Abstract Background. The aim of this study was to determine the effect of Finnish sauna as a regeneration method post-exercise on the oxidant-antioxidant balance in healthy men. Material. 43 men aged 24.0 ± 4.3 years performed a 30-min aerobic exercise on a cycle ergometer and rested for 39 min at a room temperature (Day 1; 20°C) or in a sauna for post-workout recovery (Day 2; 90°C, air humidity 10%). Blood was taken 3 times during both study days: Before the exercise (baseline), 20 and 40 min after the recovery. Methods. The activity of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) was determined in the subjects’ erythrocytes. The concentration of thiobarbituric acid reactive substances (TBARS) was measured both in plasma (TBARSpl) and in the erythrocytes (TBARSer). Results. A 12.7% increase in the TBARSpl concentration versus the baseline was observed 40 min after the Finnish sauna (p < 0.01). The CAT activity observed 20 and 40 min after the sauna was also found higher by 8.1% and 8.9%, respectively, in comparison with the baseline (p < 0.05). In turn, the TBARSer concentration was lower by 17.5% 40 min after the recovery in the sauna, as compared with the TBARSer concentration 40 min after the recovery at the room temperature (p < 0.05). Conclusions. A single Finnish sauna bath as a source of free radicals per se is able to reduce oxidative stress induced by a 30-min aerobic exercise in healthy men.

The influence of exercise combined with whole-body cryotherapy (WBC) on the oxidant/antioxidant balance in healthy men was assessed. The study included 16 kayakers of the Polish National Team, aged 22.7 ± 2.6, subjected to WBC (−120°C–−145°C; 3 min) twice a day for the first 10 days of a 19-day physical training cycle: pre exercise morning stimulation and post exercise afternoon recovery. Blood samples were taken on Day 0 (baseline) and on Days 5, 11 and 19. The serum concentration of malondialdehyde (MDA), conjugated dienes (CD), thiobarbituric acid reactive substances (TBARS), protein carbonyls, vitamin E, urea, cortisol, and testosterone were determined, along with the glutathione peroxidase (GPx) activity, the total antioxidant capacity (TAC), and morphological blood parameters. On 5th day of exercise/WBC, the baseline GPx activity decreased by 15.1% (P < 0.05), while on 19th day, it increased by 19.7% (P < 0.05) versus Day 5. On Day 19 TBARS concentration decreased versus baseline and Day 5 (by 15.9% and 17.4%, resp.; P < 0.01). On 19 Day urea concentration also decreased versus 11 Day; however, on 5th and 11th days the level was higher versus baseline. Combining exercise during longer training cycles with WBC may be advantageous.

Background. The aim of this study was to determine the effects of exposure to cold and heat on the activity of selected lysosomal enzymes as well as on the activity of the protease inhibitor, which are all considered to be markers of cellular damage. Material. Two groups of healthy volunteers were included in the study. The first group consisted of experienced winter swimmers who practiced bathing in ice-cold water once a week, while the other group was comprised of persons who had never taken part in winter swimming before. During the experiment all the participants bathed in a river with a water temperature of 0°C. The same volunteers were later subjected to a sauna bath at an air temperature of 85°C. Methods. The activity of cathepsin D, alpha-1-antitrypsin, arylsulphatase and acid phosphatase was measured in the participants blood serum. Results. After exposure to cold water no changes in the parameters studied could be found. However, after the sauna an increase in the activity of arylsulphatase and alfa-1-antitrypsin accompanied by a decrease of cathepsin D activity could be observed. Conclusions. Winter swimming seems to have no effect on the activity of the lysosomal enzymes. By contrast, an increase of certain lysosomal enzymes after the sauna suggests that it may be deleterious to the lysosomal membranes. Furthermore it seems that regular winter swimming combined with sauna, according to hormesis theory, induces some adaptive response.

The aim of the study was to determine the effect of a 5 min head-out ice-cold water bath on the oxidant-antioxidant balance in response to exercise. The crossover study included the subjects (n = 24; aged 28.7 ± 7.3 years) who performed two identical stationary cycling bouts for 30 min and recovered for 10 min at room temperature (RT = 20°C; session 1) or in a pool with ice-cold water (ICW = 3°C, 5 min immersion; session 2). The concentration of thiobarbituric acid reactive substances (TBARS) in blood plasma (TBARSpl) and erythrocytes (TBARSer) and the erythrocytic activity of superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) were measured three times during each of the two study sessions: before the exercise (baseline) and 20 and 40 min after the appropriate recovery session. Lower concentration of TBARSpl 40 min after postexercise recovery in ICW was revealed as compared with that after recovery at RT (P < 0.05). Moreover, a statistically significant postexercise increase in the TBARSpl and TBARSer concentrations was found (P < 0.01 and P < 0.05, resp.). A short-term ice-cold water bath decreases postexercise lipid peroxidation.

Obesity is one of the major global health problems. Melatonin deficiency has been demonstrated to correlate with obesity. The aim of the study was to estimate the effect of melatonin on oxidative stress and adipokine levels in obese patients on a calorie-restricted diet. Thirty obese patients were supplemented with a daily dose of 10 mg of melatonin (n = 15) or placebo (n = 15) for 30 days with a calorie-restricted diet. Serum levels of melatonin, 4-hydroxynonenal (HNE), adiponectin, omentin-1, leptin, and resistin, as well as erythrocytic malondialdehyde (MDA) concentration and Zn/Cu-superoxide dismutase, catalase, and glutathione peroxidase (GPx) activities, were measured at baseline and after supplementation. Significant body weight reduction was observed only in the melatonin group. After melatonin supplementation, the adiponectin and omentin-1 levels and GPx activities statistically increased, whereas the MDA concentrations were reduced. In the placebo group, a significant rise in the HNE and a drop in the melatonin concentrations were found. The results show evidence of increased oxidative stress accompanying calorie restriction. Melatonin supplementation facilitated body weight reduction, improved the antioxidant defense, and regulated adipokine secretion. The findings strongly suggest that melatonin should be considered in obesity management. This trial is registered with CTRI/2017/07/009093.

The AlterG antigravity treadmill allows running with a considerable weight reduction. Physical exercise practiced on this treadmill is an innovative method supporting the treatment of injuries in sports and rehabilitation of patients. The aim of the study was to investigate the effect of a 30 min run on the AlterG treadmill with 80% body weight reduction comparing the effect to the similar effort on the classic treadmill on the redox equilibrium and the activity of selected lysosomal enzymes and a serine protease inhibitor in the blood of amateur minor female volleyball players. Venous blood samples were taken before the exercise and 30 minutes and 24 hours after its completion. The obtained results were analysed using Tukeys test and Pearsons linear correlations were calculated. 24 h after the running test on classic treadmill, the erythrocytic superoxide dismutase activity was higher than before and 30 min after it, as well as compared to the run on AlterG treadmill (p < 0.001). The erythrocytic-conjugated diene concentration 24 h after the exercise on the classic treadmill was meaningly higher compared to that after the exercise on the AlterG treadmill (p < 0.001). The cathepsin D activity was significantly lower after the exercise in AlterG conditions compared to the baseline value and that measured after the exercise on classic treadmill (p < 0.001). It seems that the exercise on the AlterG treadmill keeps the oxidant–antioxidant equilibrium and stabilizes lysosomal membranes in young, physically active women in contrast to the exercise on the classic treadmill. Abstract The activity of selected antioxidant enzymes: superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) in erythrocytes, and the concentration of thiobarbituric acid reactive substances (TBARS) in blood plasma and erythrocytes, were determined in people subjected to hyperbaric oxygen (HBO) therapy due to sudden sensorineural hearing loss (SSNHL). Venous blood samples were taken immediately before entering the hyperbaric chamber and 5 min after leaving it. In the study group, two age subgroups were distinguished: group I consisting of subjects under 35 and group II consisting of subjects over 50. The obtained values were analysed statistically using Student’s t-test. Differences were considered as statistically significant at p < 0.05. A statistically significant decrease in the CAT activity was shown 5 min after leaving the hyperbaric chamber in pooled subjects (p < 0.01) and group I (p < 0.05). Furthermore, a statistically significant decrease in the erythrocyte TBARS concentration was observed in group II (p < 0.05). It was demonstrated that a single exposure to hyperbaric oxygen affects the oxidant-antioxidant equilibrium as evidenced by, e.g., a statistically significant decrease in the activity of catalase in erythrocytes. It is possible that the antioxidant response to HBO depends on the age of subjects.

The effect of an ice-cold water (ICW) bath as a recovery intervention from aerobic exercise on the oxidant-antioxidant balance in healthy ice swimmers was determined. Twenty ice swimmers aged 31.2 ± 6.3 years performed a 30-min cycloergometer exercise test at room temperature (20°C, RT), followed by recovery at RT or in a pool of ice-cold water (ICW bath, 3°C, 5 min). Blood for laboratory assays was collected from the basilic vein two times: before the exercise (baseline) and 40 min after the RT or ICW recovery. The concentrations of plasma and erythrocytic thiobarbituric acid reactive substances (plTBARS and erTBARS, respectively), serum concentrations of 8-iso-prostaglandin F2α, 4-hydroxynonenal and malondialdehyde, along with the erythrocytic activities of catalase (CAT) and superoxide dismutase (SOD), as well as the serum level of total antioxidant capacity, were assessed. No statistically significant changes were observed. However, a statistically significant negative linear correlation between the erTBARS concentration and the SOD activity was found 40 min after the combination of exercise/RT recovery (r=-0.571, P<0.01). The baseline CAT and SOD activities were also linearly correlated (r=0.469, P<0.05). Both the 5-min ICW bath and the 30-min aerobic exercise have practically no impact on the oxidant-antioxidant balance in healthy ice swimmers.

The activity of α 1-antitrypsin (AAT) and the lysosomal enzymes, cathepsin D (CTS D), arylsulfatase (ASA), and acid phosphatase, (AcP) was determined in patients with COPD (GOLD category A). Moreover, the diagnostic usefulness of these parameters in blood serum was assessed along with establishing whether smoking cessation affects these parameters. The study included 70 patients with COPD who ceased smoking (study group) and two control groups of 33 subjects each: nonsmokers without COPD (control I) and patients with COPD who continued smoking (control II). In control I, blood was taken once and in control II, at the start of the experiment and after the 1st, 2nd, and 3rd months. AAT in the patients exhibited higher activity than in the healthy subjects at all time points. AAT activity in the patients before the start of the experiment was ~80% higher (P < 0.001) than in control I. No statistically significant differences in CTS D, ASA, and AcP activity were found. COPD involves increased AAT activity and unchanged activities of the assessed lysosomal enzymes. Three-month tobacco abstinence does not affect these parameters in peripheral blood. Determining the AAT levels in blood serum can be used in the diagnostics of COPD.

Zmiana warunków otoczenia jest stresem dla organizmu, który może prowadzić do wzmożonej generacji reaktywnych form tlenu i licznych uszkodzeń w obrębie komórek. Istotną rolę w przeciwdziałaniu tym uszkodzeniom pełnią enzymy antyoksydacyjne. Celem niniejszej pracy było określenie wpływu zmian ciśnienia otoczenia w warunkach nurkowania na aktywność peroksydazy glutationowej oraz katalazy. W badaniu wzięło udział 11 płetwonurków w wieku od 18 do 41 lat. Badani spędzili 40 min w jeziorze, w wodzie o temperaturze 13° C, schodząc maksymalnie na głębokość około 9 m. Podczas zanurzenia wszyscy oddychali powietrzem. Krew do badań pobrano z żyły odłokciowej dwukrotnie: przed zanurzeniem w wodzie (kontrola) oraz bezpośrednio po wynurzeniu. W pracy wykazano tendencję do wzrostu aktywności katalazy oraz tendencję do obniżania się aktywności peroksydazy glutationowej, ale zmiany te nie były istotne statystycznie. Ciśnienie otoczenia wywierane przez wodę podczas nurkowania nie ma istotnego wpływu na aktywność peroksydazy glutationowej oraz katalazy w erytrocytach. Może to sugerować, że nie doszło do zwiększonej generacji nadtlenku wodoru będącego substratem reakcji katalizowanych przez oba enzymy. Brak istotnych statystycznie zmian aktywności badanych enzymów antyoksydacyjnych może być efektem zmian przystosowawczych w organizmie osób nurkujących.
