Abstract The ratio tissue-type to total ferritin was estimated in serum of 15 cancer patients, 7 normal subjects, as well as in 7 tumour cytosols, after absorption to Con A-Sepharose. As expected, in normal serum the larger part corresponded to serum-type, glycosylated ferritin, and in tumour cytosols the larger part corresponded to tissue-type, non-glycosylated ferritin. In cancer patient sera however, a significant part of tissue-type, non-glycosylated ferritin was constantly found. This observation suggests the ferritinaemia in cancer patients to be of a mixed origin, with tissue-type ferritin release by damaged cells or abnormal secretion.

Abstract In order to assess the practical influence of the higher specificity of monoclonal antibodies in ferritin assays, different samples of both normal subjects and cancer patients were measured with two different ferritin assay methods, the first using a polyclonal antibody directed against liver ferritin, the other using a monoclonal antibody against spleen ferritin. Samples included serum, red blood cells and tumour cytosol. The monoclonal antibody assay was found to yield statistically significant lower results for all sample types, except for serum in normal subjects. This difference is attributed to the non-recognition of acidic ferritins by this monoclonal antibody assay.
