Experimental infection of lambs with C and S-type strains of Mycobacterium avium subspecies paratuberculosis: immunological and pathological findings

The two main genotypes of recognized isolates of Mycobacterium avium subsp. paratuberculosis (Map) are cattle (C) and sheep (S) strains. An experimental infection was conducted to establish the effect of Map strain on the pathogenesis of ovine paratuberculosis. Twenty-four out of thirty 1.5-month-old Assaf lambs were divided into 4 groups of 6 and infected orally with three low passage field isolates, two of S- (22G and the pigmented Ovicap49) and one of C– (764) type, and the reference K-10 strain (C type). The remaining six animals were unchallenged controls. Animals were euthanized at 150 and 390 days post-infection (dpi). Throughout the experiment, the peripheral immune response was assessed and histological and molecular (PCR) studies were conducted on samples of intestine and related lymphoid tissue. Specific antibody and IFN-γ production was significantly higher in animals infected with the C strains, while no consistent IFN- γ responses were observed in the S-type strain infected groups. A positive intradermal skin test response was detected in all infected groups. Lambs infected with S-type strains had granulomatous lesions restricted to the lymphoid tissue with no differences in the lesion intensity over time. In both C–type strain groups, lesions were more severe at 150 dpi while at 390 dpi lesions, characterized by well-demarcated granulomas with fibrosis, decreased in severity. Only infected lambs were positive to PCR. These results suggest that the strain of Map has a strong influence over the immune and pathological responses developed by the host. Lesions induced by C–type strains in lambs show a regressive character and tend to decrease as the infection progresses.

Placental thrombosis in acute phase abortions during experimental Toxoplasma gondii infection in sheep

After oral administration of ewes during mid gestation with 2000 freshly prepared sporulated oocysts of T. gondii isolate M4, abortions occurred between days 7 and 11 in 91.6% of pregnant and infected ewes. Afterwards, a further infection was carried out at late gestation in another group of sheep with 500 sporulated oocysts. Abortions happened again between days 9 and 11 post infection (pi) in 58.3% of the infected ewes. Classically, abortions in natural and experimental ovine toxoplasmosis usually occur one month after infection. Few experimental studies have reported the so-called acute phase abortions as early as 7 to 14 days after oral inoculation of oocysts, and pyrexia was proposed to be responsible for abortion, although the underline mechanism was not elucidated. In the present study, all placentas analysed from ewes suffering acute phase abortions showed infarcts and thrombosis in the caruncullar villi of the placentomes and ischemic lesions (periventricular leukomalacia) in the brain of some foetuses. The parasite was identified by PCR in samples from some placentomes of only one sheep, and no antigen was detected by immunohistochemical labelling. These findings suggest that the vascular lesions found in the placenta, and the consequent hypoxic damage to the foetus, could be associated to the occurrence of acute phase abortions. Although the pathogenesis of these lesions remains to be determined, the infectious dose or virulence of the isolate may play a role in their development.

Systemic and local immune responses in sheep after Neospora caninum experimental infection at early, mid and late gestation

Besides its importance in cattle, Neospora caninum may also pose a high risk as abortifacient for small ruminants. We have recently demonstrated that the outcome of experimental infection of pregnant sheep with 106 Nc-Spain7 tachyzoites is strongly dependent on the time of gestation. In the current study, we assessed peripheral and local immune response in those animals. Serological analysis revealed earlier and higher IFN-γ and IgG responses in ewes infected at early (G1) and mid (G2) gestation, when abortion occurred. IL-4 was not detected in sera from any sheep. Inflammatory infiltrates in the placenta mainly consisted of CD8+ and, to a lesser extent, CD4+ T cells and macrophages (CD163+). The infiltrate was more intense in sheep infected at mid-gestation. In the foetal mesenchyme, mostly free tachyzoites were found in animals infected at G1, while those infected in G2 displayed predominantly particulate antigen, and parasitophorous vacuoles were detected in sheep infected at G3. A similar pattern of placental cytokine mRNA expression was found in all groups, displaying a strengthened upregulation of IFN-γ and IL-4 and milder increases of TNF-α and IL-10, reminiscent of a mixed Th1 and Th2 response. IL-12 and IL-6 were only slightly upregulated in G2, and TGF-β was downregulated in G1 and G2, suggestive of limited T regulatory (Treg) cell activity. No significant expression of TLR2 or TLR4 could be detected. In summary, this study confirms the pivotal role of systemic and local immune responses at different times of gestation during N. caninum infection in sheep.

Macrophage Subsets Within Granulomatous Intestinal Lesions in Bovine Paratuberculosis

Animals infected with Mycobacterium avium subspecies paratuberculosis show a variety of granulomatous lesions that range from focal forms, seen in the subclinical stages, to diffuse lesions associated with clinical signs. The aim of this study was to phenotypically characterize the macrophages present in the different lesion types using immunohistochemical methods. Lesions from a total of 23 animals with bovine paratuberculosis, natural and experimental, were examined by immunohistochemistry. Antibodies against inducible nitric oxide synthase (iNOS), tumor necrosis factor α (TNF-α), CD163, interleukin 10 (IL-10), transforming growth factor β (TGF-β), major histocompatibility complex (MHC) class II, natural resistance-associated macrophage protein 1 (Nramp-1), calprotectin, Ki-67, CD68, lysozyme, and ionized calcium-binding adaptor molecule 1 (Iba-1) molecules were employed. Samples were scored semiquantitatively using a complete histological score (H-score), reflecting the staining intensity and the percentage of immunolabeled macrophages. Differences in the H-score were seen depending on the lesion type. In focal lesions, with none or few acid-fast bacilli (AFB), macrophages were polarized toward M1 phenotype, with high H-scores for iNOS and TNF-α. Diffuse multibacillary lesions showed M2 differentiation, with high expression of CD163, IL-10, and TGF-β as well as Nramp-1 and MHC class II antigens. Macrophages in diffuse paucibacillary forms showed high H-scores for iNOS but low ones for TNF-α. Diffuse lesions, either multibacillary or paucibacillary, showed high calprotectin and low Ki-67 expression, suggesting a progressive character, while focal forms, with low H-scores for these antigens, would be consistent with latency. Lysozyme and CD68 expression were related to the amount of AFB. H-score for Iba-1 antibody was similar among all types. The findings of this study provide insights into the polarization status of macrophages and lesion development in bovine paratuberculosis.

Experimental ovine toxoplasmosis: influence of the gestational stage on the clinical course, lesion development and parasite distribution

The relation between gestational age and foetal death risk in ovine toxoplasmosis is already known, but the mechanisms involved are not yet clear. In order to study how the stage of gestation influences these mechanisms, pregnant sheep of the same age and genetic background were orally dosed with 50 oocysts of Toxoplasma gondii (M4 isolate) at days 40 (G1), 90 (G2) and 120 (G3) of gestation. In each group, four animals were culled on the second, third and fourth week post infection (pi) in order to evaluate parasite load and distribution, and lesions in target organs. Ewes from G1 showed a longer period of hyperthermia than the other groups. Abortions occurred in all groups. While in G2 they were more frequent during the acute phase of the disease, in G3 they mainly occurred after day 20 pi. After challenge, parasite and lesions in the placentas and foetuses were detected from day 19 pi in G3 while in G2 or G1 they were only detected at day 26 pi. However, after initial detection at day 19 pi, parasite burden, measured through RT-PCR, in placenta or foetus of G3 did not increase significantly and, at in the third week pi it was lower than that measured in foetal liver or placenta from G1 to G3 respectively. These results show that the period of gestation clearly influences the parasite multiplication and development of lesions in the placenta and foetus and, as a consequence, the clinical course in ovine toxoplasmosis.

Safety and efficacy of the bumped kinase inhibitor BKI-1553 in pregnant sheep experimentally infected with Neospora caninum tachyzoites

Neospora caninum is one of the main causes of abortion in cattle, and recent studies have highlighted its relevance as an abortifacient in small ruminants. Vaccines or drugs for the control of neosporosis are lacking. Bumped kinase inhibitors (BKIs), which are ATP-competitive inhibitors of calcium dependent protein kinase 1 (CDPK1), were shown to be highly efficacious against several apicomplexan parasites in vitro and in laboratory animal models. We here present the pharmacokinetics, safety and efficacy of BKI-1553 in pregnant ewes and foetuses using a pregnant sheep model of N. caninum infection. BKI-1553 showed exposure in pregnant ewes with trough concentrations of approximately 4 μM, and of 1  μM in foetuses. Subcutaneous BKI-1553 administration increased rectal temperatures shortly after treatment, and resulted in dermal nodules triggering a slight monocytosis after repeated doses at short intervals. BKI-1553 treatment decreased fever in infected pregnant ewes already after two applications, resulted in a 37–50% reduction in foetal mortality, and modulated immune responses; IFNγ levels were increased early after infection and IgG levels were reduced subsequently. N. caninum was abundantly found in placental tissues; however, parasite detection in foetal brain tissue decreased from 94% in the infected/untreated group to 69–71% in the treated groups. In summary, BKI-1553 confers partial protection against abortion in a ruminant experimental model of N. caninum infection during pregnancy. In addition, reduced parasite detection, parasite load and lesions in foetal brains were observed.

Immunohistochemical study and mRNA cytokine profile of the local immune response in cattle naturally infected with Calicophoron daubneyi

In order to recognize the local immune response of the definitive host to Calicophoron daubneyi natural infection, an immunohistochemical study was carried out in the reticulum and rumen in 49 naturally infected cattle. The role of cytokines (IL-4 and IL-10 interleukins and IFN-γ) in the activation of specific defence mechanisms was evaluated by reverse transcriptase quantitative polymerase chain reaction (RT-qPCR) assays to study cytokine mRNA expression. In all infected animals, CD3+ T lymphocytes seemed to be the main element of the inflammatory infiltrate in the reticular and ruminal lamina propria at the point of the parasite adhesion. Intraepithelial globule leukocytes also showed immunolabelling for CD3. Most CD3+ cells also expressed CD4 (T cell helper) antigen although sporadic CD8+-cytotoxic lymphocytes were observed. Local expression of IFN-γ was observed in damaged papillae at the site of parasite attachment and in scattered cells in the lamina propria. B cells (CD79αcy+, CD45+ and IgG+) were found constantly in relation to lymphoid aggregates. MAC387 was expressed in squamous epithelium and in macrophages of the lamina propria of affected papillae. Macrophages in this location also stained positively for CD163 and CD68. Intraepithelial Langerhans cells and macrophages located in the lamina propria showed immunopositivity for MHCII in the affected areas. RT-qPCR analysis confirmed a statistical significant increase of IFN-γ, and IL-10 expression (p<0.01) in the rumen associated with the presence of flukes. These findings suggest a predominant Th1 polarized local immune response with the probable involvement of Th regulatory cells in cattle C. daubneyi natural infection.

Immunohistochemical expression of interferon-γ in different types of granulomatous lesions associated with bovine paratuberculosis

Animals infected with Mycobacterium avium subsp. paratuberculosis (Map) show a variety of lesions, from focal forms, seen in subclinical stages to diffuse lesions in clinical cases. The purpose of this study was to evaluate the local expression of IFN-γ by immunohistochemistry in relation with the type of lesion in naturally Map-infected cows. The number of immunolabelled cells, -the majority morphologically consistent with lymphocytes-, was higher in focal and diffuse paucibacillary forms than in diffuse multibacillary lesions, where they appeared closely related to epithelioid cells. Diffuse multibacillary lesions had the lowest numbers, but higher than controls, and positive cells were intermingled among the macrophages. The peripheral IFN-γ production was higher in all Map infected cows and a positive correlation was found with the number of immunolabelled cells in the intestine. The findings of this study show that IFN-γ would play a role in the development of the different types of lesions in paratuberculosis, and also points out the importance of adequate sampling of lymphoid tissue containing samples when studying the local immune response in which IFN-γ expression may be involved, especially in cases where focal lesions are present.

Virulence attenuation of a Mycobacterium avium subspecies paratuberculosis S-type strain prepared from intestinal mucosa after bacterial culture. Evaluation in an experimental ovine model

The differences in pathogenicity between an inoculum derived directly from an intestinal tissue homogenate from a paratuberculosis affected sheep and the S-type Mycobacterium avium subsp. partuberculosis (Map) strain isolated in laboratory media from the mentioned homogenate were assessed in two experiments in lambs. Specific peripheral immune responses were significantly lower in animals inoculated with the cultured organisms that showed only granulomatous lesions in the intestinal lymphoid tissue. However, in the homogenate group, more abundant granulomata also occurred in the lamina propria. Map was isolated only in lambs infected with the culture strain. Map DNA was demonstrated by nested-PCR in all the lambs but in a lower proportion (57.1% vs 100%) in those from the culture group. Under these particular experimental conditions, the results suggest that an attenuation of Map virulence has occurred in the cultured strain compared to the initial tissue homogenate, even after a low number of passages.

Cyber insurance products have recently emerged with the potential of lowering the impact of cyberspace risks. However, they have yet to mature. In this paper we present several risk analysis models that may facilitate the implementation and adoption of cyber insurance. Our model develops the Data Specification on Protected Sites , part of the INSPIRE Annex I. Hence its orientation towards georeferenced heritage data. The data model has been developed by an interdisciplinary group made up of specialists in the field of Geomatics and Cultural Heritage. It is our aim to achieve a generic, extendable and interoperable schema. It should be generic enough to embrace all kind of protected heritage data, from an ancient pilgrim’s way to the last artifact found in an archaeological excavation, extendable to allow any kind of data producer to adapt the model to the nature of their own information and interoperable to combine spatial data sets from different sources through network services, via Internet. The achievement of these three characteristics features international norms and standards referred to our kind of data. This implies adaptation to INSPIRE as well as to several ISO norms: ISO 19100 series regarding geographical information, ISO 21127 (CIDOC-CRM Model) for heritage thematic data and ISO 15836 (Dublin Core) for document resources. The data model comprehends two main dimensions: cultural entities in a strict sense, and the legislative figures created to protect them. This allows for the representation of cultural objects (i.e. historical buildings or archaeological sites) and their link to their legislative protection, keeping them as separate realities. In order to describe the data model, an international common language has been used: UML (Unified Modeling Language), a standard itself. Este aspecto, unido a las restantes opciones de parametrizacion de los diferentes modulos, permite liberar al disenador de los detalles especificos de las implementaciones hardware asi como adaptar el consumo de recursos del FPGA a las necesidades de la aplicacion. Su transformacion en glucosa se considera la etapa mas importante en la produccion de biocombustibles a partir de biomasa lignocelulosica. Para esta transformacion es necesaria la accion sinergica de tres tipos de enzimas que hidrolizan los enlaces s-1,4 de la celulosa: (i) las endoglucanasas, que actuan al azar sobre enlaces internos, (ii) las celobiohidrolasas, que operan progresivamente por los extremos reductores y no reductores de la cadena, y (iii) las s-glucosidasas (BGL), que hidrolizan celobiosa y los celooligosacaridos mas pequenos hasta glucosa. Los hongos filamentosos son la principal fuente de celulasas, siendo los crudos del genero Trichoderma los mas estudiados y comercializados historicamente. Sin embargo, estos hongos secretan niveles insuficientes de BGL para una conversion efectiva de la celulosa, por lo que con frecuencia requieren ser suplementados con preparaciones ricas en esta enzima procedentes de otros hongos. Recientemente, se han descrito cepas de Penicillium con gran capacidad para secretar altos niveles de BGL al medio extracelular. Ademas de su papel en la hidrolisis de la celulosa las BGL tambien pueden emplearse para sintetizar compuestos de interes industrial mediante reacciones de transglicosilacion. En este trabajo se ha estudiado una nueva cepa fungica productora de celulasas, identificada como Talaromyces amestolkiae, en base a un analisis molecular y morfologico. Se han purificado a homogeneidad electroforetica tres BGL (BGL-1, BGL-2 y BGL-3) secretadas por el hongo y se han caracterizado bioquimicamente. BGL-1 y BGL-2 son proteinas monomericas, mientras que BGL-3 es un dimero funcional. Los valores de actividad maxima de estas enzimas se obtuvieron a pH 4,0 y entre 50-60 oC, siendo estables en un rango de pH de 4-7 y a 50 oC. Las tres mostraron distinto comportamiento en funcion del sustrato (pNPG o celobiosa), ensayandose tambien el efecto de determinados compuestos quimicos e inhibidores en su actividad. Las tres pueden hidrolizar celooligosacaridos de diferente longitud, disminuyendo la eficacia de hidrolisis con el aumento de la polimerizacion, y no son activas frente a polisacaridos. Ademas, mostraron actividad de transglicosilacion, formando alquilglicosidos y celooligosacaridos de mayor longitud que los usados como sustratos. Las tres BGL se identificaron mediante huella peptidica y en base la alta homologia con otras BGL fungicas relacionadas filogeneticamente, se disenaron cebadores especificos que permitieron la secuenciacion de los genes que codifican cada una de ellas. El analisis de las secuencias aminoacidicas mostro que BGL-1 es miembro de la familia 1 de las glicosil hidrolasas, mientras que BGL-2 y BGL-3 pertenecen a la familia 3. Teniendo en cuenta que existen pocas BGL fungicas cristalizadas, se construyeron modelos moleculares de estas ultimas en base a las estructuras con las que presentaron mayor identidad. El crudo enzimatico de T. amestolkiae, rico en BGL, se uso de forma individual y como suplemento de otros cocteles comerciales en experimentos de sacarificacion a partir de slurry acido de paja de trigo. Los resultados obtenidos indican que el hongo secreta, ademas de celulasas, otras enzimas complementarias que potencian la liberacion de azucares fermentables. Se analizo el secretoma de T. amestolkiae usando Avicel o slurry acido de paja de trigo como fuente de carbono. Las celulasas fueron las enzimas mas abundantes en ambas condiciones, sin embargo, en el crudo obtenido a partir de los cultivos en slurry aumento la proporcion de BGL y otras enzimas distintas a las celulasas, indicando que para la degradacion de un sustrato complejo es necesaria mayor diversidad enzimatica. El olivo (Olea europaea L.) es el cultivo lenoso no tropical que ocupa mayor superficie en todo el mundo, con el 95% de la produccion mundial localizada en la cuenca mediterranea. Espana es el pais con mayor superficie de olivar del mundo con 2,6 MHa y aproximadamente el 39% de la produccion mundial. La Verticilosis, causada por el hongo de suelo Verticillium dahliae Kleb, constituye la principal amenaza para el olivar. Esta enfermedad afecta al olivo en todos los paises de tradicion olivarera y causa importantes perdidas de rendimiento y muerte de los arboles (Jimenez-Diaz y col., 2012). Este patogeno coloniza el sistema vascular de la planta, bloqueando el flujo del agua y finalmente induciendo estres hidrico (Van Alfen, 1989).Espana es el mayor productor de citricos de la Union Europea y el quinto a nivel mundial, con una produccion anual superior a cinco millones de toneladas (MAGRAMA, 2016). Este sector genera una gran cantidad de subproductos, con un elevado aporte energetico y alto contenido en pectinas y azucares, por lo que podrian usarse como materias primas en dietas para rumiantes. Sin embargo, los subproductos de citricos son muy diversos, por lo que su composicion quimica y valor nutritivo tambien puede ser variable (FEDNA, 2010; Feedipedia, 2016). Due to its relevance for the population, the India Meteorological Department has dated the onset over the southern tip of the Indian Peninsula (Kerala) since 1901. Estos efectos se producen fundamentalmente a traves de su relacion, directa o indirecta, con la arquitectura del racimo, el comportamiento reproductivo y el grado o modo de exposicion de las bayas al ambiente. Respecto a los dos primeros, en un estudio previo, determinamos que tanto la longitud del raquis y sus primeras ramas como el numero de bayas por racimo son los caracteres con mayor incidencia en la compacidad entre los analizados. Ahora bien, el estudio de la base genetica responsable del numero de bayas requiere de su diseccion previa, dado que es una variable secundaria, resultado del numero de flores por inflorescencia y de la tasa de cuajado o conversion de flor en fruto. En este trabajo se presenta un estudio preliminar en el que se han caracterizado fenotipicamente 104 variedades de vid para estudiar su comportamiento reproductivo. Asi, sobre una base amplia de diversidad varietal, se han estimado variables como el numero de flores, la tasa de cuajado, corrimiento, millerandage, etc., evaluando su posible relacion con la compacidad del racimo. Este trabajo, una vez completado durante varios anos, permitira conocer la diversidad existente para el comportamiento reproductivo en la vid, asi como, de forma mas concreta, abordar la caracterizacion de los procesos geneticos responsables de las diferencias fenotipicas detectadas para el comportamiento reproductivo y sus elementos individuales y para la compacidad del racimo, identificando los posibles genes involucrados en dichos procesos.Trabajo presentado en la 11th International Conference on Grapevine Breeding and Genetics, celebrada en Pekin del 29 de julio al 2 de agosto de 2014.The Polarimetric and Helioseismic Imager (PHI) is the first deep-space solar spectropolarimeter, on-board the Solar Orbiter (SO) space mission. It faces: stringent requirements on science data accuracy, a dynamic environment, and severe limitations on telemetry volume. SO/PHI overcomes these restrictions through on-board instrument calibration and science data reduction, using dedicated firmware in FPGAs. This contribution analyses the accuracy of a data processing pipeline by comparing the results obtained with SO/PHI hardware to a reference from a ground computer. Todos los registros conocidos hasta el momento pertenecen al Ordovícico Medio-Superior (Darriwiliense-Katiense) y las asociaciones presentan gran diversidad aunque en general los taxones están poco o mal conocidos. La mayor parte de los yacimientos se concentran en las regiones de Amêndoa-Mação y Buçaco, aunque también existen citas de equinodermos en Arouca, Dornes, Moncorvo y Valongo. Por último se comentan futuras líneas de trabajo que ayudarán a comprender mejor estas faunas, perfeccionar y completar el conocimiento del registro fósil del grupo en el Ordovícico centroibérico.
