Isolation and Identification of Yeasts from Wild Flowers of Daejeon City and Chungcheongnam-do in Korea

Total 50 species, 80 strains: Eighty strains of yeasts were isolated from wild flowers of Daejeon city and its surroundings in Korea. Isolated yeasts were identified by comparison of their PCR-amplified 18s rDNA V3 region or internal transcribed spacer-2 (ITS-2) genes with BLAST database. Among them, 32 species, 54 strains of yeasts were isolated and identified from wild flowers of Daejeon city, and Cryptococcus species were dominant yeasts. Furthermore, 18 species, 26 strains of yeasts were isolated and identified from wild flowers of Geumsan-gun and Nonsan-gun, Chungcheongnam-do in Korea, and also Candida species and Pseudozyma species were dominant yeasts.

Isolation and Diversity of Yeasts from Wild Flowers in Ulleungdo and Yokjido, Korea

Various yeasts from wild flowers of Ulleungdo in Gyeongsangbuk-do and Yokjido in Gyeongsangnam-do, Korea were isolated and identified by comparison of nucleotide sequences for PCR-amplified D1/D2 region of 26S rDNA using BLAST. Forty eight yeast strains of twenty two species and sixty yeast strains of twenty five species were isolated from wild flowers of Ulleungdo and Yokjido, respectively. Only seven species were overlapped from the two different islands areas: Cryptococcus albidus, Cryptococcus laurentii, Metschnikowia reukafii, Pichia scolyti, Rhodotorula glutinis, Rhodotorula graminis and Rhodotorula mucilaginosa. Among forty species from two different islands, other thirty three species were restricted to specific collection site suggesting that each area has distinctive yeast flora.

Isolation and Physiological Functionality of Yeasts from Wild Flowers in Seonyudo of Gogunsanyeoldo, Jeollabuk-do, Korea

전라북도 고군산열도 선유도 일대에서 2014년 4월에 개화한 야생화들로부터 효모들을 분리한 후 이들의 26S rDNA의 D1/D2 부위의 염기서열을 결정한 다음 NCBI의 BLAST database에 등록되어 있는 효모들과의 상동성을 비교 분석하여 동정하였다. 야생화 51점에서 21종의 효모들을 61 균주 분리하였고, 이들 중 Cryptococcus aureus SY1-4 등의 Cryptococcus 속 균들이 가장 많았고, Metschnikowia reukaufii SY20-1등 Metschnikowia 속 균과 Rhodotorula ingeniosa SY1-1등 Rhodotorula 속 균들도 비교적 많이 분리되었다. 이들 61 균주들의 배양상등액과 무세포추출물을 제조한 후 생리기능성을 측정한 결과 M. reukaufii SY44-6의 배양상등액이 각각 49.6%의 XOD 저해활성과 38.4%의 미백성 Tyrosinase 저해활성을 보여 우수하였다. 【Sixty one yeast strains of 21 species were isolated from wild flowers in Gogunsanyeoldo, including Seonyudo of Jeollabuk-do, Korea, and identified by comparison of nucleotide sequences for PCR-amplified D1/D2 region of 26S rDNA using BLAST. Among them, Cryptococcus sp. including C. aureus SY1-4 were found to be dominant, and Metschnikowia sp. including M. reukaufii SY20-1 and Rhodotnula sp. such as R. ingeniosa SY1-1 were also abundantly isolated. Some physiological functionalities of the culture broth and cell-free extracts from 61 yeast strains were determined. Supernatant from Metschnilowia reukaufii SY44-6 showed anti-gout xanthine oxidase inhibitory activity of 49.6% and whitening tyrosinase inhibitory activity of 38.4%, respectively.】

Isolation and Diversity of Yeasts from Fruits and Flowers of Orchard in Sinam-myeon of Yesan-gun, Chungcheongnam-do, Korea

Several kinds of yeasts from fruits and flowers of orchard in Yesan-gun of Chungcheongnam-do, Korea were isolated and identified by comparison of nucleotide sequences for PCR-amplified D1/D2 region of 26S rDNA using BLAST. Fourty eight yeast strains of twenty five species and one hundred eight yeast strains of fourty eight species were isolated from fruits and flowers of orchard in Sinam-myeon of Yesan-gun, Chungcheongnam-do, respectively. Among total one hundred fifty-six yeast strains, only sixteen species were overlapped from fruits and flowers.

Note on Newly Isolated Yeasts from Wild Flowers in Daejeon City, Korea

We report morphological and physiological characteristics of newly isolated six kinds of yeasts which are new in Korean mycoflora. The yeasts were isolated from wild flowers in Daejeon city, Korea and identified by molecular analysis of the amplified internal transcribed space rDNA sequence or partial 18S rDNA sequence. Species of isolates were identified as Kuraishia capsulate, Lodderomyces elongisporus, Pseudozyma antarctica, Starmerella bombicola.

Yeasts Associated with Fruits and Blossoms Collected from Hanbat Arboretum, Daejeon, Korea

Yeasts are common inhabitants of the phyllosphere, but our knowledge of their diversity in various fruits and blossoms are limited. We collected different kinds of blossoms and unripened fruits from Hanbat arboretum, Daejeon, Korea at the year of 2013. Yeasts were isolated by plating of suspensions prepared for collected samples onto YPD medium containing antibiotics. BLAST searches were subsequently performed for the comparison of the partially determined sequences of D1/D2 domain of 26S rDNA. As a result, we isolated 57 yeast strains of 31 species from 29 different kinds of blossoms and 6 kinds of fruits samples. We found huge differences in yeast flora depending on the sample collection season.

New Records of Yeasts from Wild Flowers in Coast Near Areas and Inland Areas, Korea

국내 해안 근접지역과 내륙 일부 지역에 서식하고 있는 야생화들로부터 분리, 동정한 효모들 중에서 국내 미기록 효모들을 선별하여 이들의 미생물학적 특성들을 조사하였다. 동해시의 야생화들에서 Candida silvae와 Cryptococcus terrestris, 안동시 부근의 야생화에서는 Auriculibuller fuscus의 국내에서 보고되지 않은 효모들을 분리하였다. 충남 내륙지역인 금산군에서는 Candida quercitrusa, Hanseniaspora opuntiae등의 미기록 효모들을 분리하였다. 또한 서해안 지역의 대천시에서는 Hannaella zeae, 태안시의 야생화에서는 Ca-ndida saopaulonensis, Sporobolomyces carnicolor, sporobolomyces phaffii등이 미기록 효모들로 분리, 선별하였고 예산군에서는 Candida oleophila와 Jaminaea angkoriensis 미기록 효모들을 분리하였다. 우리나라 서남부지역인 광주시 야생화에서는 Hanseniaspora vineae, 최남단인 완도군에서는 Pseudozyma hubeiensis, Pichia mexicana 등 모두 8곳에서 14종의 미기록 효모들을 분리하였다. 이들 미기록 효모들의 형태적, 배양학적 성질들이 조사한 결과 유포자 효모가 3종, 무포자 효모가 11종이었고 위균사 형성 효모가 7종이었으며 각기 다른 배양학적 특성을 보였다. 【Fourteen new records of yeasts were screened from one hundred thirty-three yeasts strains which were isolated from wild flowers in east, west and south coast near areas and inland areas, Korea. Candida silvae, Cryptococcus terrestris and Auriculibuller fuscus were newly isolated from wild flowers collected at Donghae and Andong, respectively. Candida quercitrusa, Hanseniaspora opuntiae, Sporobolomyces carnicolor, Candida saopaulonensis and Sporobolomyces phaffii were newly isolated from wild flowers in Geumsan-gun and Taean-gun in Chungcheongnam-do, respectively. Candida oleophila and Jaminaea angkoriensis were newly identified from flowers in Yesan-gun and Hannaella zeae was also newly identified from flowers of Daecheon. Hanseniaspora vineae and two other yeast strains including Pseudozyma hubeiensis were newly isolated and identified from flowers collected at Gwangju and Wando-gun, respectively. Morphological and physiological characteristics of their new recorded yeasts were investigated.】

Microbiological Characteristics and Physiological Functionality of New Records of Yeasts from Wild Flowers in Yokjido, Korea

Abstract Two new yeast records, Cryptococcus adeliensis YJ19-2 and Cryptococcus uzbekistanensis YJ10-4 were screened from 60 yeasts strains that were isolated and identified from wild flowers in Yokjido, Gyeongsangnam-do, Korea. The morphological and cultural characteristics of the newly recorded yeasts and the physiological functionalities of the supernatants and cell-free extracts obtained from their cultures were investigated. The two newly recorded yeasts did not form ascospores and pseudomycelia. They also grew well in yeast extract-peptone-dextrose broth. C. uzbekistanensis YJ10-4 grew in a vitamin-free medium and was also tolerant to sugar and salt. Antihypertensive angiotensin I-converting enzyme inhibitory activity of the supernatant from C. adeliensis YJ19-2 was high (71.8%) and its cell-free extract also showed very high (81.2%) antidiabetic á-glucosidase inhibitory activity.

Isolation and Identification of Yeasts from Wild Flowers in Deogyu Mountain and their Physiological Functionalities

덕유산 무주 리조트 일대에서 수집한 야생화 27점에서 효모 8종 22균주를 분리 동정하였다. 이들 균주 중에서 Cryptococcus magnus 9균주, Cryptococcus flavescens 6균주와 Cryptococcus sp. 2균주 등 Cryptococcus속 균들이 가장 많이 분리되었다. 이들 22균주들을 YPD 배지에서 24시간 배양하여 각각의 배양 상등액과 무세포 추출물을 제조한 후 이들의 항산화 활성과 항통풍성 xanthine oxidase 저해활성 등의 생리기능성들을 측정한 결과, Sporobolomyces phaffii JS00583과 Rhodotorula graminis JS00581의 배양 상등액이 각각 40.4%와 39.1%의 항통풍성 xanthine oxidase 저해활성을 보였다. 또한 Cryptococcus magnus JS00570의 상등액 역시 36.1%의 미백성 tyrosinase 저해활성을 보였다. 【Several kinds of yeasts were isolated from wild flowers collected from Muju resort of Deogyu mountain in Korea and identified by comparison of nucleotide sequences of D1/D2 region of 26S rDNA using BLAST. Totally twenty-two strains of eight species were isolated and Cryptococcus magnus (9 strains) and Cryptococcus flavescenes (6 strains) were dominant species. The supernatant and cell-free extracts were prepared and their antioxidant activity, anti-gout xanthine oxidase inhibitory activity and whitening tyrosinase inhibitory activity were investigated. Isolation and Identification of Yeasts from Wild Flowers Collected around Jangseong Lake in Jeollanam-do, Republic of Korea, and Characterization of the Unrecorded Yeast Bullera coprosmaensis.

Abstract Several types of yeasts were isolated from wild flowers around Jangseong Lake in Jeollanam-do, Republic of Korea and identified by comparing the nucleotide sequences of the PCR amplicons for the D1/D2 variable domain of the 26S ribosomal DNA using Basic Local Alignment Search Tool (BLAST) analysis. In total, 60 strains from 18 species were isolated, and Pseudozyma spp. (27 strains), which included Pseudozyma rugulosa (7 strains) and Pseudozyma aphidis (6 strains), was dominant species. Among the 60 strains, Bullera coprosmaensis JS00600 represented a newly recorded yeast strain in Korea, and its microbiological characteristics were investigated. The yeast cell has an oval-shaped morphology measuring 1.4 × 1.7 μm in size. Bullera coprosmaensis JS00600 is an asporous yeast that exhibits no pseudomycelium formation. It grew well in vitamin-free medium as well as in yeast extract-malt extract broth and yeast extract-peptone-dextrose (YPD) broth, and it is halotolerant growing in 10% NaCl-containing YPD broth.
