Propylthiouracil for Treatment of Hyperthyroidism in a Horse

A 19-year-old 250-kg Quarter Horse mare was examined in the fall for evaluation of weight loss of 4 years’ duration. The mare had been purchased 6 years before examination and was reported to be normal in appearance, weight, and demeanor. She had been used for trail riding. The owner initially noticed weight loss following transient postpartum diarrhea 4 years before presentation. The weight loss was mild and slowly progressed with intermittent periods of soft feces. Referral for evaluation was prompted by marked weight loss over the preceding 6 months. The mare had a ravenous appetite and polydipsia. In addition, the mare did not completely shed her hair coat during summer and was heat intolerant in ambient temperatures that exceeded 32.2 1C (901F). This was manifested as lethargy, sweating, and increased respiratory rate.

Measurement of ascorbic acid concentration and glutathione peroxidase activity in biological samples collected from horses with recurrent airway obstruction

OBJECTIVE To measure the ascorbic acid (AA) concentration in bronchoalveolar lavage fluid (BALF) and cellular glutathione peroxidase (cGPx) activity in RBCs and WBCs from peripherally obtained blood and in cells from BALF to determine whether differences existed between the 2 major redox systems in recurrent airway obstruction (RAO)-affected and -nonaffected (control) horses and between systemic and local pulmonary responses in the glutathione redox system. ANIMALS 16 adult horses in pairs: 8 healthy (control) and 8 RAO-affected horses. PROCEDURES Physical examination data and biological samples were collected from horses before (remission), during, and after (recovery) environmental challenge with dusty straw and hay. At each stage, BALF cell AA concentration and RBC, WBC, and BALF cell cGPx activity were measured. RESULTS Compared with control horses, RAO-affected horses had significantly higher cGPx activity in RBCs at all points and in WBCs during remission and challenge. The BALF cell cGPx activity was higher in RAO-affected horses during recovery than during remission The BALF cell AA concentration did not differ significantly in control horses at any point, but total and free AA concentrations were significantly lower in RAO-affected horses during the challenge period than during remission and recovery periods. CONCLUSIONS AND CLINICAL RELEVANCE High cGPx activity suggested this redox system was upregulated during exposure to dusty straw and hay to combat oxidative stress, as AA was depleted in RAO-affected horses. The relative delay and lack of comparative increase in cGPx activity within the local environment (represented by BALF cells), compared with that in RBCs and WBCs, might contribute to disease in RAO-affected horses.

Influence of Age on Surfactant Isolated from Healthy Horses Maintained on Pasture

BACKGROUND Surfactant alterations are described in horses after exercise, anesthesia, and prolonged transport, in horses with recurrent airway obstruction, and in neonatal foals. The effect of horse age or bronchoalveolar lavage fluid (BALF) sample characteristics on surfactant is unknown. OBJECTIVES To evaluate surfactant phospholipid composition and function in healthy horses, and to investigate the influence of age and BALF sample characteristics on surfactant. ANIMALS Seventeen healthy horses 6-25 years of age maintained on pasture year-round. METHODS BALF was collected by standard procedures and was assessed for recovery volume, nucleated cell count (NCC), and cytology. Cell-free BALF was separated into crude surfactant pellet (CSP) and surfactant supernatant (Supe) by ultracentrifugation. Phospholipid and protein content were determined from both fractions. CSP phospholipid composition was analyzed by high-performance liquid chromatography with an evaporative light scatter detector. Surface tension of CSP was evaluated with a pulsating bubble surfactometer. Regression analysis was used to evaluate associations between age, BALF sample characteristics, and surfactant variables. RESULTS Results and conclusions were derived from 15 horses. Increasing age was associated with decreased phospholipid content in CSP but not Supe. Age did not affect protein content of CSP or Supe, or surfactant phospholipid composition or function. Age-related surfactant changes were unaffected by BALF recovery percentage, NCC, and cytological profile. CONCLUSIONS AND CLINICAL IMPORTANCE Older horses have decreased surfactant phospholipid content, which might be because of age-related pulmonary changes. Surfactant composition is unaffected by BALF sample characteristics at a BALF recovery percentage of at least 50%.

The Effect of Geographic Location on Circannual Adrenocorticotropic Hormone Plasma Concentrations in Horses in Australia

Background Longitudinal evaluation of plasma endogenous ACTH concentration in clinically normal horses has not been investigated in the Southern Hemisphere. Objectives To longitudinally determine monthly upper reference limits for plasma ACTH in 2 disparate Australian geographic locations and to examine whether location affected the circannual rhythm of endogenous ACTH in the 2 groups of horses over a 12‐month period. Animals Clinically normal horses <20 years of age from 4 properties (institutional herd and client owned animals) in Perth (n = 40) and Townsville (n = 41) were included in the study. Methods A prospective longitudinal descriptive study to determine the upper reference limit and confidence intervals for plasma ACTH in each geographic location using the ASVCP reference interval (RI) guidelines, for individual months and monthly groupings for 12 consecutive months. Results Plasma endogenous ACTH concentrations demonstrated a circannual rhythm. The increase in endogenous ACTH was not confined to the autumnal months but was associated with changes in photoperiod. During the quiescent period, plasma ACTH concentrations were lower, ≤43 pg/mL (upper limit of the 90% confidence interval (CI)) in horses from Perth and ≤67 pg/mL (upper limit of the 90% CI) in horses from Townsville, than at the acrophase, ≤94 pg/mL (upper limit of the 90% CI) in horses from Perth, ≤101 pg/mL (upper limit of the 90% CI) in horses from Townsville. Conclusions and Clinical Importance Circannual rhythms of endogenous ACTH concentrations vary between geographic locations, this could be due to changes in photoperiod or other unknown factors, and upper reference limits should be determined for specific locations.

Equine leptospirosis in tropical Northern Queensland

OBJECTIVE Determine leptospiral serodiversity, serodominant serovars and prevalence in the horse population of Northern Queensland (NQ), Australia, with special focus on the Atherton Tableland and Townsville-Burdekin regions. DESIGN Cross-sectional survey. METHODS Serum samples from 429 horses originating from 172 horse-owning properties in NQ were collected at regional horse shows and veterinary clinics. Samples were analysed using a microscopic agglutination test. Owners were interviewed on potential risk factors associated with leptospirosis. RESULTS Of the 172 properties that submitted samples, horses from 60 properties (35%, 95% confidence interval (CI) 31-39%) in NQ showed evidence of exposure to one or more serovars of Leptospira at a serum dilution of 1 : 100. The proportion of seropositive properties in the Atherton Tableland and Townsville-Burdekin regions was 25% (95% CI 17-33%) and 36% (95% CI 30-43%), respectively. In total, 81 horses had titres to a single serovar and the remained had titres to two or more serovars. Although 20 serovars representing 17 serogroups were detected, serovar Arborea was serodominant and detected in horses from 29 properties. Geldings were more at risk (odds ratio 3.3) of being seropositive to serovar Arborea. The detection of serovar Arborea-seropositive horses was strongly associated (P < 0.05) with the average annual rainfall each property received and the presence of pigs and sugarcane fields in the vicinity. CONCLUSION Serological evidence of leptospirosis in horses in NQ indicates that prevalence is high and horses have been exposed to a wide range of serovars.

Surfactant alterations in horses with recurrent airway obstruction at various clinical stages.

OBJECTIVE To evaluate the phospholipid composition and function of surfactant in horses with recurrent airway obstruction (RAO) at various clinical stages and compare these properties with findings in horses without RAO. ANIMALS 7 horses with confirmed RAO and 7 without RAO (non-RAO horses). PROCEDURES Pairs of RAO-affected and non-RAO horses were evaluated before, during, and after exposure to hay. Evaluations included clinical scoring, lung function testing, airway endoscopy, and bronchoalveolar lavage fluid (BALF) absolute and differential cell counts. Cell-free BALF was separated into crude surfactant pellet and supernatant by ultracentrifugation, and phospholipid and protein concentrations were determined. Phospholipid composition of crude surfactant pellets and surface tension were evaluated with high-performance liquid chromatography and a pulsating bubble surfactometer, respectively. Findings were compared statistically via mixed-effects, repeated-measures ANOVA. RESULTS Total phospholipid concentration in BALF was lower in RAO-affected versus non-RAO horses at all sample collection times. In the RAO-affected group, total phospholipid concentration was lower during exposure to hay than before or after exposure. There were no significant differences in BALF protein concentration, percentages of phospholipid classes, or surface tension between or within groups of horses. CONCLUSIONS AND CLINICAL RELEVANCE All clinical stages of RAO-affected horses were characterized by low surfactant concentration in BALF. Exacerbation of RAO led to an additional decrease in surfactant concentration. Causes for low surfactant concentration in RAO-affected horses remain to be determined. Low phospholipid concentration may render RAO-affected horses more susceptible than unaffected horses to surfactant alterations and contribute to clinical disease status and progression.

Equine pituitary pars intermedia dysfunction: current understanding and recommendations from the Australian and New Zealand Equine Endocrine Group

The purpose of this article is to provide a review of the current knowledge and opinions about the epidemiology, clinical findings (including sequelae), diagnosis, treatment and monitoring of equine pituitary pars intermedia dysfunction, particularly in the Australian context. This information and the recommendations provided will assist practitioners in making informed decisions regarding the diagnosis and management of this disorder.

Seroprevalence and associated risk factors of mosquito-borne alphaviruses in horses in northern Queensland

OBJECTIVE To investigate the seroprevalence and associated risk factors of alphaviruses (Ross River virus (RRV), Barmah Forest virus (BFV) and Whataroa virus (WHAV)) in northern Queensland horses. METHODS A cross-sectional study of alphavirus antibodies in horses (n = 287) from 147 properties in northern Queensland from September 2013 to June 2014 was conducted. Owners of sampled horses were interviewed on potential risk factors. Data were analysed for associations using multivariable logistic regression. RESULTS Antibody titres for RRV were demonstrated in samples from 134 properties (91%; 95% confidence interval (CI) 87-96%); 22 properties (15%) had BFV reactors (95% CI 12-18%) and 2 properties (1.4%) had WHAV reactors (95% CI -0.5-3.2%). The highest seroprevalence of RRV was in the Townsville-Burdekin region (93%; 95% CI 90-96%) followed by the Mackay-Whitsunday (90%; 95% CI 88-98%) and Far North Coast-Tableland (82%; 95% CI 74-90%) regions. No association (P ≤ 0.05) could be shown between any of the viruses and age groups, sexes, annual average temperature, degree of rainfall or proximity to wet environments. An association with reported large numbers of mosquitoes was seen for RRV but not BFV. A significant association between properties in close proximity to poultry and pigs was shown for BFV. CONCLUSION RRV is endemic within the horse population of northern Queensland, but horses exhibit few clinical signs and could play a role as amplifying hosts in the tropics. Exposure of horses to BFV is significant in northern Queensland and it should be considered a differential diagnosis for RRV. WHAV warrants further study.

Circannual variability in adrenocorticotropic hormone responses to administration of thyrotropin-releasing hormone in clinically normal horses in Australia

Thyrotropin releasing hormone (TRH)-stimulation testing for pituitary pars intermedia dysfunction (PPID) in horses is only recommended at certain times of the year. Current diagnostic cut-off values reflect testing in the northern hemisphere during this time. The aims of this study were to evaluate TRH stimulation testing during two different phases of the circannual pituitary cycle and to determine whether diagnostic cut-off values developed in the northern hemisphere are appropriate in Australia. Thirteen clinically normal horses at Perth, Western Australia, and 23 horses at Townsville, Queensland, Australia, had TRH stimulation tests performed at two different time points during the circannual pituitary cycle. At both locations, post-TRH adrenocorticotropic hormone (ACTH) concentrations were significantly different between testing time points (Perth: P=0.001; Townsville: P<0.0001). In Perth, the mean ACTH concentrations 10min post-TRH in September and March were 51.4pg/mL (95% confidence interval, CI, 46.4-56.4pg/mL) and 248.5pg/mL (95% CI 170.2-326.9pg/mL), respectively. The median percentage change in ACTH concentrations in March was 361.9%. In Townsville, the mean ACTH concentrations 30min post-TRH in September and April were 35.3pg/mL (95% CI 29.6-40.9pg/mL) and 112.3pg/mL (95% CI 93.4-131.2pg/mL), respectively. The median percentage change in ACTH concentrations in April was 144.7%. The ACTH cut-off value after TRH stimulation in normal horses in September in Perth and Townsville was similar to the values established in the northern hemisphere. However, TRH stimulation testing in March/April was highly variable at both locations.
