. In this study, we generated a recombinant adenovirus co-expressing porcine IFN-α and IFN-γ in tandem using an FMDV 2A sequence to mediate effective cleavage of the two proteins (referred to as Ad-porcine IFN-αγ). We demonstrated that both recombinant porcine IFN-α and IFN-γ were expressed and interferon stimulated gene (ISG)s related with IFN-α and IFN-γ were induced in porcine kidney (IBRS-2) cells infected with Ad-porcine IFN-αγ. Additionally, the anti-viral effects of Ad-porcine IFN-αγ against FMDV were enhanced both in IBRS-2 cells and in CD-1 (ICR) suckling mice compared to that of adenovirus expressing only a single protein. We propose that Ad-porcine IFN-αγ could be a rapid, highly efficient, convenient anti-viral agent against FMDV.

Detection and Genotyping of Coxiella burnetii and Coxiella-Like Bacteria in Horses in South Korea

Coxiella burnetii and Coxiella-like bacteria (CLB) are genetically and ecologically distinct despite some genetic similarities. Furthermore, CLB are exceptionally diverse and widespread in ticks, but rarely detected in domestic animals. Since Coxiella bacteria can be transmitted from infected horses by inhalation or by coming in contact with ticks during activities such as horseback riding, it is necessary to study their prevalence. To the best of our knowledge, this is the first large-scale nationwide investigation of the prevalence of C. burnetii and CLB among horses reared in South Korea. Of 816 blood samples collected between 2007 and 2013, 11 (1.3%) were identified as C. burnetii by ELISA, and six (0.7%) as CLB by 16S rRNA sequencing. While a sequence from Jeju Island was similar (97.9–100%) to those within clade B, five sequences obtained from the northern region were categorized into a new clade, indicating the sequence diversity of the genus Coxiella. Studies until date had detected CLB only in ticks; here, we describe their detection in mammals. Given their zoonotic potential, strategic monitoring and appropriate control programs for Coxiella species need to be established.

Detection and Determination of Toxoplasma gondii Seroprevalence in Native Korean Goats ( Capra hircus coreanae )

Toxoplasma gondii is an important zoonotic protozoan pathogen that causes serious illness in immunocompromised humans and infection in animals worldwide. The current study was conducted for detection of T. gondii infection and determination of the seroprevalence of the pathogen in native Korean goats (Capra hircus coreanae). Analysis of a total of 610 sera samples collected from 60 herds between 2009 and 2011 were performed using a commercial enzyme-linked immunosorbent assay (ELISA) kit for detection of anti-T. gondii IgG antibodies. Among the animals tested, 5.1% (31/610) showed seropositivity for anti-T. gondii antibodies, and 38.3% (23/60) of the herds were seropositive. The prevalence rates between young (<1 year) and adult (≥1 and ≤3 years) goats were 7.0% and 4.1%, respectively, without statistical significance (p>0.05). Likewise, the prevalence rates observed during cold season (October-March) and warm season (April-September) were 2.9% and 5.5%, respectively, without statistical significance. Seroprevalence rates observed in the northern, central, and southern regions were 7.9%, 3.8%, and 4.2%, respectively. In conclusion, we report for the first time on the seroprevalence of anti-T. gondii antibodies in native Korean goats (Capra hircus coreanae). The results of this study also indicate that there is a nationwide distribution of T. gondii infection among goats. Therefore, the implementation of integrated control strategies as well as measures for prevention and control of T. gondii infection within goats is recommended.

Evidence of Toxoplasma gondii exposure among horses in Korea.

The present study investigated the seroprevalence of Toxoplasma gondii (T. gondii) antibodies by ELISA in horses reared in Korea. Serum samples were collected from 2009 through 2013 from 816 horses reared in Korea. Analysis was performed using a commercial toxoplasmosis ELISA kit to detect anti-T. gondii antibodies. Overall, 24 out of 816 horses (2.9%) were seropositive for T. gondii. The result was analyzed by age, gender, breed and region. Significant differences were observed according to breed and region (P<0.05). This is the first nationwide serological investigation of T. gondii in horses reared in Korea. The study results reveal that T. gondii occurs nationwide in Korean horses.

Reemergence of Foot-and-Mouth Disease, South Korea, 2000–2011

Five outbreaks of foot-and-mouth disease have occurred in South Korea during 2000–2011. Macro-analysis of these outbreaks showed a correlation with outbreaks in countries in eastern Asia. Genetic analyses of food-and-mouth disease viruses in South Korea showed a correlation with viruses that are prevalent in neighboring countries.
