ABSTRACT To assess the presence of the four main viruses responsible for human acute gastroenteritis in a hydrographic network impacted by a disordered urbanization process, a 1-year study was performed involving water sample collection from streams in the hydrographic basin surrounding the city of Manaus, Amazonas, Brazil. Thirteen surface water sample collection sites, including different areas of human settlement characterized as urban, rural, and primary forest, located in the Tarumã-Açu, São Raimundo, Educandos, and Puraquequara microbasins, were defined with a global positioning system. At least one virus was detected in 59.6% (31/52) of the water samples analyzed, and rotavirus was the most frequent (44.2%), followed by human adenovirus (30.8%), human astrovirus (15.4%), and norovirus (5.8%). The viral contamination observed mainly in the urban streams reflected the presence of a local high-density population and indicated the gastroenteritis burden from pathogenic viruses in the water, principally due to recreational activities such as bathing. The presence of viral genomes in areas where fecal contamination was not demonstrated by bacterial indicators suggests prolonged virus persistence in aquatic environments and emphasizes the enteric virus group as the most reliable for environmental monitoring.

In the last few years, awareness in developed countries has increased regarding the importance of urban watercourses as essential natural resources for human well being. Macroinvertebrates have been used as bioindicators to complement physico-chemical evaluation of water quality after environmental perturbations. The city of Manaus is closely associated with the Amazonian rain forest and with its dense hydrographic network. Any perturbation, such as deforestation and/or water pollution in the city’s streams, therefore causes changes in the local ecosystem as the population increases. In this study, 65 streams were sampled in October and November 2003. Samples were taken from stream-bed sediment in the center of the channel and litter/sediment at the edge of the stream. Deforestation, total Nitrogen (TN), total Phosphorus (TP), depth, width, electrical conductivity, temperature and dissolved Oxygen (DO) were measured. A total of 115,549 specimens were collected, distributed among 152 taxa. Oligochaeta, Chironomus, Psychodidae and Ceratopogonidae were the taxa with the greatest frequencies of occurrence and the highest total abundances. Higher deforestation, TN and TP were correlated with lower DO and greater electrical conductivity, pH and water temperature. Deforestation, TN and TP were not associated with water velocity and stream width. Depth was the only variable correlated (negatively) with deforestation and not correlated with TN and TP. Greater deforestation, TN and TP were correlated with lower richness of taxa; but these variables did not affect abundance. Canonical Correspondence Analysis ordenated the streams into two groups; the majority of the streams were in the group with high levels of deforestation and with high values of TP, TN, pH, electrical conductivity and temperature, where the macroinvertebrates were reduced to a few taxa. The other group was composed of streams that were well oxygenated and deep, where richness of taxa was higher. These results indicate changes in community composition in response to changes in environmental conditions. The highest taxa correlation was with streams that were well oxygenated and had the greatest depth and water velocity. Species Indicator Analysis identified 29 taxa as indicators of nonimpacted streams, 16 as indicators of deforested streams and three as indicators of streams impacted by deforestation and domestic sewage. Of the total sampled streams, 80% were impacted by deforestation and water pollution and had fauna tolerant of these perturbations. Water pollution, represented by TN and TP, affected the macroinvertebrate fauna in a way similar to deforestation, i.e., causing reduction in taxa richness, simplifying the insect community composition without changing abundance. Use of the taxa suggested in this study as environmental indicators could improve the evaluation of water quality in the streams in Central Amazonia.

Aedes aegypti and Ae. albopictus are vectors of dengue viruses, which cause endemic disease in the city of Manaus, capital of the state of Amazonas, Brazil. More than 53 thousand cases have been registered in this city since the first epidemic in 1998. We evaluated the hypothesis that different ecological conditions result in different patterns of vector infestation in Manaus, by measuring the infestation level in four neighborhoods with different urbanization patterns, during the rainy (April), dry (August), and transitional (November) seasons. Ae. aegypti predominated throughout the study areas and sampling periods, representing 86% of all specimens collected in oviposition traps. High frequencies of houses positive for both species were observed in all studied sites, with Ae. aegypti present in more than 84% of the houses in all seasons. Ae. albopictus, on the other hand, showed more spatial and temporal variation in abundance. We found no association between infestation level and house traits. This study highlights the homogeneity of dengue vector distribution in Manaus.

An anopheline survey was carried out in two simian malaria areas in the Brazilian Amazon, Balbina and Samuel, to determine the potential vectors of Plasmodium brasilianum. The most abundant and/or acrodendrophilic anophelines in the forest and the most likely vector were Anopheles mediopunctatus, An. nuneztovari, An. oswaldoi, An. triannulatus and An. shannoni. An. darlingi and An. marajoara were captured essentially in anthropic habitats outside the forest and are unlikely to be involved in the transmission of P. brasilianum among monkeys within the forests and from monkeys to man in their surroundings in the Amazon.

We present filaria-nested polymerase chain reaction (PCR), which is based on amplification of first internal transcribed spacer rDNA to distinguish three parasitic filarial species (Onchocerca volvulus, Mansonella ozzardi and Mansonella perstans) that can be found in the Amazon Region. Nested PCR-based identifications yielded the same results as those utilizing morphological characters. Nested PCR is highly sensitive and specific and it detects low-level infections in both humans and vectors. No cross-amplifications were observed with various other blood parasites and no false-positive results were obtained with the nested PCR. The method works efficiently with whole-blood, blood-spot and skin biopsy samples. Our method may thus be suitable for assessing the efficacy of filaria control programmes in Amazonia by recording parasite infections in both the human host and the vector. By specifically differentiating the major sympatric species of filaria, this technique could also enhance epidemiological research in the region.

Age-related seroprevalence studies that have been conducted in Brazil have indicated a transition from a high to a medium endemicity of hepatitis A virus (HAV) infection in the population. However, most of these studies have focused on urban populations that experience lower incidence rates of HAV infection. In the current study, the prevalence of anti-HAV antibodies was investigated in children with a low socioeconomic status (SES) that live on the periphery of three capital cities in Brazil. A total of 1,162 dried blood spot samples were collected from individuals whose ages ranged from one-18 years and tested for anti-HAV antibodies. A large number of children under five years old (74.1-90%) were identified to be susceptible to HAV infection. The anti-HAV antibody prevalence reached ≥ 50% among those that were 10-14 years of age or older. The anti-HAV prevalence rates observed were characteristics of regions with intermediate level of hepatitis A endemicity. These data indicated that a large proportion of children with a low SES that live at the periphery of urban cities might be at risk of contracting an HAV infection. The hepatitis A vaccine that is currently offered in Brazil is only available for high-risk groups or at private clinics and is unaffordable for individuals with a lower SES. The results from this study suggest that the hepatitis A vaccine should be included in the Brazilian National Program for Immunisation.

In a study of sandfly species in the Samuel Ecological Station, in Porto Velho, Rondonia State, the following species were identified: Lutzomyia brasiliensis, L. evangelistai, L. gomezi, L. anduzei, L. flaviscutellata, L. richardwardi, L. shawi, L. umbratilis, L. yuilli yuilli, L. dendrophyla, L. puctigeniculata, L. shannoni, L. amazonensis, L. ayrozai, L. carrerai carrerai, L. claustrei, L. davisi and L. lainsoni. L. richardwardi, L. umbratilis and L. c. carrerai were the predominant species captured of man forming 60.30% of the total catch. L. richarwardi was the most frequent at ground level (29.9%), while L. umbratilis predominated in the canopy (48.5%).

Sand flies (Diptera: Psychodidae) are responsible for the transmission of protozoan parasites that cause leishmaniases. They are found predominantly in forests, but some species exploit environments that have been subject to deforestation and subsequent human colonization. Studies conducted in Brazil over the past 30 years show that some species are adapting to peri-urban and urban settings. We evaluated sand fly diversity and abundance in the rural settlement of Rio Pardo, Presidente Figueiredo Municipality, Amazonas State, Brazil. Settlement households were divided into four categories. These categories were determined by the human population density and the degree of deforestation in the immediate area. We used CDC light traps to sample the area surrounding 24 households (6 households in each category). Samples were taken on six occasions during September-November 2009 and June-August 2010. A total of 3074 sand fly specimens were collected, including 1163 females and 1911 males. These were classified into 13 genera and 52 species. The greatest abundance of sand flies and the greatest richness of species were observed in areas where human population density was highest. Our results show that changes in the human occupancy and vegetation management in rural settlements may affect the population dynamics and distribution of sand fly species, thereby affecting the local transmission of cutaneous leishmaniases.

Background Arboviral diseases are major global public health threats. Yet, our understanding of infection risk factors is, with a few exceptions, considerably limited. A crucial shortcoming is the widespread use of analytical methods generally not suited for observational data – particularly null hypothesis-testing (NHT) and step-wise regression (SWR). Using Mayaro virus (MAYV) as a case study, here we compare information theory-based multimodel inference (MMI) with conventional analyses for arboviral infection risk factor assessment. Methodology/Principal Findings A cross-sectional survey of anti-MAYV antibodies revealed 44% prevalence (n = 270 subjects) in a central Amazon rural settlement. NHT suggested that residents of village-like household clusters and those using closed toilet/latrines were at higher risk, while living in non-village-like areas, using bednets, and owning fowl, pigs or dogs were protective. The “minimum adequate” SWR model retained only residence area and bednet use. Using MMI, we identified relevant covariates, quantified their relative importance, and estimated effect-sizes (β±SE) on which to base inference. Residence area (β Village = 2.93±0.41; β Upland = −0.56±0.33, β Riverbanks = −2.37±0.55) and bednet use (β = −0.95±0.28) were the most important factors, followed by crop-plot ownership (β = 0.39±0.22) and regular use of a closed toilet/latrine (β = 0.19±0.13); domestic animals had insignificant protective effects and were relatively unimportant. The SWR model ranked fifth among the 128 models in the final MMI set. Conclusions/Significance Our analyses illustrate how MMI can enhance inference on infection risk factors when compared with NHT or SWR. MMI indicates that forest crop-plot workers are likely exposed to typical MAYV cycles maintained by diurnal, forest dwelling vectors; however, MAYV might also be circulating in nocturnal, domestic-peridomestic cycles in village-like areas. This suggests either a vector shift (synanthropic mosquitoes vectoring MAYV) or a habitat/habits shift (classical MAYV vectors adapting to densely populated landscapes and nocturnal biting); any such ecological/adaptive novelty could increase the likelihood of MAYV emergence in Amazonia.
