Quantitative analysis of biodiesel in blends of biodiesel and conventional diesel by comprehensive two-dimensional gas chromatography and multivariate curve resolution.

In this paper, a method to determine the composition of blends of biodiesel with mineral diesel (BXX) by multivariate curve resolution with Alternating Least Squares (MRC-ALS) combined to comprehensive two-dimensional gas chromatography with Flame Ionization Detection (GC×GC-FID) is presented. Chromatographic profiles of BXX blends produced with biodiesels from different sources were used as input data. An initial evaluation carried out after multiway principal component analysis (MPCA) was used to reveal regions of the chromatograms were the signal was likely to be dependent on the concentration of biodiesel, regardless its vegetable source. After this preliminary step MCR-ALS modeling was carried out only using relevant parts of the chromatograms. The resulting procedure was able to predict accurately the concentration of biodiesel in the BXX samples regardless of its origin.

Discriminating Brazilian crude oils using comprehensive two-dimensional gas chromatography–mass spectrometry and multiway principal component analysis ☆

The geochemical characterization of petroleum is an essential task to develop new strategies and technologies when analyzing the commercial potential of crude oils for exploitation. Due to the chemical complexity of these samples, the use of modern analytical techniques along with multivariate exploratory data analysis approaches is an interesting strategy to extract relevant geochemical characteristics about the oils. In this work, important geochemical information obtained from crude oils from different production basins were obtained analyzing the maltene fraction of the oils by comprehensive two-dimensional gas chromatography coupled to quadrupole mass spectrometry (GC×GC-QMS), and performing multiway principal component analysis (MPCA) of the chromatographic data. The results showed that four MPC explained 93.57% of the data variance, expressing mainly the differences on the profiles of the saturated hydrocarbon fraction of the oils (C13-C18 and C19-C30n-alkanes and the pristane/phytane ratio). The MPC1 grouped the samples severely biodegraded oils, while the type of the depositional paleoenvironments of the oils and its oxidation conditions (as well as their thermal maturity) could be inferred analysing others relevant MPC. Additionally, considerations about the source of the oil samples was also possible based on the overall distribution of relevant biomarkers such as the phenanthrene derivatives, tri-, tetra- and pentacyclic terpanes.

Characterization of crude oil biomarkers using comprehensive two-dimensional gas chromatography coupled to tandem mass spectrometry

Oil samples from Recôncavo basin (NE Brazil), previously analyzed by traditional techniques such as gas chromatography coupled to tandem mass spectrometry, were evaluated using comprehensive two-dimensional gas chromatography coupled to quadrupole mass spectrometry and comprehensive two-dimensional gas chromatography coupled to tandem mass spectrometry along with simplified methods of samples preparation to evaluate the differences and advantages of these analytical techniques to better understand the development of the organic matter in this basin without altering the normal distribution of the compounds in the samples. As a result, the geochemical parameters calculated by comprehensive two-dimensional gas chromatography coupled to tandem mass spectrometry described better the origin, maturity, and biodegradation of both samples probably by increased selectivity, resolution, and sensitivity inherent of the multidimensional technique. Additionally, the detection of the compounds such as, the C(14α-) homo-26-nor-17α-hopane series, diamoretanes, nor-spergulanes, C19 -C26 A-nor-steranes and 4α-methylsteranes resolved and detected by comprehensive two-dimensional gas chromatography coupled to tandem mass spectrometry were key to classify and differentiate these lacustrine samples according to their maturity and deposition conditions.

Exploratory Analysis of Biodiesel by Combining Comprehensive Two-Dimensional Gas Chromatography and Multiway Principal Component Analysis

Comprehensive two-dimensional gas chromatography (GC×GC) was used in this work in order to reveal and identify minor compounds of several raw materials in biodiesel samples, which are not detected by conventional GC analysis. Multiway principal component analysis (MPCA) of the chromatographic profile allowed to identify compounds as C15:1n-3, C18:2n-6, C20:1n-6, C20:3n-3, C20:3n-6, C20:4n-6 C20:5n-3, C21:1n-6, C22:1n-9, C23:1n-9, C24:1n-9 multiunsaturated which were useful tracers to discriminate between biodiesel samples. In this way, the separation power of GC×GC combined with the MPCA algorithm proved to be a valuable strategy for biodiesel samples classification, allowing identification and providing additional compounds of the commonly known as chemical profile of biodiesel samples.

The constant evolution of science and the growing demand for new technologies have led to new techniques in instrumentation that can improve detection, separation, resolution, and peak capacity. Comprehensive two-dimensional liquid chromatography (LC×LC) is presented as a powerful tool in complex sample analyses. During an analysis, a sample is subjected to two independent separation mechanisms that are combined, resulting in increased resolving power. For appropriate application of LC×LC, understanding the influence of parameters that require optimization is necessary. The main purpose of optimization is to predict the combination of stationary phases, separation conditions, and instrumental requirements to obtain the best separation performance. This review discusses theoretical, intrumental, and chemometric aspects of LC×LC and focuses on its applications in foods. It aims to provide a clear understanding of the aspects that can be used as strategies in the optimization of this analytical method.

In vitro effects of Crotalus atrox snake venom on chick and mouse neuromuscular preparations

The neuromuscular effect of venoms is not a major clinical manifestation shared between rattlesnakes native to the Americas, which showed two different venom phenotypes. Taking into account this dichotomy, nerve muscle preparations from mice and chicks were used to investigate the ability of Crotalus atrox venom to induce in vitro neurotoxicity and myotoxicity. Unlike crotalic venoms of South America, low concentrations of C. atrox venom did not result in significant effects on mouse neuromuscular preparations. The venom was more active on avian nerve-muscle, showing reduction of twitch heights after 120 min of incubation with 10, 30 and 100 μg/mL of venom with diminished responses to agonists and KCl. Histological analysis highlighted that C. atrox was myotoxic in both species of experimental animals; as evidenced by degenerative events, including edematous cells, delta lesions, hypercontracted fibers and muscle necrosis, which can lead to neurotoxic action. These results provide key insights into the myotoxicity and low neurotoxicity of C. atrox in two animal models, corroborating with previous genomic and proteomic findings and would be useful for a deeper understanding of venom evolution in snakes belonging to the genus Crotalus.

Comprehensive two-dimensional gas chromatography–mass spectrometry combined with multivariate data analysis for pattern recognition in Ecuadorian spirits

The current methodology used in quality control of Ecuadorian beverages such as Pájaro azúl, Puro and Pata de vaca is carried out by using conventional gas chromatography; however, it does not allow the fingerprinting of these Ecuadorian spirit beverages and their possible cases of adulteration. In order to overcome this drawback, comprehensive two-dimensional gas chromatography–mass spectrometry (GC × GC–MS) was combined with multivariate data analysis, revealing that compounds like citronellal, citronellol, geraniol, methyl anthranilate, (−)-trans-α-bergamotene, (−)-cis-α-bergamotene and d-limonene can be considered key elements for pattern recognition of these traditional beverages and product adulteration cases. Thus, the two-dimensional chromatographic fingerprints obtained by GC × GC–MS coupled with chemometric analysis, using Principal Component Analysis and Fisher-ratio can be considered as a potential strategy for adulteration recognition, and it may used as a quality assurance system for Ecuadorian traditional spirits.
