Protein and collagen synthesis in rat diaphragm muscle incubated in vitro: The effect of alterations in tension produced by electrical or mechanical means

Abstract 1. 1. The synthesis of non-collagen protein (NCP) and collagen (CP) have been measured in vivo and in isolated diaphragm muscle with labelled proline. 2. 2. Under non-stimulated conditions the rates of synthesis of NCP (0.9%/day) and CP (0.2%/day) were 10% of those in vivo . Intermittent changes in tension induced by stimulation of the phrenic nerve, direct stimulation or rhythmic stretching stimulated NCP synthesis (+ 170%: +97%; +153% respectively) but had little effect on CP synthesis. 3. 4. Stimulation did not alter amino acid transport but increased glucose uptake and lactate output. 4. 5. The results are discussed in the context of an in vitro model for stretch-induced hypertrophy.
