Effect of hesperidin extraction on cell proliferation and apoptosis of CNE-2Z cells

Objective: To study the effect of hesperidin extracted from the peel of Gannan kumquat on cell proliferation and apoptosis of human poorly diferentiated nasopharyongeal carcinoma epithelial cell (CNE-2Z). Methods: Effect of hesperidin on proliferation of CNE-2Z cells was detected by the MTT method. Effect of hesperidin on apoptosis of CNE-2Z cells was utilized by the acridine orange / ethidium bromide staining method. Results: After being treated with different hesperidin for 24, 48 and 72 h respectively, the proliferation of CNE-2Z cells was inhibited in vitro; different time had high significant difference on the proliferation of CNE-2Z cells (F = 1898.865, P < 0.001), different pharmacal concentration had had high significant difference on the proliferation of CNE-2Z cells (F = 5454.040, P < 0.001). Time and pharmacal concentration had interaction on the proliferation of CNE-2Z cells (F = 118.909, P < 0.001). During the concentration of 1µmol/L-100µmol/L, inhibition ratio of CNE-2Z cells increased gradually along with the increment of pharmacal concentration and the extension of time. When the concentration was higher than 100 µmol/L, its inhibition no more reinforced accompanied with the increasement of pharmacal concentration (P> 0.05) and presented the determinate saturability. The aggregation of cellular nuclear chromatin, karyopycnosis, karyorrhexis and apoptotic body was present in CNE-2Z cells detected by acridine orange/ethidium bromide fluorescent staining after the utilization of hesperidin. Conclusion : Hesperidin inhibited the cellular division growth and induced the cell apoptosis of CNE-2Z cells.

Effect of hesperidin extraction on cell proliferation and apoptosis of Alzheimer's disease induced by Aβ 25–35

Objective: To study the effect of hesperidin extracted from the peel of Gannan kumquat on Alzheimers disease and its possible mechanisms. Methods: Cellular model of Alzheimers disease was established using PC12 cell induced by serum starvation method and treated by β-amyloid peptide 25–35 (Aβ<inf>25–35</inf>). Effect of hesperidin on proliferation of PC12 cell induced by Aβ<inf>25–35</inf> was detected by the MTT method. Effect of hesperidin on apoptosis of PC12 cell induced by Aβ<inf>25–35</inf> was utilized by the acridine orange / ethidium bromide staining method. Results: PC12 cells stayed at G<inf>0</inf> stage induced by serum starvation method in order to simulate the neuron in the state of terminal differentiation. After this, the PC cells induced by serum atarvation were treated by Aβ<inf>25–35</inf> to establish the cellular model of Alzheimers disease. After synchronized PC12 cells being pretreated with different concentrations of hesperidin (0–200 µmol/L) for 1 h, and then 30 µmol/L Aβ<inf>25–35</inf> for 24 h, it was found that the survival rate increased along with the increasing dosage of hesperidin (F = 995.8,P < 0.001); however, there was no significant difference between 100 µmol/L group and 200 µmol/L group (P > 0.05). After synchronized PC12 cells being pretreated with 100µmol/L hesperidin for 1 h, and then 30 µmol/L Aβ<inf>25–35</inf> for 24h, the cell nuclei were gradually similar to the cell nuclei of control group by by acridine orange/ethidium bromide fluorescent staining.Conclusion:Hesperidin inhibited the cytotoxicity and apoptosis induced by Aβ<inf>25–35</inf> and had protection for neurocyte.

Investigation on Current Situation of T2DM in Ganzhou City and Countryside and Analysis of Correlation Factors

Objective: to study the current situation and distributional characteristics of T2DM in Ganzhou city and countryside and to explore the correlation factors of T2DM. Methods: the baseline of T2DM and its correlation factors was investigated in the 35- to 64-year-old residents in Ganzhou by multiple horizon Hierarchies random sampling method from 2009 to 2016. The WHO diagnostic code of 1999 was adopted for the diagnosis of T2DM. Results: the prevalence of T2DM and IFG in Ganzhou was 7.7% (683/8906) and 13.1% (1,167/8,906) respectively. The prevalence of T2DM was 9.6% (283/2947) and 14.9% (112/751) in overweight population and obesity population respectively which was higher than in BMI normal population [6.7% (289/4317), x2=62.185, P<0.001]. The prevalence of IFG was 21.5% (634/2947) and 25.5% (192/751) in overweight population and obesity population which was obviously higher than in BMI normal population [16.2% (699/4317), x2=55.191, P<0.01]. The prevalence of T2DM and IFG in hypertension population was 12.4% (306/2469) and 23.7% (457/1929) which was obviously higher than in non-hypertension population [7.0% (369/5274),x2=19.309, P<0.001 and 17.1%(705/4121),x2= 14.474, P<0.001]. The prevalence of T2DM and IFG in hyperlipemia population was 12.3% (297/2412) and 23.7% (451/1902) respectively which was obviously higher than in non-hyperlipemia population [6.2% (319/5152),x2=61.709, P<0.001 and 17.7% (709/4003), x2=15.674 , P<0.001]. The prevalence of male T2DM and IFG was 8.5% (383/4504) and 13.2% (479/3630) respectively. The prevalence of female T2DM and IFG was 7.0% (296/4224) and 19.4% (660/3404) respectively. There was not statistically significant between the male and female T2DM (x2 =3.208, P=0.073), but there was statistically significant between the male and female IFG (x2 =29.222, P<0.001). The prevalence of T2DM and IFG in city was 8.0%(349/4366) and 20.0% (670/3350). The prevalence of T2DM and IFG in countryside was 7.3%(240/3293) and 19.4% (490/2527) respectively. There was not significant in T2DM or IFG between in the city and in the countryside (x2=0, P=1 and x2=0.193, P=0.660). Conclusion: the prevalence of T2DM and IFG in Ganzhou city and countryside was severe. The health education should be carried out for moderate and small city which has just recently transformed and been rich. The early screening for T2DM should be evolved through the onset risk prediction model of T2DM in order to decrease the prevalence of T2DM.

Meta-analysis of association between SLC30A8 rs13266634 and T2DM in Chinese population

We use Stata software to perform a meta-analysis for evaluating the relationship between rs13266634 polymorphism in Solute carrier family 30member8(SLC30A8) and type2 diabetes mellitus(T2DM). Through the Stata software, we calculated the combined effect and checked the publication bias. A total of 13 studies matched with the inclusion criteria are collected, including 11923 T2DM cases and 14702 control cases. The total OR(95%CI) was calculated and the publication bias was assessed with funnel plots and Egger method. Heterogeneity test show statistically significant heterogeneity between studies (Q=26.72, P=0.008), the different of frequency distribution of allele gene of SLC30A8 rs13266634(C/T) in case and control group was statistically significant; the frequency distribution of C-allele in case group is outstanding higher than control group. Total OR (95%CI) is 1.21 (1.14, 1.28), P=0.005; the risk of T2DM with CC genotype carrier is 1.57 times than TT gene type, OR (95%CI) is 1.57 (1.26, 1.95), P=0.005; the frequency of CT and TT genotype carrier has no significant difference, Publication bias analysis and sensitive analysis show that the results of Meta-analysis is stable and reliable. Our meta-analysis show the association between C-allele of SLC30A8 rs13266634 polymorphism and Chinese T2DM is related, CC genotype may be the risk gene of Chinese T2DM.

Effect of hesperidin extraction on the bone marrow polychromatophilic erythroblast micronucleus of exercise fatigue

To study the effect of hesperidin extracted from the peel of Gannan kumquat on the bone marrow polychromatophilic erythroblast micronucleus of exercise fatigue. Methods: Hesperidin was extracted from the peel of kumquat by ultrasound and ethanol precipitation and was detected qualitatively by the high performance liquid chromatogramphy. Effect of hesperidin on the micronuclear rate of polychromatophilic erythroblasts of rat marrow was used by the micronucleus test. Results: The rats of exercise fatigue had some phenomena, such as depilation, tired appearance, low appetite, degressive locomotory capacity and slow-moving growth of body weight. Exercise fatigue caused the rats to increase the micronuclear rate of marrow polychromatophilic erythroblasts (P < 0.001). Hesperidin decreased the micronuclear rate of marrow polychromatophilic erythroblasts (P < 0.05–0.001). Conclusion: Hesperidin had inhibited the exercise fatigue through inhibiting the production of micronuclear rate of marrow polychromatophilic erythroblasts.
