Abstract Acid hydrolysis of the saponins from the stem bark of Guaiacum officinale yielded the new sapogenin 3β,20η-dihydroxy-30-norolean-12-en-28-oic acid and the artifacts 3β-hydroxy-30-norolean-12,19-dien-28-oic acid and its methyl ester. Larreagenin, sitosterol and oleanolic acid were also isolated.

Abstract Two new saponins guaianin D (1) and E (2), obtained from the bark of Guaiacum officinale have been characterised by 13C-nmr spectroscopy and chemical reactions.The complete structure of these saponins have been determined as 3-0[α-L-rhamnopyranosyl (1 → 3) [α-L-rhamnopyranosyI] (1→ 2) α-L-arabinopyranosyl (3 → 1)-(β-D-glucopyranosyl]-30-norolean-12,20 (29) dien-28-oic acid (1) and 3-0-[α-L-rhamnopyranosyl] (1 → 3)-[α-L-rhamno-pyranosyl] (1 → 2)-[a-L-arablnopyranosyl (3 → 1)-[β -D-glucopyranosyl] 30-norolean-12, 20(29)-dien-28-0-[β-D-glycopyranosyl] ester (2).
