Carcass quality of 10 beef cattle breeds of the Southwest of Europe in their typical production systems

A sample of about 70 young bulls of each of 10 beef cattle breeds from France and Spain, reared in their typical production systems, was studied regarding growth and carcass quality traits. There were large differences between and within breed-systems. The animals slaughtered in Spain weighed between 444 and 551 kg, whereas the range in France was from 610 to 750 kg. Growth showed a linear tendency; the daily weight gain ranging from 1.03 to 1.65 kg/day. Asturiana de los Valles and Pirenaica were, among the Spanish populations, the breed-systems with more muscle (around 75%) and less fat (8-10%) in the carcass, whereas Avilena-Negra Iberica, Morucha and Retinta showed opposite characteristics (60-65% muscle and 17-19% total fat at the 6th rib). Bruna dels Pirineus and Asturiana de la Montana occupied an intermediate position. All of the French breed-systems, Aubrac, Gasconne and Salers surpassed 73% muscle, while the percentage of total fat ranged from 7.6 to 10%. Salers breed-system also showed the longest and thinnest hind-limb. In the wide range of carcass weights studied in this paper, the general relationships among carcass traits were confirmed. Animals with the better conformation were also leaner. Longer carcasses tended to be associated with poor conformation and fatness. Bone content was clearly opposed to carcass conformation and muscle content.

Muscle transcriptomic profiles in pigs with divergent phenotypes for fatness traits

BackgroundSelection for increasing intramuscular fat content would definitively improve the palatability and juiciness of pig meat as well as the sensorial and organoleptic properties of cured products. However, evidences obtained in human and model organisms suggest that high levels of intramuscular fat might alter muscle lipid and carbohydrate metabolism. We have analysed this issue by determining the transcriptomic profiles of Duroc pigs with divergent phenotypes for 13 fatness traits. The strong aptitude of Duroc pigs to have high levels of intramuscular fat makes them a valuable model to analyse the mechanisms that regulate muscle lipid metabolism, an issue with evident implications in the elucidation of the genetic basis of human metabolic diseases such as obesity and insulin resistance.ResultsMuscle gene expression profiles of 68 Duroc pigs belonging to two groups (HIGH and LOW) with extreme phenotypes for lipid deposition and composition traits have been analysed. Microarray and quantitative PCR analysis showed that genes related to fatty acid uptake, lipogenesis and triacylglycerol synthesis were upregulated in the muscle tissue of HIGH pigs, which are fatter and have higher amounts of intramuscular fat than their LOW counterparts. Paradoxically, lipolytic genes also showed increased mRNA levels in the HIGH group suggesting the existence of a cycle where triacylglycerols are continuously synthesized and degraded. Several genes related to the insulin-signalling pathway, that is usually impaired in obese humans, were also upregulated. Finally, genes related to antigen-processing and presentation were downregulated in the HIGH group.ConclusionOur data suggest that selection for increasing intramuscular fat content in pigs would lead to a shift but not a disruption of the metabolic homeostasis of muscle cells. Future studies on the post-translational changes affecting protein activity or expression as well as information about protein location within the cell would be needed to to elucidate the effects of lipid deposition on muscle metabolism in pigs.

Characterisation of young bulls of the Bruna dels Pirineus cattle breed (selected from old Brown Swiss) in relation to carcass, meat quality and biochemical traits

Seventy-four young bulls of the Bruna dels Pirineus beef cattle breed were reared in a typical production system and slaughtered at an average age of 381 days and live weight of 541 kg. The animals were evaluated for productive traits, carcass quality, meat quality, eating quality and biochemical characteristics of m. longissimus thoracis (LT). Biochemical measurements included intramuscular fat (IMF) and collagen proportion, haem pigment concentration, lactate dehydrogenase and isocitrate dehydrogenase activities and type I (MHC I) fibre percentage determined by ELISA. Bruna dels Pirineus bulls achieved high growth rates during the fattening period (1.63 kg/day) and showed good carcass quality, with a high dressing-out proportion (607 g/kg, hot carcass), a good conformation score (U; EUROP) and a moderate fatness score (3;1-5). Carcass composition was estimated from the sixth rib joint dissection (682 g/kg lean proportion, 127 g/kg total dissectable fat and 163 g/kg bone). IMF (24.3 g/kg) and MHC I (27.9%) showed high variabilities (CV>30%). Sensory analysis of LT included beef and livery odour and flavour intensity, and overall tenderness and juiciness assessment of loin samples (14-day ageing). Beef odour and flavour were slightly positively correlated with IMF and carcass fatness score (P<0.05). Fatness, MHC I, insoluble collagen and cooking losses tended to affect the livery flavour intensity positively. This variable was significantly higher in meat from bulls of lower carcass quality (i.e. lower conformation score, lower lean proportion; P<0.05) and higher type I fibre percentage. Loin overall tenderness and juiciness were not affected by the biochemical traits studied, however, they were negatively affected by cooking loss (P<0.05).

Mapping of quantitative trait loci for cholesterol, LDL, HDL, and triglyceride serum concentrations in pigs

The fine mapping of polymorphisms influencing cholesterol (CT), triglyceride (TG), and lipoprotein serum levels in human and mouse has provided a wealth of knowledge about the complex genetic architecture of these traits. The extension of these genetic analyses to pigs would be of utmost importance since they constitute a valuable biological and clinical model for the study of coronary artery disease and myocardial infarction. In the present work, we performed a whole genome scan for serum lipid traits in a half-sib Duroc pig population of 350 individuals. Phenotypic registers included total CT, TG, and low (LDL)- and high (HDL)-density lipoprotein serum concentrations at 45 and 190 days of age. This approach allowed us to identify two genomewide significant quantitative trait loci (QTL) for HDL-to-LDL ratio at 45 days (SSC6, 84 cM) and for TG at 190 days (SSC4, 23 cM) as well as a number of chromosomewide significant QTL. The comparison of QTL locations at 45 and 190 days revealed a notable lack of concordance at these two time points, suggesting that the effects of these QTL are age specific. Moreover, we have observed a considerable level of correspondence among the locations of the most significant porcine lipid QTL and those identified in humans. This finding might suggest that, in mammals, diverse polymorphisms located in a common set of genes are involved in the genetic variation of serum lipid levels.

Porcine intramuscular fat content and composition are regulated by quantitative trait loci with muscle-specific effects.

Intramuscular fat (IMF) storage is a biological process with a strong impact on nutritional and technological properties of meat and also with relevant consequences on human health. The genetic architecture of IMF content and composition phenotypes has been thoroughly studied in pigs through the identification of QTL and the estimation of genetic parameters. A question that has not been elucidated yet is if the genetic determinants of IMF-related phenotypes are muscle specific or, conversely, have broad effects on the whole skeletal muscle compartment. We have addressed this question by generating lipid QTL maps for 2 muscles with a high commercial value, gluteus medius (GM) and longissimus thoracis et lumborum (LTL), in a Duroc commercial population (n = 350). Our data support a lack of concordance between the GM and LTL QTL maps, suggesting that the effects of polymorphisms influencing IMF, cholesterol, and fatty acid contents are modulated to some extent by complex spatial factors related to muscle location, metabolism, and function. These results have important implications on the implementation of genomic selection schemes aimed to improve the lipid profile of swine meat.

Polymorphism of the pig acetyl-coenzyme A carboxylase α gene is associated with fatty acid composition in a Duroc commercial line

Acetyl-coenzyme A carboxylase alpha (ACACA) catalyses the first committed step in the biosynthesis of long-chain fatty acids (FA) by converting acetyl-CoA into malonyl-CoA. In pigs, the ACACA gene maps to a chromosome 12 QTL with important effects on FA composition. In the present study, we have sequenced the coding region of the pig ACACA gene in 15 pigs, identifying 21 polymorphic sites that were either synonymous or non-coding. Ten of these SNPs segregated in a Duroc commercial population (n = 350) for which lipid metabolism and meat and carcass quality trait records were available. Significant associations were found between two linked single nucleotide polymorphisms (c.4899G>A and c.5196T>C) and percentages of carcass lean, intramuscular fat, monounsaturated, saturated (myristic, palmitic and stearic) and polyunsaturated (linoleic) FAs in the longissimus thoracis et lumborum muscle, along with serum HDL-cholesterol concentration. The most important allele substitution effects were observed for the polyunsaturated/saturated FA ratio (13-21% of the phenotypic mean) as well as for the percentages of omega-6 and polyunsaturated FAs, especially linoleic acid (7-16% of the phenotypic mean). These results suggest the existence of a causal mutation, mapping to the chromosomal region containing the pig ACACA gene, with marked effects on FA composition of meat.

Bayes factor analyses of heritability for serum and muscle lipid traits in Duroc pigs.

Concern about pork quality has increased during last decades. Given the influence of fat content and composition on sensorial, nutritional, and technological variables of pork meat, an accurate knowledge about genetic control of pig lipid metabolism is required. This study focused on providing a broad characterization for serum and meat lipid trait heritability estimates in pigs. Analyses were performed on a population of 370 Duroc barrows and measured the additive polygenic background for the serum concentrations of cholesterol, triglyceride, and low- and high-density lipoproteins at 45 and 190 d of age (at slaughter), as well as intramuscular fat, cholesterol content, and C:12 to C:22 fatty acid content in longissimus thoracis et lumborum and gluteus medius muscles at slaughter. These traits were analyzed under Bayesian univariate animal linear models, and the statistical relevance of heritability estimates was evaluated through Bayes factor (BF); the model with polygenic additive effects was favored when BF >1. All serum lipid traits showed relevant genetic determinism, but the BF reached greater values at 190 d of age. Serum lipid traits displayed moderate modal estimates for heritability that ranged from 0.18 to 0.30. On the other hand, the genetic determinism for meat quality traits showed a heterogeneous behavior with large and less-than-1 BF. In general, longissimus thoracis et lumborum and gluteus medius muscles showed a similar pattern, with strong evidence of polygenic additive effects for intramuscular fat and palmitic, stearic, and cis-vaccenic fatty acids content, whereas oleic and muscle cholesterol content showed moderate to weak BF with moderate heritabilities. Similarly, results regarding linoleic, arachidonic, n-3, and n-6 fatty acids suggested a moderate genetic determinism, but only in gluteus medius muscle. For the remaining traits (myristic and palmitoleic fatty acids in both muscles, along with linoleic, arachidonic, n-3, and n-6 fatty acids in the longissimus thoracis et lumborum muscle), no statistical evidence for genetic control was observed in this study. As a whole, these results confirm the complexity of lipid metabolism in pigs.

Functional and association studies on the pig HMGCR gene, a cholesterol-synthesis limiting enzyme

The 3-hydroxy-3-methylglutaryl-CoA reductase (HMGCR) is the rate-limiting enzyme in the biosynthesis of cholesterol. We have studied the role of the HMGCR gene in pig lipid metabolism by means of expression and structural analysis. We describe here the complete coding region of this gene in pigs and report two synonymous single nucleotide polymorphisms in the coding region. We have, additionally, studied the association of one of these polymorphisms (HMGCR:c.807A>C) with several lipid deposition- and cholesterol-related traits in a half-sib population generated from a commercial Duroc line, showing in some families a positive relationship of HMGCR:c.807A allele with serum low-density lipoprotein (LDL)-bound cholesterol and triglyceride levels, and also with intramuscular fat (IMF) content of gluteus medius muscle. We have also assessed the expression levels in muscle and in liver from 68 Duroc individuals corresponding to the most extreme animals for the analysed traits. Liver HMGCR expression correlated negatively with the serum high-density lipoprotein (HDL) levels, carcass lean percentage and stearic acid content, while muscle expression correlated also negatively with the carcass lean percentage, stearic and linoleic acids content, but showed a positive correlation with the serum lipid cholesterol (HDL, LDL and total cholesterol), IMF and muscle oleic and palmitic fatty acid content. With this information, we have performed an association analysis of expression data with lipid metabolism phenotypic levels and the HMGCR genotype. The results indicate that HMGCR expression levels in muscle are different in the two groups of pigs with extreme values for fat deposition and total cholesterol levels, and also between animals with the different HMGCR genotypes.

Bayesian threshold analysis of direct and maternal genetic parameters for piglet mortality at farrowing in Large White, Landrace, and Pietrain populations.

A Bayesian threshold model was fitted to analyze the genetic parameters for farrowing mortality at the piglet level in Large White, Landrace, and Pietrain populations. Field data were collected between 1999 and 2006. They were provided by 3 pig selection nucleus farms of a commercial breeding company registered in the Spanish Pig Data Bank (BDporc). Analyses were performed on 3 data sets of Large White (60,535 piglets born from 4,551 litters), Landrace (57,987 piglets from 5,008 litters), and Pietrain (42,707 piglets from 4,328 litters) populations. In the analysis, farrowing mortality was considered as a binary trait at the piglet level and scored as 1 (alive piglet) or 0 (dead piglet) at farrowing or within the first 12 h of life. Each breed was analyzed separately, and operational models included systematic effects (year-season, sex, litter size, and order of parity), direct and maternal additive genetic effects, and common litter effects. Analyses were performed by Bayesian methods using Gibbs sampling. The posterior means of direct heritability were 0.02, 0.06, and 0.10, and the posterior means of maternal heritability were 0.05, 0.13, and 0.06 for Large White, Landrace, and Pietrain populations, respectively. The posterior means of genetic correlation between the direct and maternal genetic effects for Landrace and Pietrain populations were -0.56 and -0.53, and the highest posterior intervals at 95% did not include zero. In contrast, the posterior mean of the genetic correlation between direct and maternal effects was 0.15 in the Large White population, with the null correlation included in the highest posterior interval at 95%. These results suggest that the genetic model of evaluation for the Landrace and Pietrain populations should include direct and maternal genetic effects, whereas farrowing mortality could be considered as a sow trait in the Large White population.

Quantitative trait loci analysis of a Duroc commercial population highlights differences in the genetic determination of meat quality traits at two different muscles

We performed a whole-genome scan with 110 informative microsatellites in a commercial Duroc population for which growth, fatness, carcass and meat quality phenotypes were available. Importantly, meat quality traits were recorded in two different muscles, that is, gluteus medius (GM) and longissimus thoracis et lumborum (LTL), to find out whether these traits are determined by muscle-specific genetic factors. At the whole-population level, three genome-wide QTL were identified for carcass weight (SSC7, 60 cM), meat redness (SSC13, 84 cM) and yellowness (SSC15, 108 cM). Within-family analyses allowed us to detect genome-wide significant QTL for muscle loin depth between the 3rd and 4th ribs (SSC15, 54 cM), backfat thickness (BFT) in vivo (SSC10, 58 cM), ham weight (SSC9, 69 cM), carcass weight (SSC7, 60 cM; SSC9, 68 cM), BFT on the last rib (SSC11, 48 cM) and GM redness (SSC8, 85 cM; SSC13, 84 cM). Interestingly, there was low positional concordance between meat quality QTL maps obtained for GM and LTL. As a matter of fact, the three genome-wide significant QTL for colour traits (SSC8, SSC13 and SSC15) that we detected in our study were all GM specific. This result suggests that QTL effects might be modulated to a certain extent by genetic and environmental factors linked to muscle function and anatomical location.
