UNLABELLED IIntroduction and objective. In recent years there has been intensified research on medicinal preparations of fungal origin. Some fungal polysaccharides may directly affect the inhibition of cancer cells proliferation which, stopping the cell cycle, leads to apoptosis. One of these substances (component of extract of Ganoderma spp) is extensively tested for its anti-cancer properties on the tumor cell lines. Electric cell-substrate impedance sensing (ECIS) is an in vitro impedance measuring system using alternating current (AC) to determinate the behaviour of the cells in physiological conditions. OBJECTIVE The aim of the study was to examine the electric properties (resistance, capacitance and impedance) of mouse fibroblasts cell line L929 after treatment by different concentration of crude exopolysaccharides from Ganoderma applanatum (GpEPS) in real time by ECIS technique. MATERIALS AND METHODS For the study, the L929 cell line culture was treated by different concentrations of GpEPS: C1=228.5 µg/mL; C2=22.85 µg/mL; C3=2.285 µg/mL; C4=0.2285 µg/mL; and C5=0.02285 µg/mL. Default optimal frequencies were used: Resistance (R) 4000Hz, Impedance (Z) 16000Hz, Capacitance (C) 64000Hz. RESULTS The study demonstrated that GpEPS had no significant effect on the resistance, capacitance and impedance cells cultures, which implies that there is no significant effect on the physiological processes of L929 fibroblasts. This indicates the possibility of using GpEPS preparation in anti-cancer therapy. CONCLUSIONS In the future, following further studies (comprising in preventive and therapeutic actions), GpEPS can be safely used in anti-cancer therapy which does not cause side-effects or damage to healthy cells.

OBJECTIVES The aim of this study was to examine whether obesity affects cognitive functions in people suffering from mental illness. METHODS 91 persons suffering from mental illness, including 51 women and 40 men took part in the study. Mean age of patients was 46 years. These persons were under constant psychiatric care, they were the participants of the daily support centre. Overweight and obesity was measured by Body Mass Index (BMI). Abdominal obesity was measured according to IDF guidelines and waist-hip ratio (WHR). Cognitive functions were examined using STMS, Verbal Fluency Test and Rey Auditory Verbal Test. RESULTS Abdominal obesity was diagnosed in 70% of patients according to IDF guidelines and in 61% according to WHR, in women these results were respectively: 83% and 94%, while in men 62% and 32%. BMI distribution in the study group was respectively: obesity class II - 5 persons (5%), obesity class I - 26 persons (28%), overweight - 32 persons (35%), correct BMI - 27 persons (30%), underweight - 1 person (2%). There was a negative correlation between WHR, waist circumference and abstract reasoning, direct memory and delayed memory. BMI and body mass correlated negatively only with delayed memory. Number of medications taken by the studied persons showed a positive correlation with body mass and waist circumference. Negative correlations between cognitive functions and body mass, overweight and abdominal obesity was observed in women. In men these correlations were not observed, there were only correlations between cognitive functions and age. CONCLUSIONS Obesity contributes to a decline in cognitive functions especially in direct memory and abstract reasoning.

This paper presents issues related to the creation of research plates with titanium electrodes, which are a new alternative to currently used plates with gold electrodes. Methods for obtaining metallization of titanium, necessary for the production of electrodes are described. Various lithography techniques are presented for processing the obtained layer of the material. The stages of designing and manufacturing comb capacitors and ready-made research plates have been presented. Finally, substrates with a titanium layer have been designed and manufactured to allow the impedance of living cells to be measured.

The article presents the construction and technology of comb capacitors on a polycarbonate substrates. The structures have been designed to be used in the ECIS measurement system that allows monitoring the activity of the examined cells by analyzing the changing electrical parameters impedance, resistance and capacitance in real time. The copper layer was embedded by sputtering method. The resulting comb capacitors were used in a test sample of substances with different chemical properties.

In recent years, tannase has gained increasing interest mainly because of its potential applications. One of the most important functions of tannic acid (TA) hydrolase is the release of gallic acid (GA) from complex tannins. The aim of the study was to determine the dynamic changes in tannase activity depending on the carbon source in the culture medium. An extracellular and intracellular tannase activity analysis was carried out with the use of spectrophotometric analysis and confirmed by capillary electrophoresis in cultures of white‐rot fungi: Phellinus pini, Fomes fomentarius, and Tyromyces pubescens. The inducible potential of TA and rapeseed meal on the activity of tannin acyl hydrolase was confirmed during 14 days of culturing. Different effects of the tested compounds on stimulation of tannase activity in selected fungal strains have been demonstrated. We concluded that rapeseed meal was the best inducer of tannase activity in the case of P. pini. However, the highest concentrations of GA were observed after stimulation by the TA in the cultures of F. fomentarius and T. pubescens.

BACKGROUND Hypertension-induced endothelial dysfunction is associated with an impaired bioavailability of nitric oxide regulated through interactions between nitric oxide synthase and heat shock protein-90 (Hsp-90). The role of Hsp-90 in the development of arterial hypertension remains unclear. OBJECTIVES The objective of the study was to evaluate serum concentrations of Hsp-90a in patients with arterial hypertension in comparison to their normotensive counterparts. MATERIAL AND METHODS The study was performed on 49 adults (mean age 55.6 years) with an elevated waist circumference. The individuals presented no subjective feeling of any disease, admitted no drug treatment for any condition, and had not previously been diagnosed with the metabolic syndrome. Patients were screened for arterial hypertension and other component disorders of the metabolic syndrome. Hsp-90a concentrations were evaluated by enzyme-linked immunosorbent assay (ELISA). RESULTS Twenty-eight subjects were diagnosed with arterial hypertension, while 21 individuals had normal blood pressure. Twenty-five patients satisfied the metabolic syndrome diagnostic criteria. Hsp-90a concentrations were significantly higher (p = 0.002) in the individuals with arterial hypertension than in their normotensive counterparts (median ± interquartile range): 19.42 ng/mL ± 5.17 vs. 16.86 ng/mL ± 3.18. The concentrations of Hsp-90a correlated positively with systolic blood pressure (R = 0.39; p = 0.005) and diastolic blood pressure (R = 0.29; p = 0.046). CONCLUSIONS An increase in Hsp-90a concentrations in patients with arterial hypertension may be a compensatory mechanism for the impaired bioavailability of nitric oxide. The role of Hsp-90a as an early marker of hypertension-associated endothelial injury should be confirmed in further studies on a larger group of patients.

The effect of supplementation of medium with rapeseed meal (RM) on production of biotechnologically important enzymes was investigated in submerged cultures of the white rot fungus Cerrena unicolor. The addition of RM (3.5% w/v) distinctly stimulated the activities of laccase, chitinase, and β-glucosidase. As compared to the control, the activities of chitinase, β-glucosidase, and laccase in the RM supplemented cultures were up to 4.1, 8.4, and 3.9 times higher, respectively. The results of the spectrophotometric and spectrofluorometric measurements were additionally confirmed by zymographic analysis of the samples. The level of sugars and phenolic compounds as well as the antioxidative ability of fungal preparations were also determined. The results obtained indicate that the submerged liquid fermentation of rapeseed meal can be proposed as an inexpensive and very effective method for biotechnological production of chitinase, β-glucosidase, and laccase by C. unicolor.
