Novel Pseudomonas strains INA04, INA05, and INA06, were isolated from activated sludge. Strain INA06 was found to degrade long chain alkylphenols such as 4-n-amylphenol and 4-n-hexylphenol as the sole source of carbon, apart from co-metabolic degradation of 4-n-nonylphenol in the presence of phenol, while INA04 and INA05 could grow on phenol, but could not grow well on alkylphenols. Induction studies on strain INA06 revealed a broad substrate-specific phenol hydroxylase, for the metabolism of phenol and alkylphenols, inducible with phenol or para-substituted alkylphenol. They bore close resemblance to members of Pseudomonas sensu stricto. 16S rDNA sequence homology of INA06 was closest to P. veronii (99.7%). DNA-DNA hybridization pointed out higher linkage (64% similarity) to the type strain of P. veronii than to other species of Pseudomonas sensu stricto (>60%). The BOX-PCR profile of all INA strains was similar, but different from that of P. veronii. Since biochemical characteristics were similar to those of P. veronii, and genetic relatedness was at the margin of species differentiation level (70%), we propose these strains to be treated as a new subspecies of P. veronii. The type strain of this new subspecies, named P. veronii subsp. inensis subsp. nov., is strain INA06. The accession number of strain INA05 is CIP 107595=JCM11829, and that of INA06 is CIP107594(T)=JCM11828(T). The 16S rDNA sequence accession number (DDBJ/EMBL/GenBank) of strain INA06 is AB056120.

A novel filamentous bacterium, strain NAF001, was isolated from suspended water of a domestic wastewater treatment tank. It formed an extremely long filamentous trichome and produced endospores. It formed spore-like resting cells (SLRCs) which were heat-resistant. SLRCs grew by budding to form short filaments resembling the gonidia of filamentous bacteria such as LEUCOTHRIX: This is the first report of a Bacillus species that exhibits budding growth. The filamentous form was neither restricted to any particular growth stage nor dependent on cultural conditions. Phylogenetic analysis of the 16S rRNA gene revealed that this isolate was a member of the genus Bacillus, with no close relatives at the species level (sequence similarity <95.3%). Strain NAF001 thus probably belongs to a new and novel species of Bacillus.

A novel filamentous Bacillus strain, NAF001T, was reported previously that produces endospores and spore-like resting cells; the latter outgrow by budding. Phylogenetic analysis based on 16S rDNA gene sequences reported in the same paper speculated on the proposal of a novel species for this isolate. This communication describes the DNA-DNA relatedness of strain NAF001T to various members of the genus Bacillus and its whole-cell fatty acid and quinone profiles, in order to authenticate the creation of a novel species, for which the name Bacillus funiculus sp. nov. is proposed. The type strain is NAF001T (= JCM 11201T = CIP 107128T). Further, features of the binding points between filaments of strain NAF001T that enable it to form extremely long filaments are captured by electron microscopy.

Bacillus species were observed and quantified by molecular approaches, using the 165 rDNA primers/probes, in a wastewater treatment plant designed for the purpose of stimulating the growth ofBacillus species. The plant has been operating as a test plant since 1997 in the city of Ina, Japan, with excellent treatment performance. Observations byin situ hybridization, usingBacillus-specific probes, indicated thatBacillus strains were inhabited in the plant and their numbers decreased during the treatment process. Similar results were obtained from a quantitative PCR analysis using aBacillus-specific primer set, and the amount of DNA originating from variousBacillus species was maximally 1.91% of the total DNA in the wastewater treatment tank. Clone library analysis using theBacillus-specific primers suggested that, while the population was noticeably increased, the phylogenetic diversity of the increasingBacillus species was very low.

Treatment of a five-membered cyclic oxosulfonium ylide 3 with β-acetoxy-α-methylene ketones in the presence of two equimolar amounts of base afforded the cycloheptene oxide derivatives 7a−i as single stereoisomers in 19−77% yields. The products were considered to form through a Michael-type addition of the ylide, followed by elimination of the acetoxy group and an intramolecular Corey−Chaykovsky reaction. On the other hand, the same treatment, when using a six-membered oxosulfonium ylide, gave the corresponding cyclooctene oxide derivatives in moderate yields, with the products formed in a ca. 4:1 mixture of stereoisomers, a fact correlated with differences in the configuration of the sulfur atom in the sulfinyl group.

Reaction of methyl 4,6‐di‐O‐acetyl‐2,3‐dideoxy‐d‐erythro‐hex‐2‐enopyranoside 1 with two equivalents of diphenylphosphenium cation at 0°C gave the 1‐phosphonylated 2‐enopyranosides 3α and 3β as major products. Similarly, reaction of diphenylphosphenium cation with tri‐O‐acetylglycal afforded the same products in a similar ratio. In contrast, a reaction with tri‐O‐benzylglucal at reflux temperature of dichloromethane afforded the 3‐phosphonylated sugar 13 as a major product. These reactions may proceed via stable allyl cations.

Abstract Aminophosphonic chlorides 1–3 reacted with silver thiocyanate to give corresponding isothiocyanates 4–6 in good to moderate yields. Similarly, aminophosphonic chlorides 7–9 reacted with unsaturated alcohols and allylmagnesium bromide to give trienes 15–18 containing phosphorus. An intramolecular Diels-Alder reaction of triene 18 gave isophosphindoline derivative 19. The conformation of 19 is also discussed.
