Zinc mapping in bone tissues by histochemistry and synchrotron radiation-induced X-ray emission: correlation with the distribution of alkaline phosphatase.

Zinc distribution in osteons was mapped by synchrotron radiation-induced X-ray emission analysis in both human and porcine adult bone, as well as in porcine bone by histochemistry using Timms method. Both procedures showed that zinc is not uniformly distributed, being in its highest concentration on haversian bone surfaces. When Timms method was applied in conjunction with a procedure leading to partial zinc extraction, three zinc pools were specifically detected: a loose one, found in the mineralizable osteoid; a mineral one, bound to the bone mineral; and a tenacious one, firmly bound to an organic component located in the osteoid and mineralizing organic matrix. The alkaline phosphatase distribution was also mapped in porcine adult bone by histochemistry and immunohistochemistry and it was found codistributed with tenacious zinc mainly at the calcification front. The data suggest that alkaline phosphatase is buried as a bone matrix protein during initial mineralization.

Prevalence of abdominal obesity in Spanish children and adolescents. Do we need waist circumference measurements in pediatric practice

Background Evidence indicates that central adiposity has increased to a higher degree than general adiposity in children and adolescents in recent decades. However, waist circumference is not a routine measurement in clinical practice. Objective This study aimed to determine the prevalence of abdominal obesity based on waist circumferences (WC) and waist to height ratio (WHtR) in Spanish children and adolescents aged 6 to 17 years. Further, the prevalence of abdominal obesity (AO) among normal and overweight individuals was analyzed. Design Data were obtained from a study conducted from 1998 to 2000 in a representative national sample of 1521 children and adolescents aged 6 to 17 years (50.0% female) in Spain. WC and WHtR measurements were obtained in addition to BMI. AO was defined as WHtR ≥0.50 (WHtR-AO), sex and age specific WC≥90th percentile (WC-AO1), and sex and age specific WC cut-off values associated with high trunk fat measured by by dual-energy X-ray absorptiometry (WC-AO2). Results IOTF- based overweight and obsity prevalence was 21.5% and 6.6% in children and 17.4% and 5.2% in adolescents, respectively. Abdominal obesity (AO) was defined as WHtR≥0.50 (WHtR-AO), sex- and age-specific WC≥90th percentile (WC-AO1), and sex- and age-specific WC cut-off values associated with high trunk fat measured by dual-energy X-ray absorptiometry (WC-AO2). The respective prevalence of WHtR-AO, WC-AO1, and WC-AO2 was 21.3% (24.6% boys; 17.9% girls), 9.4% (9.1% boys; 9.7% girls), and 26.8% (30.6% boys;22.9% girls) in children and 14.3% (20.0% boys; 8.7% girls), 9.6% (9.8% boys; 9.5% girls), and 21.1% (28.8% boys; 13.7% girls) in adolescents. Conclusion The prevalence of AO in Spanish children and adolescents is of concern. The high proportion of AO observed in young patients who are normal weight or overweight indicates a need to include waist circumference measurements in routine clinical practice.

Mediterranean diet impact on changes in abdominal fat and 10-year incidence of abdominal obesity in a Spanish population.

Abdominal obesity is a strong predictor of metabolic disorders. Prospective data on the association between the Mediterranean diet and surrogate markers of abdominal adiposity are scarce. The present study evaluated the relationship between adherence to the Mediterranean diet and (1) changes in waist circumference (WC) and (2) 10-year incidence of abdominal obesity. We conducted a prospective, population-based study in 3058 male and female Spaniards aged 25-74 years, followed from 2000 to 2009. Dietary intake and leisure-time physical activity levels were recorded using validated questionnaires. Weight, height and WC were measured. Adherence to the Mediterranean diet, determined using the previously validated REGICOR-Mediterranean diet score (R-MDS), based on the distribution of population food intake and on the dietary recommendations (MDS-rec), was negatively associated with WC gain (P = 0.007 and 0.024, respectively) in fully adjusted models. In the multivariate logistic analysis, the odds of abdominal obesity incidence decreased across the tertiles of the R-MDS, but the association was not significant. In conclusion, adherence to the Mediterranean diet was associated with lower abdominal fat gain, but not with 10-year incidence of abdominal obesity.

Labeling studies on cortical bone formation in the antlers of red deer (Cervus elaphus)

The formation and mineralization process of antlers, which constitute the fastest growing bones in vertebrates, is still not fully understood. We used oxytetracycline injections to label different stages of bone formation in antlers of 14 red deer between days 28 and 156 of antler growth. Results show that initially a trabecular scaffold of woven bone is formed which largely replaces a pre-existing scaffold of mineralized cartilage. Lamellar bone is then deposited and from about day 70 onwards, primary osteons fill in the longitudinal tubes lined by the scaffold in a proximal to distal sequence. Mineral apposition rate (MAR) in early stages of primary osteon formation is very high (average 2.15 μm/d). Lower MARs were recorded for later stages of primary osteon formation (1.56 μm/d) and for the smaller secondary osteons (0.89 μm/d). Results suggest a peak in mineral demand around day 100 when the extent of mineralizing surfaces is maximal. A few secondary osteons were formed in a process of antler modeling rather than remodeling, as it occurred simultaneously with formation of primary osteons. The degree of cortical porosity reflects a reduction in MAR during later stages of osteonal growth, whereas cortical thickness is determined earlier. Injections given when the antlers were largely or completely clean from velvet produced no labels in antler bone, strongly suggesting that antlers are dead after velvet shedding. The rapidity of antler mineralization and the short lifespan of antlers make them an extraordinary model to assess the effects of chemicals impairing or promoting bone mineralization.

Does nutrition affect bone porosity and mineral tissue distribution in deer antlers? The relationship between histology, mechanical properties and mineral composition

It is well known that porosity has an inverse relationship with the mechanical properties of bones. We examined cortical and trabecular porosity of antlers, and mineral composition, thickness and mechanical properties in the cortical wall. Samples belonged to two deer populations: a captive population of an experimental farm having a high quality diet, and a free-ranging population feeding on plants of lower nutritive quality. As shown for minerals and mechanical properties in previous studies by our group, cortical and trabecular porosity increased from the base distally. Cortical porosity was always caused by the presence of incomplete primary osteons. Porosity increased along the length of the antler much more in deer with lower quality diet. Despite cortical porosity being inversely related to mechanical properties and positively with K, Zn and other minerals indicating physiological effort, it was these minerals and not porosity that statistically better explained variability in mechanical properties. Histochemistry showed that the reason for this is that Zn is located around incomplete osteons and also in complete osteons that were still mineralizing, whereas K is located in non-osteonal bone, which constitutes a greater proportion of bone where osteons are incompletely mineralized. This suggests that, K, Zn and other minerals indicate reduction in mechanical performance even with little porosity. If a similar process occurred in internal bones, K, Zn and other minerals in the bone may be an early indicator of decrease in mechanical properties and future osteoporosis. In conclusion, porosity is related to diet and physiological effort in deer.

Energy density, diet quality, and central body fat in a nationwide survey of young Spaniards

OBJECTIVES To determine the impact of energy density on diet quality and anthropometric proxy measures for central body fat in young Spaniards. METHODS Data were obtained from a representative national sample of 2513 young Spaniards aged 10 to 24 y. Dietary assessment was performed with a 24-h recall. Adherence to the Mediterranean diet was measured by the KIDMED index. Reduced rank regression analysis was used to identify energy-density dietary patterns. Waist circumferences and height were measured. RESULTS Pastries and sausages showed the strongest loading for the dietary energy density factor score (DED-FS), with fruits and vegetables at the opposite end of the scale. Multiple linear regression analysis showed a positive association (P < 0.01) of dietary energy density (DED) and the DED-FS with waist circumferences regressed on height and age (WCheight+age) and waist-to-height ratio (WHtR). The adjusted odds ratio for elevated WCheight+age and WHtR increased across quartile distribution of DED (P < 0.001) and DED-FS (P < 0.05). Diet quality characterized by the KIDMED index was negatively associated (P < 0.001) with DED and the DED-FS. CONCLUSIONS Higher DED is a risk for increased central fat distribution and is associated with low adherence to the Mediterranean diet.

Enamel-Calibrated Lamellar Bone Reveals Long Period Growth Rate Variability in Humans

Mammalian teeth exhibit incremental structures representing successive forming fronts of enamel at varying time scales, including a short daily increment called a cross striation and a long period called a stria of Retzius, the latter of which, in humans, occurs on average every 8–9 days. The number of daily increments between striae is called the repeat interval, which is the same period as that required to form one increment of bone, i.e. the lamella, the fundamental – if not archetypal – unit of bone. Lamellae of known formation time nevertheless vary in width, and thus their measures provide time-calibrated growth rate variability. We measured growth rate variability for as many as 6 years of continuously forming primary incremental lamellar bone from midshaft femur histological sections of sub-Saharan Africans of Bantu origin and known life history. We observed periodic growth rate variability in approximately 6- to 8-week intervals, and in some cases annual rhythms were visible. Endogenous biological periodicities, cycles manifest in the external environment, and/or perturbations of development are all potentially contained within growth rate variability studies of lamellar incremental patterns. Because lamellae are formed within defined periods of time, quantitative measures of widths of individual lamellae provide time-resolved growth rate variability that may reveal rhythms in human bone growth heretofore unknown.

Production and in vitro characterization of 3D porous scaffolds made of magnesium carbonate apatite (MCA)/anionic collagen using a biomimetic approach.

3D porous scaffolds are relevant biomaterials to bone engineering as they can be used as templates to tissue reconstruction. The aim of the present study was to produce and characterize in vitro 3D magnesium-carbonate apatite/collagen (MCA/col) scaffolds. They were prepared by using biomimetic approach, followed by cross-linking with 0.25% glutaraldehyde solution (GA) and liofilization. Results obtained with Fourier-transform infrared spectroscopy (FT-IR) confirmed the type-B carbonate substitution, while by X-ray diffraction (XRD), a crystallite size of ~10nm was obtained. Optical and electron microscopy showed that the cylindrical samples exhibited an open-porous morphology, with apatite nanocrystals precipitated on collagen fibrils. The cross-linked 3D scaffolds showed integrity when immersed in culture medium up to 14 days. Also, the immersion of such samples into an acid buffer solution, to mimic the osteoclastic resorption environment, promotes the release of important ions for bone repair, such as calcium, phosphorus and magnesium. Bone cells (SaOs2) adhered, and proliferated on the 3D composite scaffolds, showing that synthesis and the cross-linking processes did not induce cytotoxicity.

The structure of pedicle and hard antler bone in the European roe deer (Capreolus capreolus): a light microscope and backscattered electron imaging study

Deer antlers are deciduous bony structures that develop from permanent frontal outgrowths, the pedicles. While growth and bone architecture of antlers have been studied in greater detail, information on pedicle formation and structure is scarce. The present study provides information on the structure of pedicle and hard antler bone in the European roe deer. A pronounced seasonal variation in pedicle architecture was observed, with high porosity around antler casting and a very compact bone structure during the hard antler stage. These observations suggest a corresponding marked variation also in the biomechanical properties of the pedicles. The seasonally alternating extensive resorption and formation processes make the pedicles of older deer heavily remodeled structures. Pedicles increase in thickness by apposition of primary bone that subsequently becomes replaced by secondary osteons. The antler cortex of roe deer is largely composed of a tubular framework of woven bone trabeculae with some remnants of mineralized cartilage, and primary osteons that have filled in the intertrabecular spaces. Secondary osteons are scarce, denoting little remodeling in antlers, which can be related to their short lifespan. The occurrence of cement lines around primary osteons indicates resorption on the trabecular scaffold prior to infilling of the intertrabecular spaces. The outer cortex showed a higher autofluorescence and a more immature structure than the main cortex, suggesting that it was secondarily formed by periosteal activity. Pedicles and antlers constitute a functional entity, and future histological and/or biomechanical studies should therefore consider both components of the cranial appendages.

Study protocol: effects of the THAO-child health intervention program on the prevention of childhood obesity - the POIBC study

BackgroundThe speeding increase and the high prevalence of childhood obesity is a serious problem for Public Health. Community Based Interventions has been developed to combat against the childhood obesity epidemic. However little is known on the efficacy of these programs. Therefore, there is an urgent need to determine the effect of community based intervention on changes in lifestyle and surrogate measures of adiposity.Methods/designParallel intervention study including two thousand 2249 children aged 8 to 10 years ( 4th and 5th grade of elementary school) from 4 Spanish towns. The THAO-Child Health Program, a community based intervention, were implemented in 2 towns. Body weight, height, and waist circumferences were measured. Children recorded their dietary intake on a computer-based 24h recall. All children also completed validated computer based questionnaires to estimate physical activity, diet quality, eating behaviors, and quality of life and sleep. Additionally, parental diet quality and physical activity were assessed by validated questionnaires.DiscussionThis study will provide insight in the efficacy of the THAO-Child Health Program to promote a healthy lifestyle. Additionally it will evaluate if lifestyle changes are accompanied by favorable weight management.Trial registrationTrial Registration Number ISRCTN68403446
