Pre-parturition profile of steroids and prostaglandin in cows with or without foetal membrane retention

Retained foetal membranes in cattle is one of the most common complications associated to the reduction in milk yield and impaired fertility in dairy cattle. In order to determine some endocrine mechanisms controlling parturition and delivery of foetal membranes, plasma concentrations of steroids and prostanoids were determined in 20 healthy Holstein cows. Samples were taken within the interval of 5 days pre-parturition to 12h after calving. Progesterone (P4) levels were similar in cows with (PR) and without (NPR) placental retention. While the estradiol-17beta (E2) peak at parturition was lower in PR than in NPR cows, cortisol levels were greater in PR cows 12 and 24h pre-parturition. The Prostaglandin F2alpha metabolite (PGFM) levels were higher at parturition in NPR compared with the PR group, but 12h later, these levels in the PR group increased so that concentrations were greater as compared with NPR cows. The Prostaglandin E2 metabolite (PGEM), 24, 48 and 72 h pre-parturition, were higher in PR cows. However, the PGFM:PGEM ratio was greater in cows up NPR at all time when included, indicating the importance of higher levels of Prostaglandin F2alpha (PGF2alpha) than Prostaglandin E2 (PGE2) for normal placental delivery. In conclusion, placental retention was related to both estrogen and PGF2alpha deficiency, which may be a consequence of metabolic stress leading to PGE2 and maternal cortisol synthesis before parturition.

A field evaluation of anthelmintics for control of cyathostomes of horses in Brazil.

The anthelmintic efficacy of ivermectin (IVM), piperazine citrate (PPZ) and fenbendazole (FBZ) alone, and combinations of FBZ with varying dosages of PPZ, was evaluated for the control of strongyles of Mangalarga equids. In all cases of positive strongly egg counts per gram of faeces (EPG) before and after treatment, pure populations of cyathostome larvae with eight gut cells were consistently observed in faecal cultures. The faecal egg count reductions (FECR) were evaluated between Day 7 and Day 56 post-treatment. Significant differences (P less than or equal to 0.05) were found between Day 7 and Day 49 when comparing the mean EPG values of IVM with those of the other anthelmintics. PPZ alone and the combination of FBZ (7.5 mg kg-1) and PPZ (55 mg base kg-1) did not show significant differences during the trial; both groups exhibiting an FECR of greater than 90% between Day 7 and Day 30. Horses treated with the combinations of FBZ (7.5 mg kg-1) and PPZ at either 40 or 25 mg base kg-1, and with FBZ alone at either 7.5 or 15 mg kg-1, showed an FECR of less than 90% 2 weeks after treatment. The emergence of a degree of resistance of strongyles to FBZ and PPZ in a segment of the cyathostomes in this population is suggested.

Resposta esteroidogênica induzida por hCG em garanhões jovens Mangalarga: testosterona e sulfato de estrona plasmáticos

Leydig cell acute steroidogenic response, in seven young Mangalarga stallions (2.6 - 4.5 ys), was evaluated through human Corionic Gonadotrophin (hCG) stimulation. The animals were treated with 5,000 UI (hCG) Vetecor®, i.v., in bollus, at 8 a.m. Plasma samples were collected by venopuncture at 7 a.m. (basal), 4, 24, 48, and 72 hours after stimulation (from 28 to August 31). Testosterone (T) and estrone sulphate (E1SO4) was measured by radioimmunoassay (RIA), intra-assay variation £ 5%. Data were analysed by Friedman and Wilcoxon Matched-Pairs tests (p £ 0.05). The magnitude of hormonal production after stimulation was analysed by Percentual Variation (D %). At 48 hours, 57.1% of the animals (n = 4) had maximal response (T) with values 5.7 greater than basal. And although 57.1% of them (n = 4) had maximal response (E1SO4) at 4 hours, the maximal value was 1.6 greater than basal at 24 hours. The hCG may be employed to stimulate the Leydig cell steroidogenic response and 5,000 UI is enough to stimulate testicular steroidogenesis. The estrogen basal concentrations are higher than testosterone basal concentrations. This protocol could be applied to identify fertility problems of endocrine origin in horses of this breed.
