Studies on the Cell Wall of Piricularia oryzae†: Part II. The Chemical Constituents of the Cell Wall

The chemical constituents of the cell wall of Piricularia oryzae, the pathogenic fungus of rice blast disease, were studied with the aids of chemical analysis, X-ray diffraction, infra-red absorption and enzymatic degradation. The sugar constituents were identified by chromatography as glucose (62%), mannose (4%), galactose (0.5%), and hexosamine (13%). The acidic amino acid rich protein was comprised 4.6% in the cell wall. The cell wall consists of at least three different polysaccharide complexes: a) α-Heteropolysaccharide protein complex containing mannose, glucose and galactose, b) β-1,3-Glucan containing β-1, 6-linked branch, c) Chitin like substance.

(1) Pir.の極大吸収を示す波長(蒸溜水または中性緩衝液では240mμ,強酸性溶液では251mμ)を用いて,分光光度計によるPir.の微量定量法を確立した. (2) 分光光度計による定量法とカップ検定法によりPir.の熱に対する安定性の実験を行って, Pir.は酸性溶液中では120°, 30分の加熱,或いは38°で28日間放置の条件では全く分解せずに安定であるが, pHの上昇に伴って安定性は少しずつ減少する.しかし強アルカリ性溶液を120°, 30分加熱しても30%の分解しか見られないことから,他の同様な物質に比較してPir.は極めて安定な物質と考えられる.

47菌株の稲熱病菌を用いてT. C. A-cycleに関与する有機酸塩の利用度,セルローズ利用度,ペクチナーゼの差異並びに北大農学部菌株のNo. 1及びNo. 2のビオチン生成量等の試験を行い次の如き結果を得た. (1) ビオチンを要求しないNo. 1及びNo. 2をビオチン欠合成培地で培養してビオチン生成能の有無を検した.即ち培養濾液及び洗滌菌体の酸加水分解物についてBioassayでビオチン量を試験した結果,ビオチン生成能は認められなかった.此等の菌株は糖代謝の型式が他の菌株とは異るものと考えられる. (2) 炭素源としてはコハク酸,フマール酸,クエン酸の利用度並びに此等の酸に1/100Mマロン酸を添加した培地の利用度に就て試験した結果, 9菌株に就てはT. C. A-cycle以外の経路も予想せられる. (3) セルローズの利用度試験の結果,利用度と病原性との間に関連性は認められるが分類の鍵にはなり得ない. (4) ペクチナーゼについては明確な差異が認められなかった. (5) 以上の如き結果よりセルローズ分解性,ペクチナーゼ等の差異より分類の鍵にする事は出来なかった.
