Comparative Ovulation-Inducing Capacity of a Synthetic Luteinizing Hormone-Releasing Hormone and Two Analogs in Adult Rats and Mice*

Two analogs of luteinizing hormone-releasing hormone (LH-RH), H-Pyr-His-Trp-Ser-Tyr-[d-Ala]-Leu-Arg-Pro-NH-CH 2 -CH 3 (AY-25,205) and H-Pyr-His-Trp-Ser-Tyr-[d-Trp]-Leu-Arg-Pro-Gly-NH 2 (AY-25,650), were more potent than the synthetic LH-RH, H-Pyr-His-Trp-Ser-Tyr-Gly-Leu-Arg-Pro-Gly-NH 2 , in inducing ovulation in fluphenazine dihydrochloride-pretreated proestrous rats. The ED 50 values for AY-25,205, AY-25,650, and LH-RH were 3.0, 2.1, and 176.8ng, respectively. AY-25,205 and AY-25,650 were also more effective in inducing ovulation in pregnant rats (ED 50 26.2ng for AY-25,205 and 37.9ng for AY-25,650) and pseudopregnant rats (ED 50 23.0ng for AY-25,205 and 26.4ng for AY-25,650) than was LH-RH (ED 50 values of 14.1μg and 11.1μg, respectively, for pregnant and pseudopregnant rats). Again, these two analogs were more efficient in advancing premature ovulation in diestrous rats (ED 50 10.8ng for AY-25,205 and 14.1ng for AY-25,650) than was LH-RH (ED 50 1.2μg). At a dose of 80ng/rat or higher, AY-25,205 and AY-25,650 were partially effective in inducing premature ovulation in metestrous rats, whereas LH-RH was ineffective even at a dose of 10μg/rat. The ED 50 values for AY-25,205, AY-25,650, and LH-RH in inducing ovulation in diestrous mice were 4.6ng, 3.5ng, and 0.22μg, respectively. These findings show that AY-25,205 and AY-25,650 have similar activity in inducing ovulation in rats and mice and are much more potent than LH-RH in causing maturation and rupture of growing follicles.

A simple, sensitive, and reproducible method for the detection of urinary human chorionic gonadotropin (hCG) and diagnosis of early human pregnancy is reported. A 5-ml aliquot of filtered early-morning urine sample was concentrated in a microconcentrator (M) to 0.1 ml of retentate which was diluted with 0.4 ml of distilled water and tested in a hemagglutination inhibition test (M-HIT). Also, a 0.1-ml aliquot of filtered unconcentrated urine sample was diluted with 0.4 ml of distilled water and tested in the same hemagglutination inhibition test (HIT). Urine samples from women of reproductive age; from perimenopausal, menopausal, and proteinuric women; and from adult males were tested in the HIT and M-HIT. Some of these urine samples were also tested in the mouse ovulation bioassay (MOB). The M-HIT was significantly more reliable than the HIT for diagnosis of early pregnancy 25 to 55 days after menses. Correct negative results with the M-HIT were obtained in urine samples of most of the nonpregnant cycling, perimenopausal, and menopausal women, and adult males. Urine samples from subjects with severe proteinuria gave false-positive types of reactions in the M-HIT. Positive results were obtained in the MOB with a number of urine samples from pregnant, perimenopausal, and menopausal women. A properly conducted M-HIT should be very valuable in diagnosing pregnancy as early as the 26th day of the cycle in regularly menstruating women.
