Química e farmacologia de quimioterápicos antineoplásicos derivados de plantas

This review demonstrates the importance of plants as sources of molecules used in anticancer therapies. The approach is performed by relating the active molecules to their origins, details, mechanisms of action, structure-activity relationship and chemical characteristics of chemotherapeutical medicines. It was also described the development of anticancer agents from plants by the pharmaceutical industry and the difficulties to release these compounds as a trademark. These include the well known paclitaxel, docetaxel, vincristine, vinblastine, vinorelbine, vindesine, etoposide, teniposide, and other molecules that are undergoing clinical trials.

Antinociceptive Properties of Bergenin

Bergenin (1) is a C-glucoside of 4-O-methylgallic acid with known antiarthritic activity attributed to modulation of cytokine production. The present study was undertaken to evaluate whether 1 has antinociceptive properties in models of inflammatory pain and to investigate its possible mechanisms of action. Pretreatment with 1 (12.5-100 mg/kg, ip) produced a dose-related inhibition of acetic acid-induced writhing in mice. Furthermore, treatment with 1 (50 and 100 mg/kg) inhibited both the early and late phases in a formalin test. In addition, 1 (50 and 100 mg/kg, ip) inhibited mechanical hyperalgesia, edema, and paw production of hyperalgesic cytokines (TNF-α and IL-1β) induced by complete Freunds adjuvant. However, the local production of IL-10, an anti-inflammatory cytokine, was not altered by 1 (100 mg/kg, ip). Treatment with 1 produced a similar profile of antinociception in wild-type and IL-10-deficient mice. Mice treated with 1 did not show any motor performance alterations or apparent systemic toxicity. The results presented herein demonstrate that bergenin has consistent antinociceptive and anti-inflammatory properties, acting by the inhibition of IL-1β and TNF-α production, and suggest its potential for the control of inflammatory pain.

Mechanisms involved in the antinociceptive effects of 7-hydroxycoumarin.

7-Hydroxycoumarin (umbelliferone, 1), the main metabolite of coumarin, has been reported to produce potent antinociceptive effects in animal models of pain. However, the biochemical events involved in antinociception mediated by 1 are currently not well understood. In the present study, the mechanisms by which 1 exerts its pharmacological actions were investigated. Acute pretreatment of mice with 1 produced a long-lasting antinociceptive effect against complete Freunds adjuvant (CFA)-induced hyperalgesia. The subchronic administration of 1 inhibited CFA-induced hyperalgesia and paw edema, while it did not cause any apparent toxicity. Another set of experiments showed that 1 inhibited carrageenan-induced mechanical hyperalgesia, but not mechanical hyperalgesia induced by prostaglandin E(2) (PGE(2)), suggesting that it acts upstream of PGE(2.) Treatment with 1 was able to prevent the plantar tissue neutrophil influx induced by local inflammatory stimuli. In addition, 1 exhibited inhibitory effects on the release of hyperalgesic cytokines (TNF-α and IL-1β) and the production of PGE(2), a directly acting hyperalgesic mediator. The present results suggest that the antinociceptive effect of 1 is correlated with the inhibition of neutrophil migration, cytokine release, and PGE(2) production and are supportive of the further investigation of the therapeutic potential of 1 to control inflammatory pain.

Estrutura, metabolismo e funções fisiológicas da lipoproteína de alta densidade

Several experimental, clinical and epidemiological researches have shown the incontestable causal relationship between low high-density lipoprotein (HDL) plasma concentrations and cardiovascular pathology on an atherosclerotic basis. Low HDL levels characterize about 10% of the general population and they represent the most frequent dyslipidemia in patients with coronary artery disease. Reduced HDL concentrations would be unable to effectively eliminate the cholesterol excess at the vascular wall, contributing to the inflammatory phenomenon that characterizes the pathogenesis of atherosclerosis since its initial phases. Results of numerous studies reasonably allow supposing that HDL is able to exert, also directly, anti-inflammatory actions through the modulation of expression of diverse acute phase proteins. Furthermore, HDL also exerts several other atheroprotective effects, such as antioxidants affects, inhibition of platelets aggregation and monocytes migration. This paper is a review on recent literature data about HDL metabolism and its role in the prevention of coronary artery disease.

Razão triglicérides/HDL-C e proteína C reativa de alta sensibilidade na avaliação do risco cardiovascular

INTRODUCTION: Calculated indexes from laboratory measurements and the determination of plasma markers are commonly used in cardiovascular risk assessment. OBJECTIVE: To use triglycerides/high-density lipoprotein cholesterol (HDL-C) ratio, estimate of low-density lipoprotein (LDL) particle size and high sensitive C-reactive protein (hs-CRP) levels to the clinical and laboratorial assessment of coronary risk among patients with coronary disease. MATERIAL AND METHODS: Sixty patients were chosen accordingly, 36 female (F) (65 ± 9 years of age) and 24 male (M) (55 ± 10 years of age) diagnosed with stable ischemic cardiopathy at Ana Neri Hospital, Salvador, Bahia, Brazil. The investigation was carried out at the Clinical Biochemistry Laboratory of the College of Pharmacy at Federal University of Bahia (UFBA). Blood samples were collected after a 12-hour fast to determine hs-CRP, lipid profile and risk indicators. All data were assessed through Syncron LX®20 Beckman Coulter. The inferential analysis results were considered significant when p < 5%. Data analysis was carried out with GraphPad Prism® (4.03 version) software. RESULTS: Data analysis showed a positive correlation between hs-CRP and triglycerides/HDL-C ratio only among women (r = 0.4; p = 0.0442), n = 36. The analysis of other parameters did not show any significance. CONCLUSION: The triglycerides/HDL-C ratio and hs-CRP may be used as indicators of increase in cardiovascular risk among women with coronary artery disease (CAD).

Levels of high-density lipoprotein cholesterol (HDL-C) among children with steady-state sickle cell disease

BackgroundThe search for sickle cell disease (SCD) prognosis biomarkers is a challenge. These markers identification can help to establish further therapy, later severe clinical complications and with patients follow-up. We attempted to study a possible involvement of levels of high-density lipoprotein cholesterol (HDL-C) in steady-state children with SCD, once that this lipid marker has been correlated with anti-inflammatory, anti-oxidative, anti-aggregation, anti-coagulant and pro-fibrinolytic activities, important aspects to be considered in sickle cell disease pathogenesis.MethodsWe prospectively analyzed biochemical, inflammatory and hematological biomarkers of 152 steady-state infants with SCD and 132 healthy subjects using immunochemistry, immunoassay and electronic cell counter respectively. Clinical data were collected from patient medical records.ResultsOf the 152 infants investigated had a significant positive association of high-density lipoprotein cholesterol with hemoglobin (P < 0.001), hematocrit (P < 0.001) and total cholesterol (P < 0.001) and a negative significant association with reticulocytes (P = 0.046), leukocytes (P = 0.015), monocytes (P = 0.004) and platelets (P = 0.005), bilirubins [total bilirubin (P < 0.001), direct bilirubin (P < 0.001) and indirect bilirubin (P < 0.001], iron (P < 0.001), aminotransferases [aspartate aminotransferase (P = 0.004), alanine aminotransferase (P = 0.035)], lactate dehydrogenase (P < 0.001), urea (P = 0.030), alpha 1-antitrypsin (P < 0.001), very low-density lipoprotein cholesterol (P = 0.003), triglycerides (P = 0.005) and hemoglobin S (P = 0.002). Low high-density lipoprotein cholesterol concentration was associated with the history of cardiac abnormalities (P = 0.025), pneumonia (P = 0.033) and blood transfusion use (P = 0.025). Lipids and inflammatory markers were associated with the presence of cholelithiasis.ConclusionsWe hypothesize that some SCD patients can have a specific dyslipidemic subphenotype characterized by low HDL-C with hypertriglyceridemia and high VLDL-C in association with other biomarkers, including those related to inflammation. This represents an important step toward a more reliable clinical prognosis. Additional studies are warranted to test this hypothesis and the probably mechanisms involved in this complex network of markers and their role in SCD pathogenesis.

Could the essential oil of Lippia alba provide a readily available and cost-effective anaesthetic for Nile tilapia (Oreochromis niloticus)?

Abstract We report the optimal concentration of the essential oil of Lippia alba (EOLA) for the induction and recovery of anesthesia in Nile tilapia (Oreochromis niloticus) during handling. Groups of 10 juveniles were separately subjected to a concentration of EOLA: 0 (control group), 10, 20, 50, 100, 200, 300, or 500 µL L-1. The plasma cortisol levels were significantly decreased in anesthetized fish 4 h after stress of handling. The plasma glucose levels were significantly increased 1 and 4 h after stress of handling in fish exposed to EOLA. Sensory analysis showed that tasters did not detect differences through a blind test in fillet taste and odor between exposed and non-exposed fish. The ideal concentration to promote the best anesthesia and recovery times and to reduce plasma cortisol 4 h after handling is 500 µL L-1.

Serum levels of cytokines and chemokines associated with cardiovascular disease in Brazilian patients treated with statins for dyslipidemia

The anti-inflammatory effect of 3-hydroxy-3-methyl-glutaryl-CoA reductase inhibitors (statins) has been investigated in dyslipidemic patients treated with these pharmacologic agents. The aim of this study was to investigate the serum levels of cytokines and chemokines that have been associated with atherosclerosis and cardiovascular disease in Brazilian patients treated for hypercholesterolemia with statin. The serum levels of the cytokines IL-1β, IL-6, IL-10, TNF-α and TGF-β, and the levels of the chemokines IL-8 (CXCL8) and MCP-1 were determined by enzyme-linked immunosorbent assay and tested for their association with cardiovascular disease. The suppression of circulating levels of TNF-α, MCP-1 and IL-8 and their enhancing effect on IL-10 and TGF-β production were more pronounced in male patients. Female patients treated with statins who had a previous myocardial infarction presented higher median levels of both TNF-α and IL-8 (P<0.05) and a lower median level of IL-10 than female patients without MI (P<0.05). Except in women with a previous myocardial infarction, the treatment of dyslipidemic Brazilian patients with statins down-modulates the production of atherogenic cytokines and chemokines and increases the circulating levels of anti-atherogenic cytokines.

Maternal and post-weaning exposure to a high fat diet promotes visceral obesity and hepatic steatosis in adult rats

AIM considering the frequent consumption of fat-rich diets by women of reproductive age, the aim of the present study was to investigate the effects of maternal consumption of a high-fat diet during the perinatal and/ or post-weaning period on the liver parameters and lipid metabolism of young rats. METHODS Wistar female rats were fed a high-fat (H) or control (C) diet during pregnancy and lactation. The offspring were allocated to four groups: Control Control (CC, n = 11), offspring fed a control diet after weaning; Control High-fat (CH, n = 10), offspring fed a high-fat diet after weaning; High-fat High-fat (HH, n = 10), offspring of mothers H fed a high-fat diet after weaning; and High-fat Control (HC, n = 9), offspring of mothers H fed with control diet after weaning. RESULTS AND DISCUSSION the food intake did not differ among the groups, however, the relative weight of the adipose tissue was higher in animals from the HC, HH and CH groups (p ≤ 0.005). Liver steatosis was found in the CH and HH animals, which also exhibited hypercholesterolemia (p ≤ 0.05). The levels of the liver enzymes alanine aminotransferase (ALT) and gamma-glutamyl transpeptidase (GGT) were higher in the HH group, and the LDL level was higher in the CH group compared to the CC. The consumption of an obesogenic diet during critical periods of development may contribute to the occurrence of visceral obesity, liver steatosis and hypercholesterolemia in adult rats, even in the absence of changes in dietary intake.

Environmental effects on the gills and blood of Oreochromis niloticus exposed to rivers of Bahia, Brazil

Through the integration of chemical, biochemical and morphological analyses, this study investigated the effects of multiple pollutants on environmental biomarkers, such as gill histopathological changes and hematological and biochemical parameters, in Oreochromis niloticus exposed to four sites in the Jacuipe and Subaé rivers over seven days. Sediment analyses identified Sapelba as the most contaminated site, followed by Oliveira de Campinhos, Santo Amaro and Jacuípe. Water analyses revealed aluminum, iron and manganese at all sites. Aluminum and other metal were also detected in the gills of fishes. Fish exposed to the Sapelba site exhibited significant necrosis formation, as well as higher hematological parameters and trend to increase of cortisol levels. However, filament epithelium proliferation was higher at the Oliveira de Campinhos and Santo Amaro sites, at which the lowest levels of the hematological variables were observed. Multivariate analysis grouped some gill histopathological changes together, such as epithelial detachment with edema and lamellar epithelial proliferation with the lamellar fusion of adjacent filaments, revealing relationships among them. Positive associations were identified between sediment contamination and necrosis and cortisol, while water contamination was related with filament epithelium proliferation, aneurism, lamellar fusion and several hematological parameters. Furthermore, relationships between blood parameters and gill histopathological changes demonstrated a joint physiological response that may have resulted from environmental variables such as dissolved oxygen. The results exhibited the direct influence of xenobiotics on these biomarkers but also highlighted the need to consider the complexity of environmental factors to optimize the adoption of these environmental predictive tools.
