N-Glykosyl-Derivate. Teil XIII. Der nachträgliche ausbau des aglykons. Synthese von N-Glykosyl-Derivaten des 2-amino-thiazols, 2-amino-1,3,4-thiadiazols und 5-amino-1,2,3,4-thiatriazols

Zusammenfassung Einige neue N -Glykosyl-Derivate werden aus einfacheren Glykosylamin-Abkommlingen durch nachfolgenden Aufbau des Aglykons synthetisiert. Die Kondensation von (Tetra- O -acetyl- d -glucopyranosyl)-thioharnstoff ( 1 ) mit Chloraceton ergibt 4-Methyl-2-(tetra- O -acetyl- d -glucopyranosylamino)-thiazol ( 2 ), mit Phenacylbromid das 4-Phenyl-Analogon ( 4 ). Die Reaktion von 4-(Tetra- O -acetyl- d -glucopyranosyl)-thiosemicarbazid ( 5 ) mit Orthoameisensauretriathylester liefert das 2-(Tetra- O -acetyl- d -glucopyranosylamino)-1,3,4-thiadiazol ( 6 ). Das 5-(Tetra- O -acetyl- d -glucopyranosylamino)-1,2,3,4-thiatriazol ( 9 ) wird aus 5 mit salpetriger Saure bzw. aus (Tetra- O -acetyl- d -glucopyranosyl)-isothiocyanat ( 8 ) mit Azoimid synthetisiert. Die Addition von p -Chloranilin an 8 oder von Tetra- O -acetyl- d -glucopyranosylamin ( 11 ) an p -Chlorphenylisothiocyanat liefert den N-p -Chlorphenyl- N′ -(tetra- O -acetyl- d -glucopyranosyl)-thioharnstoff ( 12 ). Die Acetate der neuen Glykosylamin-Abkommlinge wurden desacetyliert. Das 1-[(Tetra- O -acetyl- d -glucopyranosyl)-thiocarbamoyl]-thiosemicarbazid( 14 ) wird aus 8 mit Thiosemicarbazid, das 1,5-Bis[(tetra- O -acetyl- d -glucopyranosyl)-thiocarbamoyl]-thiocarbonohydrazid ( 15 ) aus 8 mit Thiocarbohydrazid synthetisiert. Anschliessend wird die Herstellung einiger neuer d -Glucopyranosyl-thiosemicarbazone bzw. ihrer O -Acetyl-Derivate beschrieben; die physikalischen Eigenschaften und IR-Daten dieser Verbindungen werden mitgeteilt.

Abstract Intramolecular Diels-Alder reactions of E,Z mixtures of nonatrienes 6 and 9 led to the exclusive formation of the bicyclic compounds 10 and 11 respectively.

Abstract The title glycoside [phenyl 2- O -(3-amino-2,3,6-trideoxy-α- l - arabino-hexopyranosyl -β- d -glucopyranoside] was prepared from phenyl 3- O -benzyl-4,6- O -benzylidene-β- d -glucopyranoside and 3-azido-2,3,6-trideoxy-1,4-di- O - p -nitrobenzoyl- l - arabino -hexopyranose or 3-azido-2,3,6-trideoxy-4- O - p -nitrobenzoyl- l - arabino -hexopyranosyl chloride.

Application of the Ferriers carbocyclic ring closure to methyl 3-azido-2,3,6-trideoxyhex-5-enopyranosides leads to cyclitol derivatives (3)and (5) suitable for the convenient synthesis of 1,3-diaminocyclitol -type aminoglycoside antibiotic analogues.
